Reviving chloroquine for anti-SARS-CoV-2 treatment with cucurbit[7]uril-based supramolecular formulation

The wide-spreading SARS-CoV-2 virus has put the world into boiling water for more than a year,however pharmacological therapies to act effectively against coronavirus disease 2019(COVID-19)remain ***(CQ),an antimalarial drug,was found to exhibit promising antiviral activity in vitro and in vivo at a high dosage,thus CQ was approved by the FDA for the emergency use authorization(EUA)in the fight against COVID-19 in the US,but later was revoked the EUA status due to the severe clinical ***,we show that supramolecular formulation of CQ by a macrocyclic host,curcurbit[7]uril(CB[7]),reduced its non-specific toxicity and improved its antiviral activity against coronavirus,working in synergy with CB[7].CB[7]was found to form 1:1 host-guest complexes with CQ,with a binding constant of$104 L/*** CQ-CB[7]formulation decreased the cytotoxicity of CQ against Vero E6 and L-02 cell *** particular,the cytotoxicity of CQ(60 mmol/L)against both Vero E6 cell line and L-02 cell lines was completely inhibited in the presence of 300 mmol/L and 600 mmol/L CB[7],***,the CB[7]alone showed astonishing antiviral activity in SARS-CoV-2 infected Vero E6 cells and mouse hepatitis virus strain A59(mHV-A59)infected N2 A cells,and synergistically improved the antiviral activity of CQ-CB[7],suggesting that CB[7]-based CQ formulation has a great potential as a safe and effective antiviral agent against SARS-CoV-2 and other coronavirus.

Selected drug release to targeted tissues has been one of the most challenging tasks in drug delivery, which, if works as expected, would improve therapeutic efficacy and reduce side effects. In reality, the amount of drug released in targeted tissue is often far from adequacy, as most of the drugs are usually metabolized or eliminated during blood circulation before reaching targeted tissue.1 Recently, a variety of stimuli-sensitive drug delivery systems exhibiting selective drug release behaviors in response to the microenvironment in the specific tissues have been developed, however, few of these materials are able to achieve ideal targeting efficiency2. Herein, we report macrophage cell membrane coated ROS responsive nanoparticles（mm-NPs） based on oligochitosan. On the one hand, the membrane of immune cells significantly lowered the immunogenicity of the NPs and contributed to the targeted drug transport to inflammatory tissues where a high level of ROS is present. On the other hand, after reaching the targeted inflammatory tissue, the ROS sensitive NPs were broken apart and the encapsulated therapeutic payload was released in response to the high level of ROS. Transmission electron microscope（TEm） and dynamic light scattering（DLS） analysis exhibited a uniform nanosphere and good stability in PBS and cell culture medium. In vitro drug release profiles of the mm-NPs under various ROS conditions as well as in ROS-generating cellular model exhibited decent ROS-responsiveness. Furthermore, atorvastatin loaded NPs（mm-AT-NPs） improved the cell viability and lowered apoptosis rates of macrophage treated by lipopolysaccharide（LPS）, similar to those treated by free AT. The NO level and inflammatory cytokines were also attenuated on LPS treated macrophages by mm-AT-NPs. Finally, in vivo experiment on mouse showed significantly improvement on the accumulation of mm-NPs at the site of atherosclerosis in comparison to free drug and ROS responsive NPs. Rationale:Comorbidity of depression and anxiety is one of the most common mental disorders in *** experimental procedures,however,have focused on identifying mixed depression-and anxiety-like behaviors in animal ***:To develop a novel testing paradigm,the so-called Liang’s contextual-stress Box(briefly,Liang’s Box),for evaluating depression-and/or anxiety-like behaviors in ***:Liang’s Box consisted of a central area(CA)with three peripheral arms(PA).mice were placed individually in the end of a peripheral arm facing to the central area and were allowed to travel around Liang’s *** a test period,behavioral parameters were recorded and collected by a video-tracking system named SmART 3.0(Panlab,Spain),including①Latency to CA,②Time spent in CA(CA-time),③Distance traveled in CA(CA-distance),④Distance traveled in PA(PA-distance),⑤Transition number between CA and PA,⑥Global activity,⑦CA-time/cm2,⑧PA-time/cm2,⑨CAdistance/cm2,and⑩PA-distance/*** order to assess the reliability and validity of Liang’s Box and get baseline information,we undertook a series of experiment,including verifying consistency and stability in the various strains of mice(Km,ICR,and C57 BL/6 J mice),imposing high illumination,restraint and tail suspension stress,pre-treating with an anxiolytic(diazepam,buspirone)or antidepressant(desipramine,mianserin).Results:①There were not remarkable differences in all the parameters among Km,ICR,and C57 BL/6 J mice;②High illumination,restraint and tail suspension significantly increased the latency to CA and decreased the CA-time,CA-distance and transition ***,these three types of stresses evoked different effect on global *** illumination increased activity,but restraint stress had no significant *** contrast with high illumination,tail suspension stress markedly reduced activity in mice;③The anxiolytics diazepam(0.25 mg·kg-1 and 0.5 mg·kg-1,i.p.)and buspirone(0.5 mg·kg-1 and 1.0 mg·kg-1,i.p.)

In situ characterization of particle formation in spray flame synthesis using wide-angle light scattering

Wide-angle light scattering(WALS)was used for in situ measurements of droplet and nanoparticle size distributions during the synthesis of titania and iron oxide particles from liquid precursor solutions in the standardized SpraySyn burner for spray flame *** was synthesized from titanium tetraisopropoxide(TTIP)and iron oxide from iron(II)nitrate nonahydrate(INN)using ethanol(EtOH)as *** images were taken at heights up to 120 mm above the burner surface and classified into droplet and particle *** size distributions were derived from a sequential analysis of scattering data containing the oscillating mie pattern,the lognormal size distribution parameters for spherical and fractal particle fractions from a multivariate approach on averaged particle scattering *** results show that the precursor addition leads to altered evaporation behavior and even droplet disruption probably induced by puffing or micro-explosions compared to pure *** the case of TTIP(a hygroscopic alkoxide),the synthesis of a large fraction of spheres was observed,while the nitrate INN leads to the formation of mostly fractal aggregates.

Internal flow in sessile droplets induced by substrate oscillation:towards enhanced mixing and mass transfer in microfluidic systems

The introduction of flows within sessile droplets is highly effective for many lab-on-a-chip chemical and biomedical ***,generating such flows is difficult due to the typically small droplet ***,we present a simple,non-contact strategy to generate internal flows in sessile droplets for enhancing mixing and mass *** flows are driven by actuating a rigid substrate into oscillation with certain amplitude distributions without relying on the resonance of the droplet *** oscillation characteristics and corresponding flow patterns are documented *** indices and mass transfer coefficients of sessile droplets on the substrate surface are measured using optical and electrochemical *** demonstrate complete mixing within the droplets in 1.35 s and increases in mass transfer rates of more than seven times static *** of concept was conducted with experiments of silver nanoparticle synthesis and with heavy metal ion sensing employing the sessile droplet as a microreactor for synthesis and an electrochemical cell for *** degrees of enhancement of synthesis efficiency and detection sensitivity attributed to the internal flows are experimentally documented.

Phase change materials(PCms) designate materials able to store latent *** change state from solid to liquid over a defined temperature *** process is reversible and can be used for thermo-technical *** present paper aims to study the thermal performance of an inorganic eutectic PCm integrated into the rooftop slab of a test room and analyze its potential for building thermal *** experiment is conducted in two test rooms in Antofagasta(Chile) during summer,fall,and *** PCm is integrated into the rooftop of the first test room,while the roof panel of the second room is a sealed air *** work introduces a numerical model,which is built using the finite difference method and used to simulate the rooms' thermal *** thermal simulations of the PCm room are performed for other Chilean locations to evaluate and compare the capability of the PCm panel to store latent heat thermal energy in different *** show that the indoor temperature of the PCm room in Antofagasta varies only 21.1℃±10.6℃,while the one of the air-panel room varies 28.3℃±18.5℃.Under the experiment's conditions,the PCm room's indoor temperature observes smoother diurnal fluctuations,with lower maximum and higher minimum indoor temperatures than that of the air-panel *** simulations in other cities show that the PCm panel has a better thermal performance during winter,as it helps to maintain or increase the room temperature by some degrees to reach comfort *** demonstrates that the implementation of such PCm in the building envelope can effectively reduce space heating and cooling needs,and improve indoor thermal comfort in different climates of Chile.

Occupant-centric controls(OcC)is an indoor climate control approach whereby occupant feedback is used in the sequence of operation of building energy *** OcC has been used in a wide range of building applications,an OcC category that has received considerable research interest is learning occupants'thermal preferences through their thermostat interactions and adapting temperature setpoints *** recent studies used reinforcement learning(RL)as an agent for OcC to optimize energy use and occupant *** studies depended on predicted mean vote(PmV)models or constant comfort ranges to represent comfort,while only few of them used thermostat *** paper addresses this gap by introducing a new off-policy reinforcement learning(RL)algorithm that imitates the occupant behaviour by utilizing unsolicited occupant thermostat *** algorithm is tested with a number of synthetically generated occupant behaviour models implemented via the Python APl of *** simulation results indicate that the RL algorithm could rapidly learn preferences for all tested occupant behaviour scenarios with minimal exploration *** substantial energy savings were observed with most occupant scenarios,the impact on the energy savings varied depending on occupants'preferences and thermostat use behaviour stochasticity.
