Plant-specific transcriptional regulators called TElOMERE REPEAT BINDING proteins(TRBs)combine two DNA-binding domains,the GH1 domain,which binds to linker DNA and is shared with H1 histones,and the Myb/SANT domain,which specifically recognizes the telobox DNA-binding site ***1,TRB2,and TRB3 proteins recruit Polycomb group complex 2(PRC2)to deposit H3K27me3 and jMj14 to remove H3K4me3 at gene promoters containing telobox motifs to repress ***,we demonstrate that TRB4 and TRB5,two related paralogs belonging to a separate TRB clade conserved in spermatophytes,regulate the transcription of several hundred genes involved in developmental responses to environmental ***4 binds to several thousand sites in the genome,mainly at transcription start sites and promoter regions of transcriptionally active and H3K4me3-marked genes,but,unlike TRB1,it is not enriched at H3K27me3-marked gene ***,TRB4 can physically interact with the catalytic components of PRC2,SWINGER,and CURlY lEAF(ClF).Unexpectedly,we show that TRB4 and TRB5 are required for distinctive phenotypic traits observed in clf mutant plants and thus function as transcriptional activators of several hundred ClF-controlled genes,including key flowering *** further demonstrate that TRB4 shares multiple target genes with TRB1 and physically and genetically interacts with members of both TRB ***,these results reveal that TRB proteins engage in both positive and negative interactions with other members of the family to regulate plant development through both PRC2-dependent and-independent mechanisms.

Purpose:To examine the effects of a school-based karate intervention on academic achievement,psychosocial functioning,and physical fitness in children aged 7-8 ***:Twenty schools in 5 different European countries(2 second-grade classrooms per school)participated in a cluster randomized controlled trial(Sport at School trial).Participants were assigned to either a control group,which continued with their habitual physical education lessons,or to an intervention group,which replaced these lessons with a 1-year karate intervention(Karate Mind and Movement program).A total of 721 children(344 girls and 377 boys,7.4±0.5 years old,mean±SD)completed the study,of which 333 and 388 were assigned to the control group and intervention group,*** included academic performance(average grade),psychosocial functioning(Strengths and Difficulties Questionnaire for parents),and different markers of physical fitness(cardiorespiratory fitness,balance,and flexibility).Results:The intervention provided small but significant benefits compared to the control group for academic achievement(d=0.16;p=0.003),conduct problems(d=-0.28;p=0.003),cardiorespiratory fitness(d=0.36;plance(d=0.24;p=0.015).There was a trend towards significant benefits for flexibility(d=0.24;p=0.056).No significant benefits were observed for other variables,including psychosocial difficulties,emotional symptoms,hyperactivity/inattention,peer problems,or prosocial behaviour(all p>0.05).Conclusion:A 1-year school-based karate intervention was effective in improving academic achievement,conduct problems,and physical fitness in primary school *** results support the inclusion of karate during physical education lessons.

Utilizing machine learning techniques for data-driven diagnosis of high temperature PEM fuel cells is beneficial and meaningful to the system durability. Nevertheless, ensuring the robustness of diagnosis remains a critical and challenging task in real application. To enhance the robustness of diagnosis and achieve a more thorough evaluation of diagnostic performance, a robust diagnostic procedure based on electrochemical impedance spectroscopy (EIS) and a new method for evaluation of the diagnosis robustness was proposed and investigated in this work. To improve the diagnosis robustness: (1) the degradation mechanism of different faults in the high temperature PEM fuel cell was first analyzed via the distribution of relaxation time of EIS to determine the equivalent circuit model (ECM) with better interpretability, simplicity and accuracy;(2) the feature extraction was implemented on the identified parameters of the ECM and extra attention was paid to distinguishing between the long-term normal degradation and other faults;(3) a Siamese Network was adopted to get features with higher robustness in a new embedding. The diagnosis was conducted using 6 classic classification algorithms—support vector machine (SVM), K-nearest neighbor (KNN), logistic regression (lR), decision tree (DT), random forest (RF), and Naive Bayes employing a dataset comprising a total of 1935 collected EIS. To evaluate the robustness of trained models: (1) different levels of errors were added to the features for performance evaluation;(2) a robustness coefficient (Roubust_C) was defined for a quantified and explicit evaluation of the diagnosis robustness. The diagnostic models employing the proposed feature extraction method can not only achieve the higher performance of around 100% but also higher robustness for diagnosis models. Vitamin A and its biologically active derivative,retinoic acid(ra),are important for many immune ***,in particular,is essential for the development of immune cells,including neutrophils,which serve as a front-line defense against *** vitamin A deficiency has been linked to higher susceptibility to infections,the precise role of vitamin A/ra in host-pathogen interactions remains poorly ***,we provided evidence that ra boosts neutrophil killing of methicillin-resistant Staphylococcus aureus(MRSA).ra treatment stimulated primary human neutrophils to produce reactive oxygen species,neutrophil extracellular traps and the antimicrobial peptide cathelicidin(ll-37).Because ra treatment was insufficient to reduce MRSA burden in an in vivo murine model of skin infection,we expanded our analysis to other infectious *** did not affect the growth of a number of common bacterial pathogens,including MRSA,Escherichia coli K1 and Pseudomonas aeruginosa;however,ra directly inhibited the growth of group A Streptococcus(GAS).This antimicrobial effect,likely in combination with ra-mediated neutrophil boosting,resulted in substantial GAS killing in neutrophil killing assays conducted in the presence of ***,in a murine model of GAS skin infection,topical ra treatment showed therapeutic potential by reducing both skin lesion size and bacterial *** findings suggest that ra may hold promise as a therapeutic agent against GAS and perhaps other clinically significant human pathogens.

Potato cyst nematodes(PCNs)are a significant threat to potato production,having caused substantial damage in many *** the future distribution of PCN species is crucial to implementing effective biosecurity strategies,especially given the impact of climate change on pest species invasion and *** learning(Ml),specifically ensemble models,has emerged as a powerful tool in predicting species distributions due to its ability to learn and make predictions based on complex data ***,this research utilised advanced machine learning techniques to predict the distribution of PCN species under climate change conditions,providing the initial element for invasion risk *** first used Global Climate Models to generate homogeneous climate predictors to mitigate the variation among ***,five machine learning models were employed to build two groups of ensembles,single-algorithm ensembles(ESA)and multi-algorithm ensembles(EMA),and compared their *** this research,the EMA did not always perform better than the ESA,and the ESA of Artificial Neural Network gave the highest performance while being *** results indicated that the distribution range of PCNs would shift northward with a decrease in tropical zones and an increase in northern ***,the total area of suitable regions will not change significantly,occupying 16-20%of the total land surface(18%under current conditions).This research alerts policymakers and practitioners to the risk of PCNs’incursion into new ***,this Ml process offers the capability to track changes in the distribution of various species and provides scientifically grounded evidence for formulating long-term biosecurity plans for their control.

Phase change materials(PCMs) designate materials able to store latent *** change state from solid to liquid over a defined temperature *** process is reversible and can be used for thermo-technical *** present paper aims to study the thermal performance of an inorganic eutectic PCM integrated into the rooftop slab of a test room and analyze its potential for building thermal *** experiment is conducted in two test rooms in Antofagasta(Chile) during summer,fall,and *** PCM is integrated into the rooftop of the first test room,while the roof panel of the second room is a sealed air *** work introduces a numerical model,which is built using the finite difference method and used to simulate the rooms' thermal *** thermal simulations of the PCM room are performed for other Chilean locations to evaluate and compare the capability of the PCM panel to store latent heat thermal energy in different *** show that the indoor temperature of the PCM room in Antofagasta varies only 21.1℃±10.6℃,while the one of the air-panel room varies 28.3℃±18.5℃.Under the experiment's conditions,the PCM room's indoor temperature observes smoother diurnal fluctuations,with lower maximum and higher minimum indoor temperatures than that of the air-panel *** simulations in other cities show that the PCM panel has a better thermal performance during winter,as it helps to maintain or increase the room temperature by some degrees to reach comfort *** demonstrates that the implementation of such PCM in the building envelope can effectively reduce space heating and cooling needs,and improve indoor thermal comfort in different climates of Chile.

In recent years,e-scooters have been introduced in many European *** several places we have witnessed a rapid uptake of this new mode of transport mainly as a result of public sharing schemes.A number of incidents,injuries and even fatalities have given rise to questions regarding the safety of these *** questions are being researched mainly using official crash data and data specifying injuries and hospital *** now,the research has focused on investigating typical injury patterns and estimating risk *** little is known about exactly where conflicts and crashes *** of hazard hotspots is crucial when investigating risk levels and improving safety for all road ***,this paper develops an approach to investigating locations with potentially dangerous interactions within the active mobility system in the city of *** approach consists of explorative expert interviews,an online poll,and quantitative *** the latter we combine three ***,we research crash hotspots using official ***,we use data based on acceleration sensors from cyclists'smartphones to find locations of sudden ***,we use trip data from the operators of escooter sharing *** information gathered is used in a conclusive expert workshop to identify hazard *** show that many of the conflicts with pedestrians are caused by parked ***,e-scooter trips are concentrated in the inner city and along specific *** moving traffic,various data sources are used to identify hotspots at intersections and in areas between *** present research lays the foundation for important further studies to investigate interactions at hotspots in detail by determining nine specific locations in the city of Berlin.
