Background Female patients with atrial fibrillation （AF） experience increased risk of thromboembolism compared to males, an observation that is reflected by its inclusion in the CHA2dS2VASc score. New onset AF （often associated with tachycardia） also confers upon patients increased thromboembolic risk. The mechanisms underlying this risk are uncertain, but new onset AF is associated with profound impairment of platelet nitric oxide （NO） signalling. Given that cardiovascular responses to catecholamines are gender-dependent, and that the presence of tachycardia in new onset AF may represent a response to catecholaminergic stimulation, we explored the potential impact of gender and tachycardia on platelet aggregation and NO signalling. Methods Interactions were sought in 87 AF patients between the extent of adenosine diphosphate （AdP）-induced platelet aggregation, the anti-aggregatory effects of the NO donor, sodium nitroprusside, gender, and admission heart rate. The potential impact of platelet expression of thioredoxin-interacting protein （Txnip） was also evaluated. Results Analysis ofcovariance confLrmed the presence of physiological antagonism between platelet AdP and NO responses [F （1, 74） = 12.212, P 〈 0.01 ], while female sex correlated with impaired NO responses independent of platelet aggregability [F （2, 74） = 8.313, P 〈 0.01]. Admission heart rate correlated directly with platelet aggregation （r = 0.235, P 〈 0.05）, and inversely with NO response （r = -0.331, P 〈 0.01）. Txnip expression varied neither with gender nor with heart rate. Conclusions These results indicate, that gender and heart rate are independent determinants of platelet fimction. Prospective studies of the putative benefit of reversal of tachycardia on restoration of normal platelet function are therefore a priority.

dynamic simulation of a double-skin façade(dSF)with computational fluid dynamics(CFd)can be challenging due to the lack of validated models and benchmarking ***,there is a lack of consensus in the scientific community on what constitutes a successfully validated dSF *** present review study identifies simulation trends and research gaps for dSFs simulated with ***,this article presents a series of CFd simulations in which key aspects of the dSF modelling are varied:2d or 3d modelling approaches,turbulence viscosity models(TVMs),radiation models,and wall *** simulation results are compared to the empirical data(both temperature and velocity fie.ds)of a benchmark test with laboratory-controlled boundary *** analysis shows that using the k-εRNG model with enhanced wall treatment and surface-to-surface(S2S)radiation model yields the best results for the 2d case of natural convection ***,it is shown that accounting for the velocity fie.d in the validation process is essential to ensure the suitability of a ***,the authors advocate for the use of selected dimensionless numbers to improve the comparability of the different dSF scientific *** would also help to identify relevant experimental datasets for validation and suitable CFd simulation settings for specific dSF cases.

Recent evidence suggests that genetic and epigenetic mechanisms might be associated with acquired resistance to cancer *** aim of this study was to assess the association of genome-wide genetic and epigenetic alterations with the response to anti-HER2 agents in HER2-positive breast cancer *** was screened for articles published until March 2021 on observational studies investigating the association of genome-wide genetic and epigenetic alterations,measured in breast cancer tissues or blood,with the response to targeted treatment in HER2-positive breast cancer *** studies were included in the review along with ours,in which we compared the genome-wide dNA methylation pattern in breast tumor tissues of patients who acquired resistance to treatment (case group, n = 6) to that of patients who did not develop resistance (control group, n =6). Among genes identified as differentially methylated between the breast cancer tissue of cases and controls, oneof them, PRKACA, was also reported as differentially expressed in two studies included in the review. Althoughincluded studies were heterogeneous in terms of methodology and study population, our review suggests thatgenes of the PI3K pathway may play an important role in developing resistance to anti-HER2 agents in breastcancer patients. Genome-wide genetic and epigenetic alterations measured in breast cancer tissue or blood mightbe promising markers of resistance to anti-HER2 agents in HER2-positive breast cancer patients. Further studiesare needed to confirm these data.

Non-tidal ocean loading （NTOL） is a crucial geophysical factor in resulting in the seasonal signals undermining geodetic GPS height time *** for the NTOL effects has thus been a critical prerequisite for estimating highly reliable secular rates of vertical crustal *** this study,we focus on whether a high temporal resolution model can benefit the correction for NTOL *** processed four years of GPS data from 26 stations around the North Sea of Europe and employed the Proudman Oceanographic Laboratory Storm Surge Model to correct the height *** and 12-hourly corrections were attempted *** generated 2-hourly heights to investigate the sub-daily signatures of NTOL effects,and also daily heights to assess the potential benefits of NTOL corrections of high temporal resolution to geophysical *** expected,hourly NTOL corrections lead to a reduction of the RMS of daily height residuals from 5.1 to 4.4 mm on average,and also a reduction of 21.2% for the integrated signal power over the seasonal to diurnal frequency ***,when applying the 12-hourly corrections instead,the RMS of daily height residuals is only slightly deteriorated by 0.01 mm on average and at most 0.06 mm,and the reduction of the integrated power for the seasonal to diurnal frequency band falls slightly to 20.4%.Therefore,we demonstrate that estimating the secular rates of vertical crustal motions will not benefit further from finer than 12-hourly NTOL corrections.

Erythropoietin (EPO) is one of the most successful biopharmaceuticals in history and is used for treating anemia of different origins. However, it became clear that EPO could also work in a neuroprotective, antiapoptotic, antioxidative, angiogenetic and neurotropic way. It causes stimulation of cells to delay cell apoptosis, especially in the central nervous system. In rodent models of focal cerebral ischemia, EPO showed an impressive reduction of infarct size by 30% and improvement of neurobehavioral outcome by nearly 40%. A large animal model dealing with ischemia and reperfusion of the spinal cord showed that EPO could reduce the risk of spinal cord injury significantly. In addition, some clinical studies tested whether EPO works in real live clinical settings. One of the most promising studies showed the innocuousness and improvements in follow-up, outcome scales and in infarct size, of EPO-use in humans suffering from ischemic stroke. Another study ended unfortunately in a negative outcome and an increased overall death rate in the EPO group. The most possible reason was the involvement of patients undergoing simultaneously systemic thrombolysis with recombinant tissue plasminogen activator. An experimental study on rats demonstrated that administration of EPO might exacerbate tissue plasminogen activator-induced brain hemorrhage without reducing the ischemic brain damage. This case shows clearly how useful animal models can be to check negative side effects of a treatment before going into clinical trials. Other groups looked in human trials at the effects of EPO on the outcome after ischemic stroke, relation to circulating endothelial progerdtor cells, aneurysmal subarachnoid hemorrhage, traumatic brain injury, hemoglobin transfusion thresholds and elective first-time coronary artery bypass surgery. Aim:To investigate whether the endothelin ETAreceptor blocker provides similarbenefit on cardiac remodeling and survival in a hypertensive rat model of chronicheart failure（CHF）.Methods:Male stroke-prone spontaneously hypertensive（SHR-SP）rats were subjected to permanent ligation of the left coronary artery andwere treated for 6 weeks with the endothelin ETAreceptor blocker LU 135252(***-1.d-1)starting 24 h after ligation or ***-operated rats servedas normal *** mean arterial blood pressure（MAP）,heart rate（HR）,leftventricular end-diastolic pressure（LVEdP）,left ventricular contractility(LVdp/dtmax),left ventricular inner diameter（LVd）and circumference（LVC）,septalthickness,left ventricular interstitial collagen content（ICC）and heart weight（HW）were measured at the end of the ***:Compared with the untreatedgroup,LU 135252 tended to increase HW（1.43±0.03 vs 1.38±0.04 g;P>0.05）,increased LVd（7.65±0.24 mm vs 6.58±0.14 mm;Pdly increased LVC（30.11±0.83 mm vs 24.82±0.85 mm;Pd reduced left ventricular ICC（3.79%±0.09% vs 6.71%±0.11%;Pd MAP（132±6mmHg vs142±4 mmHg;P>0.05）,reduced LVEdP（144mmHg vs 27±4 mmHg;Pdimproved LV dp/dtmax（4230±450 mmHg/s vs 1950±400 mmHg/s;Pd significantly（13% vs 11%;P=NS）.Conclusion:In this hyper-tensive rat model of CHF,chronic endothelin ETA receptor blockade with LU 135252improves cardiac hemodynamics,however,it does not affect long-term survivaland worsens cardiac ***,endothelin ETAreceptor antagonists areunlikely to have an important role in the management of patients with CHF.

The aim of this study is to assess and compare two clustering algorithms,namely,the Wave-cluster and density Based Spatial Clustering of Application with Noise（dBSCAN）algorithms,for landslide susceptibility assessment in Baota district,*** on historical reports,interpretation of aerial photographs and fie.d survey reports 293 were identified in the study *** factors associated with landslide occurrence were prepared and used as inputs into the models for landslide susceptibility *** models divided the study area grids into 478 and 465 subclasses respectively,which were further grouped into five susceptibility levels using K-means algorithm,based on the landslide density values of the obtained *** resulting two models were validated and compared using area under the receiver operating characteristics（AUC-ROC）curve and statistical measures including sensitivity,specificity and *** analysis demonstrated that the Wave-cluster model outperformed the dBSCAN model with sensitivity,specificity and accuracy values of 88.05%,89.2% and 88.54% respectively,while dBSCAN had sensitivity,specificity and accuracy values of 86.69%,86.38% and 86.56% *** addition,the Wave-cluster model had higher value AUC of 0.823 than the dBSCAN model（0.808）.This indicates that,the landslide susceptibility maps（LSMs）generated in this study had good performance for assessing landslide susceptibility in the study area and can be useful tools in assessing landslides,as the foundation for preventing and reducing landslide consequences for suitable social and economic ***,this is the first time these models are assessed and compared,especially in landslide susceptibility assessment.
