The role and clinical implications of microRNAs in hepatocellular carcinoma

Hepatocellular carcinoma (Hcc) is common and one of the most aggressive of all human cancers. Recent studies have indicated that miRNAs, a class of small noncoding RNAs that regulate gene expression post-transcriptionally, directly contribute to Hcc by targeting many critical regulatory genes. Several miRNAs are involved in hepatitis B or hepatitis c virus replication and virus-induced changes, whereas others participate in multiple intracellular signaling pathways that modulate apoptosis, cell cycle checkpoints, and growth-factor-stimulated responses. When disturbed, these pathways appear to result in malignant transformation and ultimately Hcc development. Recently, miRNAs circulating in the blood have acted as possible early diagnostic markers for Hcc. These miRNA also could serve as indicators with respect to drug efficacy and be prognostic in Hcc patients. Such biomarkers would assist stratification of Hcc patients and help direct personalized therapy. Here, we summarize recent advances regarding the role of miRNAs in Hcc development and progression. Our expectation is that these and ongoing studies will contribute to the understanding of the multiple roles of these small noncoding RNAs in liver tumorigenesis.

An examination by Gc×Gc-TOFMS of organic molecules present inhighly degraded oils emerging from caribbean terrestrial seeps of cretaceous age

For our ancestors, oil seeps were both a fascination and a resource but as the planet's reserves of high quality low density oil becomes increasingly depleted, so there is now a renewed interest in heavier,biodegraded oils such as those encountered in terrestrial seeps. One such seep is Pitch Lake in the caribbean island of Trinidad, which is the largest natural deposit of asphalt in the world. At the northern end of the caribbean, oil emerges along a tectonic contact on the island on cuba. The sources of the oils from these seeps are relatively recent and both are subject to intense weathering due to the tropical conditions. When analysed by gas chromatography(Gc) both oils appear as unresolved complex mixtures(UcM) and show a very high degree of biodegradation thus presenting an analytical challenge. In this case study, these two caribbean seep oils were analysed by comprehensive two dimensional Gc with time of flight mass spectrometry(Gc×Gc-TOFMS) to expose many thousands of the individual compounds that comprise the UcM. The high chromatographic resolution of the Gc×Gc-TOFMS produced good quality mass spectra allowing many compounds including molecular fossil ‘biomarkers' to be identified. compound classes included diamondoid hydrocarbons, demethylated hopanes and secohopanes, mono-and tri-aromatic steroids. D-ring aromatised structures of the 8,14-seco-hopanes,including demethylated forms were present in both oils but further demethylation, probably at position c-25 during biodegradation, was only observed in the Pitch Lake oil. Many polycyclic aromatic hydrocarbons(PAHs) were absent although the fungal-derived pentacyclic PAH perylene was present in both oils. The presence of the angiosperm biomarker lupane in the Pitch Lake oil constrained the age to the Late cretaceous. The higher degree of biodegradation observed in the cuban oil was likely due to relatively slow anaerobic processes whereas oil within Pitch Lake was probably subject to additional more rapid

将１５０μｇ１硝基芘（１ＮＰ）气管内或血管内给药后，对大鼠离体肺进行灌流，定时自灌流液中取样，以ＨＰＬＣ和ＧＣＭＳ测定并鉴定灌流液中未变的１ＮＰ及代谢物。经鉴定代谢物为单羟基硝基芘和二羟基硝基芘。两种途径给药后，１ＮＰ在灌流液中的浓度—时间数据符合一室药代动力学模型。预先以β萘黄酮，苯并蒽或苯巴比妥和β萘黄酮的混合物处理后的大鼠，肺灌流结果表明，显著降低１ＮＰ的平均保留时间（ＭＲＴ），促进１ＮＰ的生物转化和气管给药后的吸收。以苯巴比妥预处理后的大鼠肺则对１ＮＰ的药代动力学参数无显著影响。

Microbial phosphorus(P)turnover is critical in c utilization efficiency in *** is therefore necessary to understand the P mobilization processes occurring during P fertilization in order to ensure both crop yield and environmental ***,we established a controlled pot experiment containing soil amended with three different levels of starter P fertilizer and collected soil samples after 30,60,and 90 days of *** microbial element cycling(QMEc)smart chip technology and 16S rRNA gene sequencing were used to investigate functional gene structures involved in carbon,nitrogen and P cycling and the bacterial community composition of the collected *** P fertilization did not significantly affect the structure of the soil microbial community,some rare microbiota were changed in particular phosphorus-solubilizing bacteria were enriched at the high P fertilization level,suggesting that the rare taxa make an important contribution to P turnover.P fertilization also altered the functional gene structure,and high P concentrations enhanced the functional gene diversity and *** redundancy analysis further revealed that changes in rare taxa and functional genes of soil microorganisms drive the alteration of soil P *** findings extend our understanding of the microbial mechanisms of P turnover.

Pathogenicity of bacterium, Xenorhabdus nematophila isolated from entomopathogenic nematode (Steinernema carpocapsae) and its secretion against Galleria mellonella larvae

The entomopathogenic bacterium, Xenorhabdus nematophila was isolated from the hemolymph of Galleria mel- lonella infected with Steinernema carpocapsae. The bacterial cells and its metabolic secretions have been found lethal to the Galleria larvae. Toxic secretion in broth caused 95% mortality within 4 d of application whereas the bacterial cells caused 93% mortality after 6 d. When filter and sand substrates were compared, the later one was observed as appropriate. Similarly, bacterial cells and secretion in broth were more effective at 14% moisture and 25 °c temperature treatments. Maximum insect mortality (100%) was observed when bacterial concentration of 4×106 cells/ml was used. Similarly, maximum bacterial cells in broth (95%) were penetrated into the insect body within 2 h of their application. However, when stored bacterial toxic secretion was applied to the insects its efficacy declined. On the other hand, when the same toxic secretion was dried and then dissolved either in broth or water was proved to be effective. The present study showed that the bacterium, X. nematophila or its toxic secretion can be used as an important component of integrated pest management against Galleria.

A new spallation mechanism of thermal barrier coatings on aero-engine turbine blades

Laboratory experiments were conducted to study the spallation behaviour of thermal barrier coatings(TBcs) on aero-engine turbine blades manufactured by the electron-beam physical vapour deposition technique(EB-PVD). Intact blades were heated at temperature 1135℃ in a furnace for certain time and then cooled to the room temperature in the laboratory condition. It was found that no spallation occurred during cooling, but spallation happened at constant room temperature after cooling. The spallation mechanism is studied by using the mechanical model developed(Harvey 2017 and Wang 2017), which are based on the hypothesis of pockets of energy concentration(PEcs). Some observations of the spallation behaviour are well predicted by the model.

Understanding how natural selection shapes unique traits in mammals is a central topic in evolutionary *** mammalian order chiroptera(bats)is attractive for biologists as well as the general public due to their speciﬁc traits of extraordinary immunity and inverted resting ***,genomic resources for bats that occupy key phylogenetic positions are not sufﬁcient,which hinders comprehensive investigation of the molecular mechanisms underpinning the origin of speciﬁc traits in ***,we sequenced the transcriptomes of 5 bats that are phylo-genetically divergent and occupy key positions in the phylogenetic tree of *** combination with the available genomes of 19 bats and 21 other mammals,we built a database consisting of 10918 one-to-one ortholog genes and reconstructed phylogenetic relationships of these *** found that genes related to immunity,bone remodeling,and cardiovascular system are targets of natural selection along the ancestral branch of *** analyses revealed that the T cell receptor signaling pathway involved in immune adaptation is speciﬁcally enriched in ***,molecular adaptations of bone remodeling,cardiovascular system,and balance sensing may help to explain the reverted resting posture in *** study provides valuable transcriptome resources,enabling us to tentatively identify genetic changes associated with bat-speciﬁc *** work is among theﬁrst to advance our understanding of the molecular underpinnings of inverted resting posture in bats,which could provide insight into healthcare applications such as hypertension in humans.
