Insight into the role of lactic acid bacteria in the development of a novel fermented pistachio (Pistacia vera L.) beverage

This study aimed to assess the fermentability of a pistachio-based beverage. The effects of lactic acid fermen-tation on the odor profile and the stability of the beverage were also assessed. To this end, a procedure for obtaining a pistachio beverage was developed, and the nutritional composition of the beverage was obtained. The beverage was characterized by a high content of fiber (2.9%), fat (4.6%), and protein (2%), and contained high amounts of K and Mg. For the experiment, 43 cultures of the following lactic acid bacteria were used: Companilactibacillus paralimentarius, Companilactibacillus kimchi, Levilactibacillus brevis, Lactiplantibacillu pentosus, Lactiplantibacillus plantarum, Lactiplantibacillus paraplantarum, Latilactobacillus curvatus, Leuconostoc pseudome-senteroides, and Furfurilactobacillus rossiae. The cultures were evaluated for the following technological charac-teristics: i) growth and acidifying activity, ii) production of exopolysaccharides;iii) presence of histidine decarboxylase (hdcA) gene;iv) antimicrobial activity against Listeria innocua. The cultures were used in model pistachio-based fermentations to assess their ability to grow and acidify. The odor profile of the beverage after 24 h fermentation was assessed using an electronic nose system (E-nose). The pistachio-based beverage resulted an optimal substrate for lactic acid bacteria that reached loads between 8 and 10 log CFU/mL and acidified the beverage to pH values between 4 and 5.5. In the beverage, viable counts of all cultures remained above 8 log CFU/mL after 30 days of storage at 4 degrees C. The E-nose analysis represented a valid screening method to evaluate the effects of the fermentative activity of microbial starters on the qualitative characteristics of this novel food matrix.

Investigation of Argentinean Plant Extracts for Their Antibacterial Activity

Plants of Baccharis （Asteraceae） genus are commonly known in Argentina as ＂carqueja＂. The antimicrobial activity and minimal inhibitory concentration of B. articulata, B. trimera and B. crispa aqueous and ethanolic extracts were evaluated by using the micro-well dilution method. Previously, the components of extracts were analyzed by spectroscopial means. gram-positive bacteria were more sensitive to Baccharis species extracts than gram-negative bacteria. Out of 3 plant species, B. trimera showed significant antibacterial activity and aqueous and ethanolic extracts were active against Staphylococcus aureus （MIC = 2,500 μg/mL and 1,250 μg/mL, respectively） and Listeria monocytogenes （MIC = 625 μg/mL and 625 μg/mL, respectively）. All ethanolic extracts inhibited the growth of the selected gram-positive （MIC values ranged between 625 μg/mL and 1,250 μg/mL）. Therefore, all gram-negative bacteria were resistant to the ethanolic and aqueous extracts tested. One flavone, genkawanin, was identified from the three ethanolic extracts as the responsible of antibacterial activity. Two terpenes, hawtriwaic acid and bacrispine, were identified from ethanolic extract of B. crispa and B. trimera as the responsibles of antibacterial activity. These preliminary studies corroborated the antimicrobial activity of the selected plants used in folklore medicine. Therefore, they could be potential sources of new antimicrobial agents used in treatment of infectious diseases.

Thelesperma megapotamicum (Asteraceae) is commonly used in Argentine to treat various diseases (renal, digestive affections, and as anaesthesia). The present study showed the mechanisms involved “in vitro” cytotoxicity of T. megapotamicum Fractions. Five Fractions (F1 - F5) were separated by column chromatography (Silica gel) using hexane:diethyl ether as eluents. Viability was evaluated in Human breast carcinoma cell line (MCF-7) by staining with crystal violet. With respect to F1 Fraction treatment, the cell survival was 49.14% ± 8.87%, while the F2 and F3 ones exhibited a strong reduction of cell viability to only 26.35% ± 1.63% and 23.3%1 ± 0.53% of the control cell at 50 μg/ml, respectively. Apoptotic effect of these Fractions was detected using FITC-labeled Annexin V and propidium iodide binding assays and was confirmed by a higher proportion of apoptotic cells due to F2 and F3 treatments. T. megapotamicum active Fractions could facilitate the tumoral cells death by decreasing the activity of the enzyme gamma-glutamyltranspeptidase and causing alteration in cell membrane sialoglycoconjugates and others involved anticancer mechanisms including apoptosis.

Root hairs are single cells that develop by tip growth, a process shared with pollen tubes, axons, and fungal hyphae. However, structural plant cell walls impose constraints to accomplish tip growth. In addition to polysaccharides, plant cell walls are composed of hydroxyproline-rich glycoproteins （HRgPs）, which include several groups of O-glycoproteins, including extensins （EXTs）. Proline hydroxylation, an early post-translational modification （PTM） of HRgPs catalyzed by prolyl 4-hydroxylases （P4Hs）, defines their subsequent O-glycosylation sites. In this work, our genetic analyses prove that P4H5, and to a lesser extent P4H2 and P4H13, are pivotal for root hair tip growth. Second, we demonstrate that P4H5 has in vitro preferred specificity for EXT substrates rather than for other HRgPs. Third, by P4H promoter and protein swapping approaches, we show that P4H2 and P4H13 have interchangeable functions but cannot replace P4H5. These three P4Hs are shown to be targeted to the secretory pathway, where P4H5 forms dimers with P4H2 and P4H13. Finally, we explore the impact of deficient proline hydroxylation on the cell wall architec- ture. Taken together, our results support a model in which correct peptidyl-proline hydroxylation on EXTs, and possibly in other HRgPs, is required for proper cell wall self-assembly and hence root hair elongation in Arabidopsis thaliana.
