Root hairs are single cells that develop by tip growth, a process shared with pollen tubes, axons, and fungal hyphae. However, structural plant cell walls im.ose constraints to accom.lish tip growth. In addition to polysaccharides, plant cell walls are com.osed of hydroxyproline-rich glycoproteins （HRGPs）, which include several groups of O-glycoproteins, including extensins （EXTs）. Proline hydroxylation, an early post-translational m.dification （PTm. of HRGPs catalyzed by prolyl 4-hydroxylases （P4Hs）, defines their subsequent O-glycosylation sites. In this work, our genetic analyses prove that P4H5, and to a lesser extent P4H2 and P4H13, are pivotal for root hair tip growth. Second, we dem.nstrate that P4H5 has in vitro preferred specificity for EXT substrates rather than for other HRGPs. Third, by P4H prom.ter and protein swapping approaches, we show that P4H2 and P4H13 have interchangeable functions but cannot replace P4H5. These three P4Hs are shown to be targeted to the secretory pathway, where P4H5 form. dim.rs with P4H2 and P4H13. Finally, we explore the im.act of deficient proline hydroxylation on the cell wall architec- ture. Taken together, our results support a m.del in which correct peptidyl-proline hydroxylation on EXTs, and possibly in other HRGPs, is required for proper cell wall self-assem.ly and hence root hair elongation in Arabidopsis thaliana.

Background Retinopathy of prem.turity(ROP)is generally considered to be m.re frequent in m.les than in fem.les.However,it is not known whether sex differences in ROP affect all degrees of the condition,are global and have changed as neonatology has developed.We aim.d to conduct a system.tic review and m.ta-analysis of studies addressing sex differences in the risk of developing ROP.m.thods Pubm.d/m.DLINE and Em.ase databases were searched.The frequentist,random.effects risk ratio(RR)and 95%confidence interval(CI)were calculated.Bayesian m.del averaged(Bm.)m.ta-analysis was used to calculate the Bayes factors(BFs).The BF10 is the ratio of the probability of the data under the alternative hypothesis(H1)over the probability of the data under the null hypothesis(H0).Results We included 205 studies(867,252 infants).Frequentist m.ta-analysis showed a positive association between m.le sex and severe ROP(113 studies,RR=1.14,95%CI=1.07–1.22)but no association with any ROP(144 studies,RR=1.00,95%CI=0.96–1.03).Bm. showed extrem. evidence in favor of H1 for severe ROP(BF10=71,174)and strong evidence in favor of H0 for any ROP(BF10=0.05).The association between m.le sex and severe ROP rem.ined stable over tim. and was present only in cohorts from.countries with a high or high-m.ddle sociodem.graphic index.Conclusions Our study confirm. the presence of a m.le disadvantage in severe ROP but not in less severe form. of the disease.There are variations in the sex differences in ROP,depending on geographical location and sociodem.graphic level of the countries.

This narrative review provides an overview of the utilization of endoscopic ultrasound-guided gallbladder drainage(EUS-GBD)as a salvage approach in cases of unsuccessful conventional m.nagem.nt.EUS-GBD is a m.nim.lly invasive and effective technique for drainage in patients with acute cholecystitis with high risk of surgery.The procedure has dem.nstrated im.ressive technical and clinical success rates with low rates of adverse events,m.king it a safe and effective option for appropriate candidates.Furtherm.re,EUS-GBD can also serve as a rescue option for patients who have failed endoscopic retrograde cholangiopancreatography or EUS biliary drainage for relief of jaundice in m.lignant biliary stricture.However,patient selection is critical for the success of EUS-GBD,and proper patient selection and risk assessm.nt are im.ortant to ensure the safety and efficacy of the procedure.As the field continues to evolve and m.ture,ongoing research will further refine our understanding of the benefits and lim.tations of EUS-GBD,ultim.tely leading to im.roved outcom.s for patients.

Biodegradable nanoparticles such as chitosan nanoparticles (CSNPs) are used in sustainable agriculture since theyavoid dam.ge to the environm.nt;CSNPs have positive effects such as the accum.lation of bioactive com.oundsand increased productivity in plants. This study aim.d to investigate the im.act of applying CSNPs on lettuce,specifically focusing on enzym.tic activity, bioactive com.ounds, and yield. The trial was conducted using a com.letelyrandom.zed design, incorporating CSNPs: 0, 0.05, 0.1, 0.2, 0.4, and 0.8 m. m.−1. The doses of 0.4 m. m.−1im.rove yields up to 24.6% increases and 0.1 m. m.−1 of CSNPs increases total phenols by 31.2% and antioxidantcapacity by 34.6%. In addition, when low concentrations of CSNPs (0.05 and 0.1 m. L−1) were applied, anincrease in catalase was determ.ned. The CSNPs represent a good alternative to be used as a biostim.lant in sustainableagriculture because they im.rove the yield and quality of lettuce by increasing the bioactive com.ounds.

The use of entom.pathogenic fungi (EF) in recent years has been highly effective against the different orders of insects considered pests of agricultural im.ortance and their conidia have been com.only applied, but it has been reported that these are sensitive to the environm.ntal conditions. For this reason, biopesticides products have been form.lated based on secondary m.tabolites, recently. These biom.lecules participate as biological control agent, such as: cyclic depsipeptides, am.no acids, polyketides, polyphenols and terpenoids, affecting their m.rphology, life cycle and insect behavior. The use of secondary m.tabolites of entom.pathogenic fungi opens the possibility of application in a m.re efficient way for the control of agricultural pests in a com.atible with the environm.nt and hum.n health;therefore, it is im.ortant to know, analyzing the type of m.lecules, their effects, and their different m.thods of application.

The dual role of m.crophages in the healing process depends on m.crophage ability to polarize into phenotypes that can propagate inflam.ation or exert anti-inflam.atory and tissue-rem.deling functions.Controlling scaf-fold geom.try has been proposed as a strategy to influence m.crophage behavior and favor the positive host response to im.lants.Here,we fabricated Polycaprolactone(PCL)scaffolds by m.lt Electrowriting(m.W)to investigate the ability of scaffold architecture to m.dulate m.crophage polarization.Prim.ry hum.n m.crophages unpolarized(m.)or polarized into m.,m.a,and m.c phenotypes were cultured on PCL film. and m.W scaffolds with pore geom.tries(square,triangle,and rhom.us grid)characterized by different angles.m.,m.a,and m.c m.crophages wrapped along the fibers,while m. m.crophages form.d clusters with rounded cells.Cell bridges were form.d only for angles up to 90◦.No relevant differences were found am.ng PCL film. and 3D scaffolds in term. of surface m.rkers.CD206 and CD163 were highly expressed by m.a and m.c m.crophages,with m.a m.crophages presenting also high levels of CD86.m. m.crophages expressed m.derate levels of all m.rkers.The rhom.us architecture prom.ted an increased release by m.a m.crophages of IL10,IL13,and sCD163 com.ared to PCL film..The proangiogenic factor IL18 was also upregulated by the rhom.us configuration in m. and m.a m.crophages com.ared to PCL film..The interesting findings obtained for the rhom.us architecture represent a starting point for the design of scaffolds able to m.dulate m.crophage phenotype,prom.ting investigations addressed to verify their ability to facilitate the healing process in vivo.

Background:Continuous and prim.ry in vitro cultures are largely used to study cellular m.chanism. occurring in several pathologic-like or pathological conditions.Continuous cell lines allow to perform.long-lasting experim.nts since they do not undergo senescence.m.thods:The im.ortalized m.orfields/Institute of Ophtalm.logy-m.ller 1(m.O-m.)cell type represents a valuable m.del to analyze the m.chanistic pathways characterizing m.ller glial cells,both in health and in disease.m.O-m. can be used to dissect the response of these glial cells following treatm.nts which m.m.c pathological condition.For instance,m.O-m. are useful to study the response of this cell type to stress condition as the case of oxidative stress(OS)(cultured with hydrogen peroxide),pathological neovascularization(cultured with VEGF),hypoxic or hyperoxic condition(cultured in low or high oxygen cham.er).On the other hand,prim.ry cultures allow to specifically analyze cellular responses without the interference of the whole organ,although the experim.ntal treatm.nt is perform.d in vivo.Prim.ry m.ller cells can be used to perform.electrophysiological analyses of different cell sites.Discussion:We describe how to m.nage m.O-m. cells and how to analyze their response to different stress conditions;m.reover,we report how to isolate and identify prim.ry m.ller cells and how to perform.patch clam. and single cell recordings on them.
