Common genetic variations in CLOCK transcription factor are associated with nonalcoholic fatty liver disease

AIM: To investigate the role of gene variants and derived haplotypes of the CLOCK transcription factor in nonalcoholic fatty liver disease (NAFLD) and their relation with the disease severity. METHODS: A total of 136 patients with NAFLD and 64 healthy individuals were studied. Liver biopsy was performed in 91 patients. Six tag SNPs showing a minor allele frequency > 10% (rs1554483 C/g; rs11932595 A/g; rs4580704 C/g; rs6843722 A/C; rs6850524 C/g and rs4864548 A/g) encompassing 117 kb of chromosome 4 and representing 115 polymorphic sites (r2 > 0.8) were genotyped. RESULTS: rs11932595 and rs6843722 showed signifi cant associations with NAFLD (empiric P = 0.0449 and 0.023, respectively). A significant association was also observed between clinical or histologic spectrum of NAFLD and rs1554483 (empiric P = 0.0399), rs6843722 (empiric P = 0.0229) and rs6850524 (empiric P = 0.00899) and between fibrosis score and rs1554483 (empiric P = 0.02697), rs6843722 (empiric P = 0.01898) and rs4864548 (empiric P = 0.02697). Test of haplotypic association showed that CLOCK gene variant haplotypes frequencies in NAFLD individuals significantly differed from those in controls (empiric P = 0.0097). CONCLUSION: Our study suggests a potential role of the CLOCK polymorphisms and their haplotypes insusceptibility to NAFLD and disease severity.

In this paper we present the science potential of the enhanced X-ray Timing and Polarimetry(eXTP) mission for studies of strongly magnetized objects. We will focus on the physics and astrophysics of strongly magnetized objects, namely magnetars, accreting X-ray pulsars, and rotation powered pulsars. We also discuss the science potential of eXTP for QED studies. Developed by an international Consortium led by the Institute of High Energy Physics of the Chinese Academy of Sciences, the eXTP mission is expected to be launched in the mid 2020s.

In VivoEvaluations of Emulsion O/W for a New Topical Anti-Aging Formulation: Short-Term and Long-Term Efficacy

This study aims to evaluate the in vivo efficacy in terms of elasticity, hydration and anti-wrinkle enhancement of a new cosmetic cream formulation: Apollinea anti-ageing facial cream. The latter is O/W (oil/water) emulsion with high volume percentages of snail secretion filtrate and donkey milk serum in its composition. The investigation has been performed on (twelve) healthy subjects: 8 (eight) females, and 4 (four) males, all presenting evident skin wrinkles in typical facial areas (e.g. forehead and eye contour). The subjects had to apply about 2 grams of Apollinea anti-ageing facial cream every day. The efficacy of the new cosmetic formulation was assessed before and after the cosmetic treatment, precisely 2 hours, to assess the short-term efficacy, and 40 days after, this to evaluate the product long-term efficacy. Elasticity enhancements have been tested through measurements of the viscoelastic properties of the skin, whereas the hydration parameter through measurements of the skin and subcutaneous tissues dielectric constant. All the latter measurements have been performed with specific instrumentation, as discussed more in detail in the following. To evaluate the anti-wrinkles efficacy instead, we proceeded with digital camera imaging and image analysis processed through ImageJ software. The new Apollinea facial cream cosmetic formulation, based on donkey milk whey and snail secretion filtrate from Helix Aspersa Müller, showed significant enhancement effects on skin elasticity and hydration as well as an important skin-wrinkles-reduction.

Synthesis, Structural Study and Spectroscopic Characterization of a Quinolin-8-Yloxy Derivative with Potential Biological Properties

We have prepared the (5-chloro-quinolin-8-yloxy) acetic acid and characterized it by using infrared, Raman and multi-dimensional nuclear magnetic resonance spectroscopies. The density functional theory (DFT) together with the 6-31g* and 6-311++g** basis sets were used to study its structure and vibrational properties. Three stable conformations of the compound were theoretically determined in gas phase and probably these conformations are present in the solid phase. The harmonic vibrational wavenumbers for the optimized geometries were calculated at the same theory levels. For a complete assignment of the observed bands in the vibrational spectra, the DFT calculations were combined with Pulay’s scaled quantum mechanical force field (SQMFF) methodology in order to fit the theoretical wavenumber values to the experimental ones. Besides, the force constants of the three conformers of (5-chloro-quinolin-8-yloxy) acetic acid were calculated and compared with those obtained by us for the 2-(quinolin-8-yloxy) acetic acid. In addition, the characteristics of the electronic delocalization of those structures were performed by using natural bond orbital (NBO), while the corresponding topological properties of electronic charge density are analysed by employing Bader’s atoms in molecules theory (AIM).

The Dof (DNA-binding with one finger) proteins are a class of plant-specific transcription factors that can trigger several processes involved in plant growth and development, as well as in stress responses. Here, we performed a systematic bioinformatics analysis to characterize all Dof genes in common bean, which included analysis of the genome sequence, conserved protein domains, chromosomal locations, subcellular locations, phylogenetic relationships, gene duplications, and gene expression profiles in different tissues. Bioinformatics analysis revealed 36 putative genes related to PvDof that were classified into seven subfamilies (A, B1, B2, C1, C2, D1, and, D2) by comparative phylogenetic analysis. Based on our genome duplication analysis, a total of 36 genes were found to be distributed on all 11 chromosomes, and they expanded through gene duplication in tandem, suggesting the involvement of segmental duplication events in the evolutionary process. Synteny events and phylogenetic comparisons of the Dof proteins of common bean with those of A. thaliana, O. sativa, and g. max L. led to the identification of several orthologous and paralogous genes, which provided further insight into the diversity of the evolutionary characteristics of genes of this family in other plant species. Expression profiles revealed that most of the PvDof genes were expressed in different tissues, indicating that PvDof genes may be involved in various physiological functions during plant development. The results of this study provide additional information and potential biotechnological resources for further understanding the molecular basis of this gene family and consequently improvement of common bean crops.

Management of hepatitis C infection before and after liver transplantation

Chronic hepatitis C(CHC) is the most common indication for liver transplantation(LT). Aggressive treatment of hepatitis C virus(HCV) infection before cirrhosis development or decompensation may reduce LT need and risk of HCV recurrence post-LT. Factors associated with increased HCV risk or severity of recurrence include older age, immunosuppression, HCV genotype 1 and high viral load at LT. HCV recurrence post-LT leads to accelerated liver disease and cirrhosis development with reduced graft and patient survival. Currently, interferon(IFN)-based regimens can be used in dualagent regimens with ribavirin, in triple-agent antiviral strategies with direct-acting antivirals(e.g., protease inhibitors telaprevir or boceprevir), or before transplant in compensated patients to reduce HCV viral load to prevent or reduce the risk of post-LT recurrence and complications; they cannot be used in patients with decompensated cirrhosis. IFN-based regimens are used in less than half of HCV-infected patients waiting for LT due to extremely low efficacy and poor tolerability. However, antiviral therapy is indicated after LT in patients with histologically confirmed CHC despite tolerability issues. Improvements in side effect management have increased survival in patients achieving therapeutic targets. HCV treatment pre- and post-LT results in significant health care costs especially when lack of efficacy leads to disease worsening, although studies have shown sofosbuvir treatment before LT vs conventional post-LT dual antiviral is cost effective. The suboptimal efficacy and tolerability of IFN-based therapies, plus the significant economic burden, means the need for effective and well tolerated IFN-free antiHCV therapy for pre- and post-LT remains high.

AIM:To verify the methylation status of CDH1, DAPK, COX2, hMLH1 and CDKN2A genes and to evaluate their association with Helicobacter pylori (H. pylori)-cagA+ and Epstein Barr virus (EBV) infections in gastric ***: Methylation-specific PCR (MSP) assay was performed in 89 primary gastric carcinomas (intestinal and diffuse types). Microsatellite instability (MSI) analysis was performed using the BAT26 primer set and PCR products were analyzed with the ABI PRISM 3100 genetic Analyzer using genescan 3.7 software (Applied Biosystems). Detection of H. pylori and genotyping were performed by PCR, using specifi c primers for ureaseC and cagA genes. The presence of EBV was assessed by in situ hybridization. Statistical analyses were performed using the χ2 or Fisher's exact ***: The most frequent hypermethylated gene was COX-2 (63.5%) followed by DAPK (55.7%), CDH1 (51%), CDKN2A (36%) and hMLH1 (30.3%). Intestinal and diffuse adenocarcinomas showed different methylation profiles and there was an association between methylation of E-CDH1 and H. pylori-cagA+ in the intestinal adenocarcinoma type. MSI was correlated with hMLH1 methylation. There was an inverse correlation between DAPK hypermethylation and ***: We found a strong association between CDH1 methylation and H. pylori-cagA+ in intestinal-type gastric cancer, association of MSI and better prognosis and an heterogeneous COX-2 overexpression.
