Functional and structural changes in internal pudendal arteries underlie erectile dysfunction induced by androgen deprivation

Androgen deficiency is strongly associated with erectile dysfunction （ED）. Inadequate penile arterial blood flow is one of the major causes of ED. The blood flow to the corpus cavernosum is mainly derived from the internal pudendal arteries （IPAs）; however, no study has evaluated the effects of androgen deprivation on IPA＇s function. We hypothesized that castration impairs IPAs reactivity and structure, contributing to ED. In our study, Wistar male rats, 8-week-old, were castrated and studied 30 days after orchiectomy. Functional and structural properties of rat IPAs were determined using wire and pressure myograph systems, respectively. Protein expression was determined by Western blot and immunohistochemistry. Plasma testosterone levels were determined using the IMMULITE 1000 Immunoassay System. Castrated rats exhibited impaired erectile function, represented by decreased intracavernosal pressure/mean arterial pressure ratio. IPAs from castrated rats exhibited decreased phenylephrine- and electrical field stimulation （EFS）-induced contraction and decreased acetylcholine- and EFS-induced vasodilatation. IPAs from castrated rats exhibited decreased internal diameter, external diameter, thickness of the arterial wall, and cross-sectional area. Castration decreased nNOS and α-actin expression and increased collagen expression, p38 （Thr180/ryr182） phosphorylation, as well as caspase 3 cleavage. In conclusion, androgen deficiency is associated with impairment of IPA reactivity and structure and increased apoptosis signaling markers. Our findings suggest that androgen deficiency-induced vascular dysfunction is an event involving hypotrophic vascular remodeling of IPAs.

Fluids in subduction zones can have major effects on subduction dynamics. However, geophysical constraints on the scale and impact of fluid flow during continental subduction are still limited. Here we analyze the VP/VSratios in the Western Alpine region, hosting one of the best-preserved fossil continental subduction zones worldwide, to investigate the impact of fluid flow during continental subduction. We found a belt of high VP/VSratios >1.9 on the upper-plate side of the subduction zone,consistent with a partially serpentinized upper-plate mantle, and a belt of unusually low VP/VSratios g past fluid flow along the continental subduction interface. Our results suggest that past fluid flow may have reduced the effective stress along the subduction interface thus favoring continental subduction.

俯冲带中的流体可能对俯冲过程产生重要影响.然而,关于大陆俯冲期间流体规模及其作用的地球物理约束仍然有限.西阿尔卑斯造山带是全球保存最完好的大陆俯冲带之一,为认识大陆俯冲过程中流体作用的影响提供了重要的参考.本文综合多种地球物理方法分析了西阿尔卑斯造山带地区的VP/VS比值,发现在俯冲区的上盘侧存在高VP/VS（>1.9）带,与部分蛇纹岩化的上覆地幔楔位置一致;而在下盘侧存在异常低的VP/VS（<1.7）带,异常深度浅于30km.本文认为,观测到的低VP/VS比值异常是由地壳内广泛分布的富硅岩脉导致,指示了在俯冲时期大陆俯冲界面之上可能存在的富硅流体流动.本文的结果暗示俯冲时期的流体流动可能降低了俯冲界面的有效应力,从而促进了大陆俯冲.

Motorized spiral enteroscopy(MSE)is the latest advance in device-assisted *** events related to MSE were discussed in a recent large systematic review and meta-analysis and were directly compared with those of balloon enteroscopy in a case-matched study and a randomized controlled *** the real-life application of MSE,an unexpected safety issue emerged regarding esophageal injury and the technique has been withdrawn from the global market,despite encouraging results in terms of diagnostic and therapeutic *** conducted an Italian multicenter real-life prospective study,which was prematurely terminated after the withdrawal of MSE from the *** primary goals were the evaluation of MSE performance(both diagnostic and therapeutic)and its safety in routine endoscopic practice,particularly in the early phase of introduction in the endoscopic unit.A subanalysis,which involved patients who underwent MSE after unsuccessful balloon enteroscopy,demonstrated,for the first time,the promising performance of MSE as a rescue *** its remarkable performance in clinical practice and its potential role as a backup technique following a previously failed enteroscopy,it may be more appropriate to refine and enhance MSE in the future rather than completely abandoning it.

Mesenchymal stem cells are multipotent stem cells that reside in many human tissues and *** stem cells are widely used in experimental and clinical regenerative medicine due to their capability to transdifferentiate into various ***,when transplanted,they lose part of their multipotency and immunomodulatory properties,and most of them die after injection into the damaged *** this review,we discuss the potential utility of melatonin in preserving mesenchymal stem cells’survival and function after *** is a pleiotropic molecule regulating critical cell functions including apoptosis,endoplasmic reticulum stress,and *** is also synthesized in the mitochondria where it reduces oxidative stress,the opening of the mitochondrial permeability transition pore and the downstream caspase activation,activates uncoupling proteins,and curtails the proinflammatory *** addition,recent findings showed that melatonin also promotes the formation of tunneling nanotubes and the transfer of mitochondria between cells through the connecting *** mitochondrial dysfunction is a primary cause of mesenchymal stem cells death and senescence and a critical issue for survival after transplantation,we propose that melatonin by favoring mitochondria functionality and their transfer through tunneling nanotubes from healthy to suffering cells could improve mesenchymal stem cellbased therapy in a large number of diseases for which basic and clinical trials are underway.

The application of organic photocatalysts towards the oxidation of pollutants in water is hampered by different limitations such as their insolubility in the ***,we report that the grafting of a photoorganocatalyst into mesoporous silica materials is an ideal approach to obtain effective ***,the photocatalyst 10-phenylphenothiazine(PTH)was easily anchored into three different mesoporous silica-based materials(MSN,MSU-2 and SBA-15)with different particle sizes and pore sizes through an amide bond *** materials were characterized using IR analysis,solid-state X-ray diffraction,porosity and microscopy(SEM and TEM)techniques,showing that PTH is immobilized inside the pores of the materials and its optical properties are maintained after the *** homogeneous PTH was inactive in water media,the three photocatalytic materials were active for the degradation of ***-15-AP-PTH exhibited the highest catalytic performance towards the degradation of acetaminophen and diclofenac under solar irradiation,finding in this manner a new strategy for the decontamination of pollutants.

Extracorporeal shock wave for plantar flexor spasticity in spinal cord injury:A case report and review of literature

BACKgROUND Approximately 65%-78%of patients with a spinal cord injury(SCI)develop any symptom of *** aim of this study was to investigate the tolerability and short-term effects of radial extracorporeal shock wave therapy(r ESWT)on plantar flexor spasticity in a patient with incomplete *** SUMMARY An 18-year-old man with an incomplete SCI completed five sessions of r *** primary outcomes were the changes in ankle-passive range of motion(APROM)and passive resistive force to ankle *** outcomes were assessed at baseline(T0),immediately after treatment(T1)and 1 wk after the end of treatment(T2).The A-PROM increased by 15 degrees at T1 and 25 degrees at T2 compared with *** passive resistive force to ankle dorsiflexion at low velocity decreased by 33%at T1 and 55%at T2 in the gastrocnemius muscle and by 41%at T1 and 39%at T2 in the soleus muscle compared with *** high velocity,it also decreased by 44%at T1 and 30%at T2 in the gastrocnemius muscle compared with ***,in the soleus muscle,the change was minor,with a decrease of 12%at T1 and increased by 39%at T2 compared with *** In this patient,the findings showed that r ESWT combined with conventional therapy was well-tolerated and could be effective in improving A-PROM and passive resistive force to ankle dorsiflexion in the *** randomized controlled clinical trials with longer period of follow-up are necessary to confirm the results obtained in patients with SCI.
