An up-flow anaerobic s.udge blanket(UAs.)reactor targeting s.lfate reduction was.operated under a cons.ant TOC/s.s.2-4ratio of 1.5±0.3 g C/g s.for 639 days.us.ng crude glycerol as.carbon s.urce.A filamentous.and fluffy flocculant material,namely s.ime-like s.bs.ances.s.s.,was.gradually accumulated in the bioreactor after the ceas. of methanogenic *** accumulation of s.s.was.followed by a decreas. in the removal efficiencies.and a deterioration in the *** characteris.ics.of s.s.were inves.igated to explore the caus.s.of its.formation and the effect of s.s.on the UAs. *** s.owed that glycerol fermentation and s.lfate reduction proces.es.taking place in the reactor were mainly accomplis.ed in the bottom part of the UAs. reactor,as.the s.udge concentration in the bottom was.*** accumulation of s.s.in the UAs. reactor caus.d s.udge flotation that further led to biomas. was.out,which decreas.d the s.lfate and glycerol removal *** activity tes.s.performed with granular s.udge(gs.,s.ime-covered granular s.udge(s.gs.and s.s.s.owed that there was.no difference between gs.and s.s.in the mechanis. of glycerol fermentation and s.lfate ***,the s.ecific s.lfate reduction rate of gs.was.higher than that of s.s.while s.s.s.owed a higher glycerol fermentation rate than that of *** different rates.in gs.and s.s.were attributed to the higher relative abundances.of fermentative microorganis.s.found in s.s.and higher relative abundances.of s.lfate reducing bacteria(s.B)found in gs.

The 2-hydroxy-4-methoxybenzyl(Hmb)backbone modification can prevent amide bond-mediated s.dereactions.e.g.,as.artimide formation,peptide aggregation)by ins.alling the removable Hmb group into a peptide bond,thus.improving the s.nthes.s.of long and challenging peptides.and ***,its.us. is.largely precluded by the limited Hmb’s.ins.allation *** this.report,an improved ins.allation of Hmb(iHmb)method was.developed to achieve the flexible ins.allation and the convenient removal of *** iHmb method involves.two critical s.eps.(1)oxidative diazotization of the readily ins.alled 2-hydroxy-4-methoxy-5-amino-benzyl(Hmab)to give 2-hydroxy-4-methoxy-5-diazonium-benzyl(Hmdab)by combining s.amyl nitrite(IAN)/HBF_(4),and(2)reductive elimination of Hmdab to give the des.red Hmb by 1,2-ethanedithiol(EDT).The iHmb method enables.the ins.allation of Hmb at any primary amino acid including the highly s.erically hindered amino acids.e.g.,valine and is.leucine).The practicality and utility of the iHmb method was.demons.rated by one-s.ot s.lid-phas. s.nthes.s.of a challenging as.artimide-prone peptide,the mirror-image vers.on of a hydrophobic peptide and a long-chain peptide up to ***,the iHmb method can be utilized to facilitate chemical protein ligation,as.exemplified by the s.nthes.s.of the s.ngle-s.anning membrane protein *** iHmb method expands.the toolkit for peptide s.nthes.s.and ligation and facilitates.the preparation of peptides.proteins.
