This study is designed to develop Artificial Intelligence(AI)based analysis tool that could accurately detect COVID-19 lung infections based on portable chest x-rays(CXRs).The frontline physicians and radiologists suffer from grand challenges for COVID-19 pandemic due to the suboptimal image quality and the large volume of CXRs.In this study,AI-based analysis tools were developed that can precisely classify COVID-19 lung infection.Publicly available datasets of COVID-19(N=1525),non-COVID-19 normal(N=1525),viral pneumonia(N=1342)and bacterial pneumonia(N=2521)from the Italian Society of Medical and Interventional Radiology(SIRM),Radiopaedia,The Cancer Imaging Archive(TCIA)and Kaggle repositories were taken.A multi-approach utilizing deep learning ResNet101 with and without hyperparameters optimization was employed.Additionally,the fea-tures extracted from the average pooling layer of ResNet101 were used as input to machine learning(ML)algorithms,which twice trained the learning algorithms.The ResNet101 with optimized parameters yielded improved performance to default parameters.The extracted features from ResNet101 are fed to the k-nearest neighbor(KNN)and support vector machine(SVM)yielded the highest 3-class classification performance of 99.86%and 99.46%,respectively.The results indicate that the proposed approach can be bet-ter utilized for improving the accuracy and diagnostic efficiency of CXRs.The proposed deep learning model has the potential to improve further the efficiency of the healthcare systems for proper diagnosis and prognosis of COVID-19 lung infection.

Metabolic puzzle: Exploring liver fibrosis differences in Asian metabolic-associated fatty liver disease subtypes

BACKGROUND Metabolic-associated fatty liver disease(MAFLD)is a liver condition marked by excessive fat buildup in the absence of heavy alcohol use.It is primarily linked with metabolic issues like insulin resistance,obesity,and abnormal lipid levels,and is often observed with other conditions such as type 2 diabetes and cardiovascular disease.However,whether the subtypes of MAFLD based on the metabolic disorder differentially impact liver fibrosis is not well explicated,especially in the Asian population.AIM To compare the severity of liver fibrosis among different MAFLD subtypes.METHODS A total of 322 adult patients of either gender with fatty liver on ultrasound were enrolled between January to December 2021.MAFLD was defined as per the Asian Pacific Association for the Study of the Liver guidelines.Fibrosis-4 index(Fib-4)and nonalcoholic fatty liver disease fibrosis score(NFS)were employed to evaluate liver fibrosis.RESULTS The mean age was 44.84±11 years.Seventy-two percent of the patients were female.Two hundred and seventy-three patients were classified as having MAFLD,of which 110(40.3%)carried a single,129(47.3%)had two,and 34(12.5%)had all three metabolic conditions.The cumulative number of metabolic conditions was related to elevated body mass index,triglyceride(TG)levels,and glycated hemoglobin,lower high-density lipoprotein(HDL)levels,higher liver inflammation(by aspartate aminotransferase andγ-glutamyl transferase),and higher likelihood of fibrosis(by NFS and Fib-4 scores)(P<0.05 for all).The proportion of advanced fibrosis also increased with an increase in the number of metabolic conditions(4.1%,25.5%,35.6%,and 44.1%by NFS and 6.1%,10.9%,17%,and 26.5%by Fib-4 for no MAFLD and MAFLD with 1,2,and 3 conditions,respectively).Among MAFLD patients,those with diabetes alone were the eldest and had the highest mean value of NFS score and Fib-4 score(P<0.05),while MAFLD patients diagnosed with lean metabolic dysfunction exhibited the highest levels of TG and alanine aminotransferase but the lowest HDL level

Vibration-reduced anxiety-like behavior relies on ameliorating abnormalities of the somatosensory cortex and medial prefrontal cortex

Tibetan singing bowls emit low-frequency sounds and produce perceptible harmonic tones and vibrations through manual tapping. The sounds the singing bowls produce have been shown to enhance relaxation and reduce anxiety. However, the underlying mechanism remains unclear. In this study, we used chronic restraint stress or sleep deprivation to establish mouse models of anxiety that exhibit anxiety-like behaviors. We then supplied treatment with singing bowls in a bottomless cage placed on the top of a cushion. We found that unlike in humans, the combination of harmonic tones and vibrations did not improve anxietylike behaviors in mice, while individual vibration components did. Additionally, the vibration of singing bowls increased the level of N-methyl-D-aspartate receptor 1 in the somatosensory cortex and prefrontal cortex of the mice, decreased the level of γ-aminobutyric acid A(GABA) receptor α 1 subtype, reduced the level of CaMKII in the prefrontal cortex, and increased the number of GABAergic interneurons. At the same time, electrophysiological tests showed that the vibration of singing bowls significantly reduced the abnormal low-frequency gamma oscillation peak frequency in the medial prefrontal cortex caused by stress restraint pressure and sleep deprivation. Results from this study indicate that the vibration of singing bowls can alleviate anxiety-like behaviors by reducing abnormal molecular and electrophysiological events in somatosensory and medial prefrontal cortex.

Bandgap tuning using rare earth metals as dopants in ferrite-based photocatalytic materials has received a lot of interest because the Fermi 4f energy of these metals generates a sub-energy state in the bandgap generated by the overlapping of Fe-3d and O-2p orbitals.Herein,dysprosium-doped cobalt-nickel mixed ferrite(D-CNFO)and its graphene-reinforced composite(D-CNFO@G)were prepared and an ideal photocatalyst material for azo dye mineralization was proposed.A cost-effective combination of wetchemical and ultrasonication methods was used to prepare the doped and composite samples.Advanced characterization methodologies were used to scrutinize the optical,compositional,structural,morphological,and photocatalytic characteristics of as-prepared materials.The X-ray diffraction analysis identified the spinel phase's(cubic)structure,while the electronic spectroscopy examination confirmed the prepared samples'rod-like morphology.The UV/visible absorbance spectrum shows the higher light harvesting behavior of the D-CNFO@G in the visible region.The mineralization performance of the DCNFO and D-CNFO@G composites was analyzed using Congo-red(an anionic dye),a well-known azo dye.The D-CNFO@G sample removes Congo-red dye at a rate almost 2.4%faster than the D-CNFO sample.The experiment involving trapping free radicals indicates that hydroxyl radical play s a crucial role in dye degradation.Since the D-CNFO@G catalyst is magnetic and can be isolated easily from the photocatalytic system,it shows an awkward cycle activity of more than 96%after five mineralization tests.The asprepared D-CNFO@G composite is proved as an excellent option for azo dye mineralization because of the combined impacts of rare earth doping,graphene reinforcement and nanotechnology.

Among the most well-known zoonotic maladies,Brucellosis is the one having a lot of clinical presentations.Brucellosis is a disease which is caused by a type of bacteria of the genus Brucella.Brucellosis is common in people who work as shepherd,milkman,butcher,knackers and vertinary assistants.The different types of the tests are used to diagnose brucellosis such as(ELISA)Enzyme Linked Immunosorbent Assay,(RBPT)Rose Bengal Plate Test and many others.From literature it is evident that the picture of brucellosis is not clear in many countries including Pakistan.So the aim of the study is to clear the exact status of the disease in the women of agro ecological zones of KP Pakistan.A multistage sampling will be used by dividing KP in different agro ecological zones and blood will be collected from the women who are in close contact with animals.The serum will be isolated and the samples will be tested for Brucella by commercial serological kits i.e.ELISA kit in the laboratory for the comparison of the disease status in women of different agro ecological zones of KP Pakistan.

旨在克隆牦牛毛球族同源蛋白3(TRIB3)基因并进行生物信息学分析,检测其在牦牛各组织中的表达情况,为后续探究该基因在牦牛中的功能提供理论依据。试验采集牦牛乳腺组织并以其cDNA为模板,克隆牦牛TRIB3基因序列,通过生物信息学分析了解其生物学特征和结构,运用荧光定量PCR(RT-qPCR)技术检测TRIB3基因在牦牛心脏、肝脏、肺脏、骨骼肌、乳腺及脂肪组织中的表达情况。结果显示:牦牛TRIB3基因CDS区全长为1074 bp,共编码357个氨基酸,为不稳定性亲水蛋白;同源性对比结果显示,牦牛TRIB3编码氨基酸与野牦牛的同源性最高;系统进化树显示,野牦牛和野牛与牦牛TRIB3基因亲缘关系最近,最远是小鼠;TRIB3蛋白属于不稳定亲水性蛋白,主要存在于细胞核中,不含信号肽和跨膜结构域;TRIB3蛋白二级结构含有α-螺旋、延伸链、β-转角、无规则卷曲,分别占35.85%、9.52%、448%和50.14%;蛋白互作分析结果显示,TRIB3与组成型光形态建成蛋白1(COP1)、过氧化物酶体增殖物激活受体α(PPARα)、CCAAT增强子结合蛋白β(CEBPβ)、激活转录因子3(ATF3)、DNA损伤诱导转录因子3(DDIT3)、丝氨酸/苏氨酸蛋白激酶2(AKT2)、AKT3和Twist相关蛋白1(TWIST1)之间均有紧密联系。RT-qPCR检测发现TRIB3基因在牦牛的不同组织中均有表达,乳腺组织中的表达量最高。研究结果为进一步探究TRIB3基因在牦牛乳腺中的作用和其生物功能提供一定的理论数据。

Trehalose(Tre)is a non-reducing disaccharide found in many species,including bacteria,fungi,invertebrates,yeast,and even plants,where it acts as an osmoprotectant,energy source,or protein/membrane protector.Despite relatively small amounts in plants,Tre concentrations increase following exposure to abiotic stressors.Trehalose-6-phosphate,a precursor of Tre,has regulatory functions in sugar metabolism,crop production,and stress tolerance.Among the various abiotic stresses,temperature extremes(heat or cold stress)are anticipated to impact crop production worldwide due to ongoing climate changes.Applying small amounts of Tre can mitigate negative physiological,metabolic,and molecular responses triggered by temperature stress.Trehalose also interacts with other sugars,osmoprotectants,amino acids,and phytohormones to regulate metabolic reprogramming that underpins temperature stress adaptation.Transformed plants expressing Tre-synthesis genes accumulate Tre and show improved stress tolerance.Genome-wide studies of Tre-encoding genes suggest roles in plant growth,development,and stress tolerance.This review discusses the functions of Tre in mitigating temperature stress—highlighting genetic engineering approaches to modify Tre metabolism,crosstalk,and interactions with other molecules—and in-silico approaches for identifying novel Tre-encoding genes in diverse plant species.We consider how this knowledge can be used to develop temperature-resilient crops essential for sustainable agriculture.

Size-controlled synthesis of Fe3O4 and Fe3O4@SiO2 nanoparticles and their superparamagnetic properties tailoring

Superparamagnetic properties and fine-tuning of colloidal Fe3O4nanoparticles are important for their widespread biomedical applications. Herein, colloidal Fe3O4nanoparticles(NPs) of different sizes(8–20 nm) were prepared,and their hydrophilization with SiO2shell coating to be Fe3O4@SiO2core-shell had been realized successively.The size of Fe3O4NPs was controlled by different heating rates. Transmission electron microscope(TEM), powder X-ray diffractometry(XRD), and vibrating sample magnetometer(VSM) were performed to examine the morphology, crystallinity, and magnetic properties of the prepared Fe3O4and Fe3O4@SiO2core-shell NPs,respectively. In addition, high resolution transmission electron microscope(HRTEM) results suggested that Fe3O4NPs had well crystallization. Enabled by such, their superparamagnetic properties can be fine-tuned accordingly and cater to their potential applications.

Studies have shown that dysregulated expression of miRNAs is a major underlying event in the onset and progression of many metabolic diseases.The pathological role of miR-548c-3p in various cancer types,including breast cancer,glioma,prostate cancer,and gastric cancer,has been documented.1 However,experimental evidences validating the role of miR-548c-3p in the development of metabolic diseases are still lacking,and no data is available on its diagnostic potential for metabolic syndrome(MS).Recently,we predicted the involvement of miR-548c-3p in the pathogenesis of MS and its progression to cardiovascular diseases(CVD)via the regulation of target genes necessary for the proper function of insulin resistance,mTOR,and JAK/STAT signaling pathways.2 Therefore,we deemed it necessary to engage in wet laboratory analysis and validate these claims in patients with metabolic syndrome compared to healthy individuals.

An Efficient Stacked Ensemble Model for Heart Disease Detection and Classification

Cardiac disease is a chronic condition that impairs the heart’s functionality.It includes conditions such as coronary artery disease,heart failure,arrhythmias,and valvular heart disease.These conditions can lead to serious complications and even be life-threatening if not detected and managed in time.Researchers have utilized Machine Learning(ML)and Deep Learning(DL)to identify heart abnormalities swiftly and consistently.Various approaches have been applied to predict and treat heart disease utilizing ML and DL.This paper proposes a Machine and Deep Learning-based Stacked Model(MDLSM)to predict heart disease accurately.ML approaches such as eXtreme Gradient Boosting(XGB),Random Forest(RF),Naive Bayes(NB),Decision Tree(DT),and KNearest Neighbor(KNN),along with two DL models:Deep Neural Network(DNN)and Fine Tuned Deep Neural Network(FT-DNN)are used to detect heart disease.These models rely on electronic medical data that increases the likelihood of correctly identifying and diagnosing heart disease.Well-known evaluation measures(i.e.,accuracy,precision,recall,F1-score,confusion matrix,and area under the Receiver Operating Characteristic(ROC)curve)are employed to check the efficacy of the proposed approach.Results reveal that the MDLSM achieves 94.14%prediction accuracy,which is 8.30%better than the results from the baseline experiments recommending our proposed approach for identifying and diagnosing heart disease.
