With the help of the solid phase peptide synthesizing method, we synthesized Kalata B2 cyclotide and the final product was purified by preparative hPLC. The sequence of Kalata B2 is GLPVCGETCFGGTCNTPGCSCTWPICTRD and its m/z M + h mass is 2955.53. It had been first isolated from Oldenlandia affinis plant leaves extract which belongs to Rubiaceae family. The synthesis of Kalata B2 has been characterized through UPLC/MS, LC/MS, CD, 1hNMR, and IR.

Redundant CAMTA Transcription Factors Negatively Regulate the Biosynthesis of Salicylic Acid and N-hydroxypipecolic Acid by Modulating the Expression of SARD1 and CBP60g

Two signal molecules, salicylic acid (SA) and N-hydroxypipecolic acid (NhP), play critical roles in plant immunity. The biosynthetic genes of both compounds are positively regulated by master immune-regulating transcription factors SARD1 and CBP60g. however, the relationship between the SA and NhP pathways is unclear. CALMODULIN-BINDING TRANSCRIPTION FACTOR 1 (CAMTA1), CAMTA2, and CAMTA3 are known redundant negative regulators of plant immunity, but the underlying mechanism also remains largely unknown. In this study, through chromatin immunoprecipitation and electrophoretic mobility shift assays, we uncovered that CBP60g is a direct target of CAMTA3, which also negatively regulates the expression of SARD1, presumably via an indirect effect. The autoimmunity of camta3-1 is suppressed by sard1 cbp60g double mutant as well as ald1 and fmo1, two single mutants defective in NhP biosynthesis. Interestingly, a suppressor screen conducted in the camta1/ 2/ 3 triple mutant background yielded various mutants blocking biosynthesis or signaling of either SA or NhP, leading to nearly complete suppression of the extreme autoimmunity of camta1/ 2/ 3, suggesting that the SA and NhP pathways can mutually amplify each other. Together, these results reveal that CAMTAs repress the biosynthesis of SA and NhP by modulating the expression of SARD1 and CBP60g, and that the SA and NhP pathways are coordinated to optimize plant immune response.

在过去的十年里,人们为了了解慢性髓性白血病（CML）的细胞遗传学基础,已经做了许多工作。费城（Ph1）染色体表现为第22号染色体的长臂缺失/易位,其与CML发生之间的关系依然不清楚。过去认为,该不正常的染色体是第22号染色体的长臂缺失（22q—）,现在已经证明,在90—95％的病例中,这是第9和第22号染色体长臂之间的相互易位（9q;22q）。其余的5—10％是变异的易位。其中有些是一部分染色体物质易位到异常的第22号染色体,使其长于正常22号染色体,Ph1染色体因此而不易被分辨出来。这就称为遮蔽的、或隐匿的Ph1染色体。这一术语看来是十分恰当的,因为已经发现在这种情况时,第22号染色体的长臂一部分转移到了另一染色体,从而提示存在着相互易位。

Mangrove Restoration and Regeneration Monitoring in Gulf of Kachchh, Gujarat State, India, Using Remote Sensing and Geo-Informatics

Indian coast harbors richly diverse and critical coastal habitats like coral reefs and mangroves. Mangroves form one of the most important ecosystems of coastal and marine areas. It safeguards the ecology of the coastal areas and provides livelihood opportunities to the fishermen and pastoral families living in these areas. In real sense, mangrove is the Kalpvriksh (divine tree which fulfills all the desires) for the coastal communities. The restoration and plantation of mangroves have received a lot of attentions worldwide. To assess the impact of mangrove plantation activities and to monitor the mangrove regeneration and restoration in various villages, a joint study under the Integrated Coastal Zone Management Project (ICZMP) was taken up by Gujarat Ecology Commission (GEC) and Bhaskaracharya Institute for Space Applications and Geo-Informatics (BISAG) in the Gulf of Kachchh, Gujarat State.?The major objective of this study was to monitor the increase in mangrove cover in coastal areas of Gulf of Kachchh using the Indian Remote Sensing Satellite data of 2005, 2011 and 2014. The mangrove regeneration was monitored using multi-temporal Indian Remote Sensing Satellite (IRS) LISS-III and LISS-IV digital data covering Gulf of Kachchh region. The multi-temporal IRS LISS-III data covering Gulf of Kachchh of October-2005, November-2011 and LISS-IV data of April-2014 was analyzed. The mangrove density and mangrove area in different talukas was estimated based on the analysis of IRS LISS-III digital data. The mangroves have been delineated based on the pink colour observed on satellite images and the area was estimated in the Geographic Information System (GIS) environment. The taluka or block-level mangrove areas were estimated and changes in the areas were monitored during the period of six years from 2005 to 2011. Bacterial infections and multidrug-resistant bacteria are major health burdens in wound *** antimicrobial agents,e.g.,ε-polylysine(ε-PL),provide a broad spectrum of antibacterial properties and support dermal cell ***,ε-PL was incorporated into polycaprolactone(PCL)/gelatin electrospun scaffolds collected at varying rotation ***,the samples were crosslinked using dopamine hydrochloride to provide highly proliferative dressings with broad antimicrobial *** morphological study showed that the electrospun wound dressings were smooth,continuous,and bead-free,with a mean diameter ranging from 267±7 to 331±8 nm for all random and aligned *** fiber alignment of the electrospun PCL/gelatin scaffolds improved their tensile strength and ***,nanofiber mats are highly hydro-philic,which is crucial for an efficient wound *** samples also demonstrated high antimicrobial properties against common wound bacterial strains,including methicillin-resistant Staphylococcus aureus(MRSA),Staphylococcus aureus(SA),Escherichia coli(EC),Acinetobacter baumannii(AB),and Pseudomonas aeruginosa(PA).Mammalian cell prolifera-tion and morphology assays involving primary human dermal fibroblasts(hDFs)and immortalized keratinocytes(haCaT)showed excellent biocompatibility of the electrospun mats and remarkably aligned ***,aligned mats showed more cell migration than randomly oriented mats,which is desirable for more efficient wound ***,it can be concluded that aligned PCL/gelatin mats containingε-PL are promising for potential use in wound dressings.

Soil and water conservation measures improve soil carbon sequestration and soil quality under cashews

Land degradation is becoming a serious problem in the west coast region of India where one of the world’s eight biodiversity hotspots, the ‘Western Ghats’, is present. Poor land management practices and high rainfall have led to increasing problems associated with land degradation. A long-term(13-year)experiment was done to evaluate the impact of soil and water conservation measures on soil carbon sequestration and soil quality at three different depths under cashew nut cultivation on a 19% slope. Five soil and water conservation measures-continuous contour trenches, staggered contour trenches, halfmoon terraces, semi-elliptical trenches, and graded trenches all with vegetative barriers of Stylosanthes scabra and Vetiveria zizanoides and control were evaluated for their influence on soil properties,carbon sequestration, and soil quality under cashews. The soil and water conservation measures improved significantly the soil organic carbon, soil organic carbon stock, carbon sequestration rate and microbial activity compared to the control condition(without any measures). Among the measures tested, continuous contour trenches with vegetative barriers outperformed the others with respect to soil organic carbon stock, sequestration rate, and microbial activity. The lower metabolic quotient with the measures compared to the control indicated alleviation of environmental stress on microbes. Using principal component analysis and a correlation matrix, a minimum dataset was identified as the soil available nitrogen, bulk density, basal soil respiration, soil p h, acid phosphatase activity, and soil available boron and these were the most important soil properties controlling the soil quality. Substituted 2-benzylidene-1-benzofuran-3-ones are commonly known as aurones. This class of bioactive heterocycles belongs to flavonoid family. The article intends to put forth the rational design and synthesis of a new series of aurones using 3’,5’-dibromo-2’,4’-dihydroxychalcones and copper bromide in presence of DMF-water mixture (8:2, v/v) for the first time. Preliminary bioassay shows that most of compounds have good trypanocidal activity against Trypanosoma cruzi at 10 μg/mL. Few compounds are equally potent to the standard drugs Benznidazole and Nifurtimox. The structures of the newly synthesized products 2a-n were established by elemental analysis, FTIR, 1h NMR, 13C NMR and mass spectroscopic studies.

Background: Epstein-Barr virus (EBV)has been implicated in B-cell lymphoma associated with iatrogenic immune dysregulation, primarily in the context of extracutaneous lymphoma. Methods: We describe six patients, five transplant recipients receiving cyclosporine and one patient with rheumatoid arthritis receiving methotrexate,who developed cutaneous presentations of EBV-associated B-cell lymphoma. human herpesvirus 8 (hhV8)and EBV thymidine kinase (vTK)expression were also explored. Results: The cases comprised plasmablastic lymphoma (one case), plasmacytic marginal zone lymphoma (two cases), and diffuse large B-cell lymphoma (three cases). There was a monoclonal gammopathy in one and concurrent extracutaneous disease in two of the six patients. EBV-associated latent small nuclear RNA was detected in all cases with coexpression of hhV8 in one of the five cases and of vTK in three of the six cases. Three patients responded to a reduction in the immunosuppressive regimen and antiviral therapy. Recurrent disease developed in two, with one patient succumbing to multiorgan dissemination. Conclusions: EBV-associated cutaneous B-cell lymphoma is characterized by a long interval between the initiation of immunosuppression and the development of lymphoma. Although previous reports have reported an indolent clinical course, an aggressive clinical course may occur. hhV8 and lytic phase EBV antigens are detected in some cases, possibly suggesting a pathogenetic role.
