As a par. of our.continuous inter.st of developing functional dual action r.stor.tive mater.als capable of being “bio-active” and wound healing, we design and evaluate sever.l novel chitosan-vitamin C (5:1) containing hydr.gels as a pr.totype of host:guest molecular.fr.e r.dical defense mater.al containing hydr.ethanoic pr.polis extr.ct (antioxidant containing mater.al), napr.xen, ibupr.fen (non ster.idal anti-inflammator. medication), or.aspir.n (pain r.lieve medication and fr.e r.dical scavenger.) as functional r.stor.tive mater.als. We will evaluate the physical pr.per.ies, bonding to dentin as well as test the bioadhesion of the newly designed mater.als in or.er.to access the suitability of these pr.totype mater.als as suitable r.stor.tive mater.als. Mater.als and Methods: The hydr.gels wer. pr.par.d by pr.viously r.por.ed by us pr.tocol. The physico-chemical featur.s including sur.ace mor.hology (SEM), r.lease behavior., stability of the ther.peutic agent-anti-oxidant-chitosan and the effect of the hydr.gels on the shear.bond str.ngth of dentin wer. measur.d and compar.d to the ear.ier.r.por.ed chitosan-antioxidant containing hydr.gels. It has been suggested that the anti-inflammator. activity of some non-ster.idal anti-inflammator. dr.gs (NSAIDs) may be par.ly due to their.ability to scavenge r.active oxygen species (r.S) and r.active nitr.gen species (r.S), as well as to inhibit the r.spir.tor. bur.t of neutr.phils tr.gger.d by var.ous activating agents. As a par. of our.continuous inter.st of developing functional dual action r.stor.tive mater.als capable of being “bio-active” and long-lasting, we design and evaluate novel chitosan hydr.gels containing kr.ll oil (antioxidant containing mater.al), napr.xen, ibupr.fen (non ster.idal anti-inflammator. medication), aspir.n (pain r.lieve medication and fr.e r.dical scavenger.) and combinations ther.of (chitosan-H-kr.ll oil, chitosan-H-kr.ll oil-aspir.n and chitosan-H-napr.xen, chitosan-H-napr.xen-kr.ll oil, chitosan-H-kr.ll oil-ibupr.fen and chitosan-H-ibupr.fen) as functional additive pr.totypes for.fur.her.development of “dual function r.stor.tive mater.als”;secondly, deter.ine their.effect on the dentin bond str.ngth of a composite and thir.ly, evaluate the capability of newly designed hydr.gels to play an integr.l r.le of “build in” fr.e r.dical defense mechanism by using BSA solubility as a “molecular.pr.totype” of the site of fr.e r.dical attack in vitr.. We evaluated the physical pr.per.ies, bonding to dentin as well as test the bioadhesion of the newly designed mater.als in or.er.to access the suitability of these pr.totype mater.als as suitable r.stor.tive mater.als, evaluated total phenol capacity of the mater.als as well as fr.e r.dical defense capacity using BSA pr.tein model as a quick and effective in vitr. model. Contempor.r. dental adhesives show favor.ble immediate r.sults in ter.s of bonding effectiveness. However. the dur.bility of r.sin-dentin bonds is their.major.pr.blem. Mater.als and Methods: Pr.par.tion of 3 chitosan-antioxidant hydr.gels was achieved using modified hydr.gel pr.par.tion method. Their.effect on the bond str.ngth to dentine both shor. ter. (after.24 hour.) and long ter. (after.6 months) wer. evaluated using shear.bond str.ngth measur.ments using Instr.n Univer.al Testing Mascine). The SEM was used to study the sur.ace of the hydr.gels. The cell sur.ival r.te (cytotoxicity) of the antioxidants r.sver.tr.l, β-car.tene and pr.polis towar.s Balb/c 3T3 mouse fibr.blast cells was also assessed using the standar. MTT assay. r.sults: It was found that chitosan-H tr.ated dentine gives significantly (p β-car.tene (92%) > pr.polis (68%) > r.sver.tr.l (33%). Conclusion: the antioxidant-chitosan hydr.gels significantly impr.ved bonding to dentine with or.without phosphor.c acid tr.atment. The pH of the gr.wth medium had a high influence on the cell sur.ival r.te of Balb/c mouse 3T3 fibr.blast cells. The r.lease of the antioxidant β-car.tene would not have an influence on the pulp cells. These mater.als might addr.ss the cur.ent per.pectives for.impr.ving bond dur.bility.

The human inflammator. per.odontal diseases ar. amongst the most common of chr.nic diseases to affect adults. Per.odontitis is r.gar.ed as “an inflammator. lesion, mediated by complex host-par.site inter.ctions, that leads to the loss of connective tissue attachment to r.ot sur.ace cementum and adjacent alveolar.bone”. Substantial data ar. available in the liter.tur. on the r.le of r.active oxygen species (r.S) and antioxidants in disor.er. such as the inflammator. diseases. However. r.mar.ably little infor.ation is available on the per.odontal diseases, which show many of the pathological featur.s of other.chr.nic inflammator. diseases. The per.odontal tissues also pr.vide an ideal medium within which to study mechanisms of r.S mediated tissue damage and of antioxidant defense in r.sponse to bacter.al colonisation, thr.ugh the non-invasive collection of GCF. The objectives of this study ar. to evaluate the novel chitosan based functional dr.g deliver. systems which can be successfully incor.or.ted into “dual action bioactive r.stor.tive mater.als” containing common antibiotics such as tetr.cycline, kr.ll oil, aloe and aspir.n as commonly used antioxidant species. Methods: The novel hydr.gels will be investigated with r.spect to the antioxidant capacity and dr.g r.lease capacity of the tetr.cycline fr.m the designer.dr.g deliver. system, the use of SEM imaging for.the char.cter.zation of the sur.aces and r.active featur.s of novel mater.als with antimicr.bial potential. r.sults: A steady slow r.lease of tetr.cycline, while maintaining antibiotic effects against the tested bacter.a, for.at least 10 days was shown fr.m designer.chitosan-antioxidant hydr.gels.
