Expression of connexin 30 and connexin 32 in hippocampus of rat during epileptogenesis in a kindling model of epilepsy

Objective Understanding the molecular and cellular mechanisms underlying epileptogenesis yields new in- sights into potential therapies that may ultimately prevent epilepsy. Gap junctions （GJs） create direct intercellular conduits between adjacent cells and are formed by hexameric protein subunits called connexins （Cxs）. Changes in the expression of Cxs affect GJ communication and thereby could modulate the dissemination of electrical discharges. The hippocampus is one of the main regions involved in epileptogenesis and has a wide network of GJs between different cell types where Cx30 is expressed in astrocytes and Cx32 exists in neurons and oligodendrocytes. In the present study, we evaluated the changes of Cx30 and Cx32 expression in rat hippocampus during kindling epileptogenesis. Methods Rats were stereotaxically implanted with stimulating and recording electrodes in the basolateral amygdala, which was electrically stimulated once daily at afterdischarge threshold. Expression of Cx30 and Cx32, at both the mRNA and protein levels, was measured in the hippocampus at the beginning, in the middle （after acquisition of focal seizures）, and at the end （after establishment of generalized seizures） of the kindling process, by real-time PCR and Western blot. Results Cx30 mRNA expression was upregulated at the beginning of kindling and after acquisition of focal seizures. Then it was downregulated when the animals acquired generalized seizures. Overexpression of Cx30 mRNA at the start of kindling was consistent with the respective initial protein increase. Thereafter, no change was found in protein abundance during kindling. Regarding Cx32, mRNA expression decreased after acquisition of generalized seizures and no other significant change was detected in mRNA and protein abundance during kindling. Conclusion We speculate that Cx32 GJ communication in the hip- pocampus does not contribute to kindling epileptogenesis. Sustainable functionalization and modification of materials via multicomponent reactions in water

In materials chemistry,green chemistry has established firm ground providing essential design criteria to develop advanced tools for efficient functionalization and modification of materials.Particularly,the combination of multicomponent reactions in water and aqueous media with materials chemistry unlocks a new sustainable way for constructing multi-functionalized structures with unique features,playing significant roles in the plethora of applications.Multicomponent reactions have received significant consideration from the community of material chemistry because of their great efficiency,simple operations,intrinsic molecular diversity,and an atom and a pot economy.Also,by rational design of multicomponent reactions in water and aqueous media,the performance of some multicomponent reactions could be enhanced by the contributing“natural”form of water-soluble materials,the exclusive solvating features of water,and simple separating and recovering materials.To date,there is no exclusive review to report the sustainable functionalization and modification of materials in water.This critical review highlights the utility of various kinds of multicomponent reactions in water and aqueous media as green methods for functionalization and modification of siliceous,magnetic,and carbonaceous materials,oligosaccharides,polysaccharides,peptides,proteins,and synthetic polymers.The detailed discussion of synthetic procedures,properties,and related applicability of each functionalized/modified material is fully deliberated in this review.

Dear Editor,We were interested to read the paper by Kuzmanovic Elabjer et al.The purpose of the authors was to assess ultrasound reliability in detecting retinal tears in patients with acute symptomatic posterior vitreous detachment （ASPVD）. They performed transpalpebral ultrasound of the eye and the orbit followed by fundus examination initially and in 6wk period.

Comparison of the Use of Conventional and Antibiotic-Coated Tracheal Tubes and EVAC on the Incidence of Ventilator-Associated Pneumonia (VAP)

Ventilator-associated pneumonia (VAP) is one of the most important hospital infections in hospitalized patients, which is associated with increased mortality and patient costs. The tracheal tube itself seems to be a major risk factor for VAP. Contaminated secretions pass through the endotracheal tube and reach the lungs. Also, bacteria form a bacterial biofilm on the tracheal tube and are transferred from there to the lungs. Different tracheal tube designs have been produced to overcome these cases. The purpose of this study is to investigate the effect of an Evac tracheal tube covered with antibiotics and normal on the incidence of pneumonia caused by the ventilator. Research method: 180 patients were randomly intubated in three groups of 60 people with three types of tracheal tubes, Evac and Bactiguard. Clinical examinations, endotracheal tube aspiration culture, and chest radiography were obtained from the patients and the incidence of VAP was calculated based on the CPIS standard. The relationship between the type of endotracheal tube and the incidence of VAP and the length of stay in the intensive care unit (ICU) and being discharged alive from the ICU were investigated. Findings and conclusions: The average incidence of VAP for the group of patients intubated with a conventional tracheal tube was 50%, EVAC was 45% and Bactiguard was 40%. The results showed that there is no significant relationship between the incidence of ventilator-induced pneumonia and the type of tracheal tube. The incidence of ventilator-induced pneumonia was not significantly reduced by suctioning subglottic secretions and Bactiguard tracheal tubes. It seems that using one method alone is not effective in reducing ventilator-induced pneumonia.

It seems that defects on micro-minerals levels have an etiologic role involved in type 1 diabetes mellitus pathogenicity. The aim of our study were to evaluate the serum levels of zinc and iron and investigate their possible relationship between these biochemical parameters with demographic conditions and glycemic control in patients with type 1 diabetes mellitus disorder. In this case-control based study, 68 female with type 1 diabetes mellitus with a mean age of 52.2 ± 2 as case group and 122 healthy women as a control group with a mean age of 49/4 ± 3/2 were investigated. for biochemical analysis,10 mL fasting venous blood sample were obtained from each subjects. FBS (fasting blood glucose), fructosamine (glycemic control parameter) were determined (spectrophotometry method, (pars azmoon, Iran), nitroblue tetrazoline method respectively). serum zinc level with colorimetric method (Biorex-UK) and serum iron with photometric method (pars azmoon, Iran) were determined. Statistical analysis using SPSS software performed. Significant levels considered as P < 0.05. According to this study there is statistically significant difference between serum levels of iron and zinc in patients with type 1 diabetes compared to controls. indeed serum level of iron and zinc had lower level in patient group toward controls. In patients group, there are a positive correlation between age and decreased level of serum zinc (P < 0.05). Also there was a significant negative correlation between the glycemic control status and serum zinc. Other studied parameters concluded BMI (body mass index), Weight and height have not significant difference between groups. The decrease in serum iron and zinc level in women with type 1 diabetes may be related to low dietary intake or increased excretion of micro minerals or the presence of confounding factors that require more extensive intervention studies to confirm it.
