Ochratoxin A induces mitochondrial apoptosis and ferroptosis by inhibiting sigma-1 receptor to disrupt redox and cholesterol homeostasis

Ochratoxin A (OTA)， a secondary fungal metabolite known for its nephrotoxic effects， is widespread in various foods and animal feeds. Our recent investigation suggests a correlation between OTA-induced nephrotoxicity and sigma-1 receptor (sig-1R)-mediated mitochondrial apoptosis in human proximal tubule epithelial-originated kidney-2 (HK-2) cells. However， the involvement of sig-1R in OTA-induced nephrotoxicity， encompassing other forms of regulated cell death like ferroptosis， remains unexplored. In this research， cell viability， apoptotic rate， cholesterol levels， mitochondrial glutathione (mGSH) levels， reactive oxygen species (ROS) levels， and protein expressions in HK-2 cells treated with OTA and/or blarcamesine hydrochloride (Anavex 2-73) were evaluated. The results suggest that OTA induces mitochondrial apoptosis and ferroptosis by inhibiting sig-1R， subsequently promoting sterol regulatory element-binding protein 2 (SREBP2)， 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR)， GRAM domain-containing protein 1B (GRAMD1B)， steroidogenic acute regulatory protein， mitochondrial (StARD1)， 78 kDa glucose-regulated protein (GRP78)， CCAAT/enhancer-binding protein (C/EBP) homologous protein (CHOP)， cyclophilin D (CypD)， Cleaved-Caspase-3， B-cell lymphoma-2-associated X protein (BAX)， and long-chain fatty acid-CoA ligase 4 (ACSL4)， inhibiting tumor necrosis factor receptor-associated protein 1 (TRAP1)， mitochondrial 2-oxoglutarate/malate carrier protein (SLC25A11)， B-cell lymphoma-2-like protein 1 (BCL-XL)， and glutathione peroxidase 4 (GPX4)， reducing mGSH levels， and increasing total cholesterol， mitochondrial cholesterol， and ROS levels. In conclusion， OTA induces mitochondrial apoptosis and ferroptosis by inhibiting sig-1R， thereby disrupting redox and cholesterol homeostasis in vitro. The regulation of cholesterol homeostasis by sig-1R and its involvement in OTA-induced mitochondrial apoptosis and ferroptosis are reported here for the first time.

量化居民点空间分布离散度并探索其与农村经济指标的关联关系,以期为＂整村推进＂扶贫开发规划的实施提供指导意见。以国家扶贫重点县——鹤峰县为研究区,以行政村为评价单元,分别从人文和自然角度引用离散度综合指数（DCI）和景观综合指数（LCI）结合空间探索性分析方法,对比分析鹤峰县居民点空间分布的离散特点,并采用线性回归模型和空间滞后回归模型剖析鹤峰县整体及不同自然与经济条件下离散度与农民人均纯收入之间的相互影响关系。研究结果表明：1）整体上,DCI与LCI均存在空间聚集效应,但两者空间分布特征存在差异,DCI评价结果介于0.59~4.56,LCI评价结果介于0.14~2.61,DCI可以更好地反映离散度。2）通过空间滞后回归模型对DCI与农民人均纯收入分析,得到两者间呈明显相关关系。3）DCI与道路密度、经济圈层、坡度均存在相关关系,同时反映收入变化趋势;LCI在经济圈层等级下未表现出此特征。对于＂整村推进＂扶贫项目的实施,既要重视道路等人文要素的影响,也要考虑行政村之间的联系。

Fully endoscopic far-lateral supracerebellar infratentorial approach for trigeminal neuralgia: illustrative case reports

Background Trigeminal neuralgia (TN) is a common cause of craniofacial pain. The retrosigmoid approach is usually used to treat TN, but no cases of endoscopic far-lateral supracerebellar infratentorial approach (EF-SCITA) were used to undergo operation for *** presentation Two patients were presented with severe facial pain and preliminary diagnosis was TN. Preoperative magnetic resonance imaging revealed that a superior cerebellar artery (SCA) compressed the trigeminal nerve in case 1, and a tumor located in the petrous apex extending into the Meckel’s cave compressed the trigeminal nerve in case 2. Operations were achieved through the EF-SCITA. The pain was totally relieved with no postsurgical complications in both *** We present the first two case reports of EF-SCITA to relieve classical and secondary TN successfully. The EF-SCITA can be a promising approach for treating TN.

Application of interventional therapy via hepatic artery in pancreatic neuroendocrine neoplasms liver metastases

Pancreatic neuroendocrine neoplasm (PNEN) is the second most common malignant tumor of the pancreas. It has the characteristic of high metastases rate, and the liver is the most common site for metastasis. Metastasis affects prognosis and survival seriously. A number of earlier studies have shown that the interventional therapy via hepatic artery could reduce hepatic tumor burden and hormone secretion safely and rapidly, significantly improve objective response rate (ORR), and enhance the efficacy and prolong the survival time when combined with system therapy. The interventional therapy via hepatic artery plays an important role in the treatment of PNEN liver metastases. Interventional therapy via hepatic artery could possibly increase ORR, prolong progression-free survival, and even overall survival for appropriate patients.

While heteroatom doping serves as a powerful strategy for devising novel polycyclic aromatic hydrocarbons(PAHs), the further fine-tuning of optoelectronic properties via the precisely altering of doping patterns remains a challenge. Herein, by changing the doping positions of heteroatoms in a diindenopyrene skeleton, we report two isomeric boron, sulfur-embedded PAHs, named Anti-B_(2)S_(2) and Syn-B_(2)S_(2), as electron transporting semiconductors. Detailed structure-property relationship studies revealed that the varied heteroatom positions not only change their physicochemical properties, but also largely affect their solid-state packing modes and Lewis base-triggered photophysical responses. With their low-lying frontier molecular orbital levels, n-type characteristics with electron mobilities up to 1.5 × 10^(-3)cm^(2)V^(-1)s^(-1)were achieved in solution-processed organic field-effect transistors. Our work revealed the critical role of controlling heteroatom doping patterns for designing advanced PAHs.

Wearable electromagnetic interference(EMI)shielding fabrics with excellent electromagnetic shielding performance,oxidation resistance,and structural stability are highly demanded for the rapid development of electronic devices and wireless *** are metallic conductive materials with exceptional EMI shielding properties,but they are prone to oxidation in air and have poor structural stability and durability on fabric ***,we present a one-step assembly method to fabricate fabrics coated with MXenes and polymeric sodium alginate(SA)composite(MXene-SA).SA protects MXenes from oxidation and forms a stable interlayer structure by bonding to *** MXene-SA coated fabrics are breathable and flexible,and have a low sheet resistance of 2.12±0.08Ω/sq and a high EMI shielding performance of 37.05 dB at X-band,which is comparable to the best 42.31 ***,the MXene-SA coated fabrics exhibit high structural stability and oxidation resistance under various conditions of sonication disintegration,mechanical abuse,chemical corrosion,and humidity,compared to pure MXenes coated *** believe that the wearable and high-performance MXene-SA fabrics have great potential for the next generation of ultra-portable and wearable EMI shielding products.

Targeted regeneration and upcycling of spent graphite by defect‐driven tin nucleation

The recycling of spent batteries has become increasingly important owing to their wide applications,abundant raw material supply,and sustainable *** with the degraded cathode,spent anode graphite often has a relatively intact structure with few defects after long ***,most spent graphite is simply burned or discarded due to its limited value and inferior performance on using conventional recycling methods that are complex,have low efficiency,and fail in performance ***,we propose a fast,efficient,and“intelligent”strategy to regenerate and upcycle spent graphite based on defect‐driven targeted *** Sn as a nanoscale healant,we used rapid heating(~50 ms)to enable dynamic Sn droplets to automatically nucleate around the surface defects on the graphite upon cooling owing to strong binding to the defects(~5.84 eV/atom),thus simultaneously achieving Sn dispersion and graphite *** a result,the regenerated graphite showed enhanced capacity and cycle stability(458.9 mAh g^(−1) at 0.2 A g^(−1) after 100 cycles),superior to those of commercial *** from the self‐adaption of Sn dispersion,spent graphite with different degrees of defects can be regenerated to similar structures and *** analysis indicates that targeted regeneration and upcycling have significantly lower energy consumption(~99%reduction)and near‐zero CO_(2) emission,and yield much higher profit than hydrometallurgy,which opens a new avenue for direct upcycling of spend graphite in an efficient,green,and profitable manner for sustainable battery manufacture.
