Optimization of the Process for Preparing Bivalent Polysaccharide Conjugates to Develop Multivalent Conjugate Vaccines against Streptococcus pneumoniae or Neisseria meningitidis and Comparison with the Corresponding licensed Vaccines in Animal Models

Objective:This study aimed to describe,optimize and evaluate a method for preparing multivalent conjugate vaccines by simultaneous conjugation of two different bacterial capsular polysaccharides(CPs)with tetanus toxoid(TT)as bivalent conjugates.Methods:Different molecular weights(MWs)of polysaccharides,activating agents and capsular polysaccharide/protein(CP/Pro)ratio that may influence conjugation and immunogenicity were investigated and optimized to prepare the bivalent conjugate bulk.Using the described method and optimized parameters,a 20-valent pneumococcal conjugate vaccine and a bivalent meningococcal vaccine were developed and their effectiveness was compared to that of corresponding licensed vaccines in rabbit or mouse models.Results:The immunogenicity test revealed that polysaccharides with lower MWs were better for Pn1-TT-Pn3 and MenA-TT-MenC,while higher MWs were superior for Pn4-TT-Pn14,Pn6A-TT-Pn6B,Pn7F-TT-Pn23F and Pn8-TT-Pn11A.For activating polysaccharides,1-cyano-4-dimethylaminopyridinium tetrafluoroborate(CDAP)was superior to cyanogen bromide(CNBr),but for Pn1,Pn3 and MenC,N-(3-dimethylaminopropyl)-N’-ethylcarbodiimide hydrochloride(EDAC)was the most suitable option.For Pn6A-TT-Pn6B and Pn8-TT-Pn11A,rabbits immunized with bivalent conjugates with lower CP/Pro ratios showed significantly stronger CP-specific antibody responses,while for Pn4-TT-Pn14,higher CP/Pro ratio was better.Instead of interfering with the respective immunological activity,our bivalent conjugates usually induced higher IgG titers than their monovalent counterparts.Conclusion:The result indicated that the described conjugation technique was feasible and efficacious to prepare glycoconjugate vaccines,laying a solid foundation for developing extended-valent multivalent or combined conjugate vaccines without potentially decreased immune function.

Atherosclerosis is a leading cause of mortality and morbidity worldwide.Despite the challenges in managing atherosclerosis,researchers continue to investigate new treatments and complementary therapies.Cordyceps is a traditional Chinese medicine that has recently gained attention as a potential therapeutic agent for atherosclerosis.Numerous studies have demonstrated the effectiveness of cordyceps in treating atherosclerosis through various pharmacological actions,including anti-inflammatory and antioxidant activities,lowering cholesterol,inhibiting platelet aggregation,and modulating apoptosis or autophagy in vascular endothelial cells.Notably,the current misuse of the terms cordyceps and Ophiocordyceps sinensis has caused confusion among researchers,and complicated the current academic research on cordyceps.This review focuses on the chemical composition,pharmacological actions,and underlying mechanisms contributing to the anti-atherosclerotic effects of cordyceps and the mycelium of Ophiocordyceps spp.This review provides a resource for the research on the development of new drugs for atherosclerosis from cordyceps.

Dynamical switchable quantum nonreciprocity induced by off-resonant chiral two-photon driving

Optical nonreciprocity,which refers to the direction-dependent emission,scattering and absorption of photons,plays a very important role in quantum engineering and quantum information processing.Here,we propose an all-optical approach to achieve the optical dynamical switchable quantum nonreciprocity by an off-resonant chiral two-photon driving in a single microring cavity,which differs from the conventional nonreciprocal schemes.It is shown that the optical field with time-dependent statistical properties can be generated and the nonreciprocity flips periodically,with switchable photon blockade and photon-induced tunneling effects.We find that the dynamical system is robust and immune to the parameter variations,which loosens the parameter range of system.Meanwhile,the time window for one-way quantum information is sufficiently wide and tunable.Our work opens a new idea for the current quantum nonreciprocal research,which can facilitate a memory functionality and be used for future inmemory superconducting quantum compute.The other nonreciprocal quantum devices,i.e.,dynamical switchable nonreciprocal squeezing and entanglement,may be inspired by our method,which is expected to have important applications in future quantum technology.

Nanoelectrodes have been shown to be powerful tools for nanoscopic research on single entities,deepening the understanding of heterogeneity and stochastics for single molecules,nanoparticles and cells[1–3].However,despite their importance,current nanoelectrode preparations are highly restrained to flame/electrochemical etching,metal sputtering,electrochemical deposition,etc.[3],resulting in a limited range of materials(mainly gold,platinum and carbon)for potential-resolved detection of redox-active species(Table S1 online).The enrichment of preparation methods to introduce novel materials and mechanisms should prove significant for the development of nanoelectrodes.

Highly elastic and degradable vitrimeric elastomers using polycondensation

Introducing covalently crosslinked network to polymer matrix can merge the advantages in reprocessing and durability of polymers.In this contribution,a series of high-performance vitrimeric elastomers were achieved via polycondensation.The topological structures of polymers were tuned by varying the feeding ratios of bisacetoacetate,hex–substituted bisacetoacetate,bisamine and tris(2-aminoethyl)amine.With these structural manipulations,the vitrimeric elastomers presented great elastic recovery properties(strain recovery value up to 80%)benefiting from the introduction of long chain branch.Furthermore,the elastomers exhibited excellent reprocessing property,water vapor/oxygen barrier and adhesive properties.Specially,the elastomers could be degraded into monomer under acid conditions which enabled the elastomer synthesis again in closed loop recycling system.The ease of the polycondensation in this work to prepare highly elastic and recyclable vitrimeric elastomers demonstrated exciting opportunities for the synthesis of sustainable polymers.

Shengmaisan combined with liuwei Dihuang Decoction alleviates chronic intermittent hypoxia-induced cognitive impairment by activating the EPO/EPOR/JAK2 signaling pathway

Chronic intermittent hypoxia(CIH),a principal pathophysiological aspect of obstructive sleep apnea(OSA),is associated with cognitive deficits.Clinical evidence suggests that a combination of Shengmaisan and liuwei Dihuang Decoctions(SMS-LD)can enhance cognitive function by nourishing yin and strengthening the kidneys.This study aimed to assess the efficacy and underlying mechanisms of SMS-LD in addressing cognitive impairments induced by CIH.We exposed C57BL/6N mice to CIH for five weeks(20%-5%O_(2),5 min/cycle,8 h/day)and administered SMS-LD intragastrically(15.0 or 30 g·kg^(-1)·day)30 min before each CIH session.Additionally,AG490,a JJanus kinase 2(JAK2)inhibitor,was administered via intracerebroventricular injection.Cognitive function was evaluated using the Morris water maze,while synaptic and mitochondrial structures were examined by transmission electron microscopy.Oxidative stress levels were determined using DHE staining,and the activation of the erythropoietin(ER)/ER receptor(EPOR)/JAK2 signaling pathway was analyzed through immunohistochemistry and Western blotting.To further investigate molecular mechanisms,HT22 cells were treated in vitro with either SMS-LD medicated serum alone or in combination with AG490 and then exposed to CIH for 48 h.Our results indicate that SMS-LD significantly mitigated CIH-induced cognitive impairments in mice.Specifically,SMS-LD treatment enhanced dendritic spine density,ameliorated mitochondrial dysfunction,reduced oxidative stress,and activated the EPO/EPOR/JAK2 signaling pathway.Conversely,AG490 negated SMS-LD’s neuroprotective and cognitive improvement effects under CIH conditions.These findings suggest that SMS-LD’s beneficial impact on cognitive impairment and synaptic and mitochondrial integrity under CIH conditions may predominantly be attributed to the activation of the EPO/EPOR/JAK2 signaling pathway.

Atomically Substitutional Engineering of Transition Metal Dichalcogenide Layers for Enhancing Tailored Properties and Superior Applications

Transition metal dichalcogenides(TMDs)are a promising class of layered materials in the post-graphene era,with extensive research attention due to their diverse alternative elements and fascinating semiconductor behavior.Binary MX2 layers with different metal and/or chalcogen elements have similar structural parameters but varied optoelectronic properties,providing opportunities for atomically substitutional engineering via partial alteration of metal or/and chalcogenide atoms to produce ternary or quaternary TMDs.The resulting multinary TMD layers still maintain structural integrity and homogeneity while achieving tunable(opto)electronic properties across a full range of composition with arbitrary ratios of introduced metal or chalcogen to original counterparts(0–100%).Atomic substitution in TMD layers offers new adjustable degrees of freedom for tailoring crystal phase,band alignment/structure,carrier density,and surface reactive activity,enabling novel and promising applications.This review comprehensively elaborates on atomically substitutional engineering in TMD layers,including theoretical foundations,synthetic strategies,tailored properties,and superior applications.The emerging type of ternary TMDs,Janus TMDs,is presented specifically to highlight their typical compounds,fabrication methods,and potential applications.Finally,opportunities and challenges for further development of multinary TMDs are envisioned to expedite the evolution of this pivotal field.

significance of Diffusion Tensor Imaging of Vastus Medialis Oblique in Recurrent Patellar Dislocation

Background： Numerous studies have investigated the influence of osseous factors on patellofemoral joint instability, but research on the influence of dynamic muscle factors in vivo is still in the exploratory stage. This study aimed to use magnetic resonance imaging （MRI） and diffusion tensor imaging （DTI） to evaluate vastus medialis oblique （VMO） fiber bundles in patients with recurrent patellar dislocation to explore the changes in muscle morphology and function. Methods： This prospective study involved 30 patients （7 males and 23 females; average age, 21.4 ± 3.8 years） clinically diagnosed with recurrent patellar dislocation in Peking University Third Hospital and 30 healthy volunteers matched for age, sex, and body mass index in our medical school between January 2014 and October 2014. None of the patients had a recent history of traumatic patellar dislocation or transient patellar dislocation. All patients underwent conventional MRI and DTI of the knee. The cross-sectional area of the VMO on MRI and the fractional anisotropy （FA）, apparent diffusion coefficient （ADC）, and primary （λ1）, secondary （λ2）, and three-level characteristic （λ3） values on DTI were measured. The independent-samples t-test was used to compare these parameters between the two groups. Results： Compared with the control group, the patient group showed significantly higher FA values （0.39 ± 0.05 vs. 0.33 ± 0.03） and significantly lower ADC （1.51 ± 0.13 vs. 1.58± 0.07）, λ2 （4.96 ±0.13 vs. 5.04 ± 0.07）, and λ3 values （4.44 ± 0.14 vs. 4.58 ± 0.07; t = 5.99, t = -2.58, t = 3.02, and t = -4.88, respectively： all P 〈 0.05）. Cross-sectional VMO area and λ1 values did not differ between the two groups （t = 1.82 and t = 0.22, respectively; both P 〉 0.05）. Conclusions： The functional status of the VMO is closely associated with recurrent patellar dislocation. MRI, especially DTI （FA, ADC, λ2, and ,λ3）, can detect early changes in VMO function and might lhcilitate the noninvasive monitoring of the functional status of the VMO in patients with recurre

High-performance si/VO_(2)-nanorod heterojunction photodetector based on photothermoelectric effect for detecting human radiation

As a typical candidate of optoelectronic materials,vanadium dioxide(VO_(2))has wide applications in photodetectors(PDs),but is still challenging in largely enhancing the photodetecting performance for low-power human radiation.Herein,high-performance si/VO_(2)nanorods(NRs)heterojunction PDs based on the photothermoelectric(PTE)effect are presented.The uniform VO_(2)-NRs array films were deposited on si by using magnetron sputtering technique,and a si/VO_(2)heterojunctions were fabricated.The device exhibits a four-stage photoresponse and broadband photoresponse from ultraviolet to long-wavelength infrared.Benefited from the unique nanorods structure and the strong PTE effect,the fabricated device exhibits a large enhancement of the photodetecting performance,showing an ultrahigh photodetectivity of 1.6×10^(13)Jones and ultrafast response rates with a risingedge time of~65.0μs,three orders of magnitude higher than other VO_(2)-based devices.Furthermore,the device exhibits unique abilities to detect human radiation even when the human fingers are far away from the device surface up to 10.0 cm.Additionally,the fabricated si/VO_(2)devices can also be applied as breath sensors to distinguish different breathing patterns.These results supply an effective route to design high-performance photodetectors toward detecting human thermal radiation and respiration.
