Valvular heart disease(VHD),clinically manifested as stenosis and regurgitation of native heart valve,is one of the most prevalent cardiovascular diseases with high mortality.Heart valve replacement surgery has been recognized as golden standard for the treatment of VHD.Owing to the clinical application of transcatheter heart valve replacement technic and the excellent hemodynamic performance of bioprosthetic heart valves(BHVs),implantation of BHVs has been increasing over recent years and gradually became the preferred choice for the treatment of VHD.However,BHVs might fail within 10-15 years due to structural valvular degeneration(SVD),which was greatly associated with drawbacks of glutaraldehyde crosslinked BHVs,including cytotoxicity,calcification,component degradation,mechanical failure,thrombosis and immune response.To prolong the service life of BHVs,much effort has been devoted to overcoming the drawbacks of BHVs and reducing the risk of SVD.In this review,we summarized and analyzed the research and progress on:(i)modification strategies based on glutaraldehyde crosslinked BHVs and(ii)nonglutaraldehyde crosslinking strategies for BHVs.

High frequency pulsating voltage injection method is a good candidate for detecting the initial rotor position of permanent magnet synchronous motor.However,traditional methods require a large number of filters,which leads to the deterioration of system stability and dynamic performance.In order to solve these problems,a new signal demodulation method is proposed in this paper.The proposed new method can directly obtain the amplitude of high-frequency current,thus eliminating the use of filters,improving system stability and dynamic performance and saving the work of adjusting filter parameters.In addition,a new magnetic polarity detection method is proposed,which is robust to current measurement noise.Finally,experiments verify the effectiveness of the method.

Background:The conversion of adenosine(A)to inosine(I)through deamination is the prevailing form of RNA editing,impacting numerous nuclear and cytoplasmic transcripts across various eukaryotic species.Millions of high-confidence RNA editing sites have been identified and integrated into various RNA databases,providing a convenient platform for the rapid identification of key drivers of cancer and potential therapeutic targets.However,the available database for integration of RNA editing in hematopoietic cells and hematopoietic malignancies is still lacking.Methods:We downloaded RNA sequencing(RNA-seq)data of 29 leukemia patients and 19 healthy donors from National Center for Biotechnology Information(NCBI)Gene Expression Omnibus(GEO)database,and RNA-seq data of 12 mouse hematopoietic cell populations obtained from our previous research were also used.We performed sequence alignment,identified RNA editing sites,and obtained characteristic editing sites related to normal hematopoietic development and abnormal editing sites associated with hematologic diseases.Results:We established a new database,"REDH",represents RNA editome in hematopoietic differentiation and malignancy.REDH is a curated database of associations between RNA editome and hematopoiesis.REDH integrates 30,796 editing sites from 12 murine adult hematopoietic cell populations and systematically characterizes more than 400,000 edited events in malignant hematopoietic samples from 48 cohorts(human).Through the Differentiation,Disease,Enrichment,and knowledge modules,each A-to-I editing site is systematically integrated,including its distribution throughout the genome,its clinical information(human sample),and functional editing sites under physiological and pathological conditions.Furthermore,REDH compares the similarities and differences of editing sites between different hematologic malignancies and healthy control.Conclusions:REDH is accessible at http://***/.This user-friendly database would aid in understanding the mechanisms of RNA editing in hema

Parameterization is a critical step in modelling ecosystem dynamics.However,assigning parameter values can be a technical challenge for structurally complex natural plant communities;uncertainties in model simulations often arise from inappropriate model parameterization.Here we compared five methods for defining community-level specific leaf area(SLA)and leaf C:N across nine contrasting forest sites along the North-South Transect of Eastern China,including biomass-weighted average for the entire plant community(AP_BW)and four simplified selective sampling(biomass-weighted average over five dominant tree species[5DT_BW],basal area weighted average over five dominant tree species[5DT_AW],biomass-weighted average over all tree species[AT_BW]and basal area weighted average over all tree species[AT_AW]).We found that the default values for SLA and leaf C:N embedded in the Biome-BGC v4.2 were higher than the five computational methods produced across the nine sites,with deviations ranging from 28.0 to 73.3%.In addition,there were only slight deviations(simplified selective sampling methods,and no significant difference between the predictions of AT_BW and AP_BW except the Shennongjia site.The findings in this study highlights the critical importance of computational strategies for community-level parameterization in ecosystem process modelling,and will support the choice of parameterization methods.

Intestinal failure-associated liver disease(IFALD)is a common hepatobiliary complication resulting from long-term parenteral nutrition(PN)in patients with intestinal failure.The spectrum of IFALD ranges from cholestasis,steatosis,portal fibrosis,to cirrhosis.Development of IFALD is a multifactorial process,in which gut dysbiosis plays a critical role in its initiation and progression in conjunction with increased intestinal permeability,activation of hepatic immune responses,and administration of lipid emulsion.Gut microbiota manipulation including pre/probiotics,fecal microbiota transplantation,and antibiotics has been studied in IFALD with varying success.In this review,we summarize current knowledge on the taxonomic and functional changes of gut microbiota in preclinical and clinical studies of IFALD.We also review the function of microbial metabolites and associated signalings in the context of IFALD.By providing microbiota-targeted interventions aiming to optimize PN-induced liver injury,our review provides perspectives for future basic and translational investigations in the field.

Deflagration and detonation induced by shock wave focusing at different Mach numbers

Shock wave focusing is an effective way to create a hot spot or a high-pressure and hightemperature region at a certain place,showing its unique usage in detonation initiation,which is beneficial for the development of detonation-based engines.The flame propagation behavior after the autoignition induced by shock wave focusing is crucial to the formation and self-sustaining of the detonation wave.In this study,wedge reflectors with two different angles(60°and 90°)and a planar reflector are employed,and the Mach number of incident shock waves ranging from 2.0 to 2.8 is utilized to trigger different flame propagation modes.Dynamic pressure transducers and the high-speed schlieren imaging system are both employed to investigate the shock-shock collision and ignition procedure.The results reveal a total of four flame propagation modes:deflagration,DDT(Deflagration-to-Detonation Transition),unsteady detonation,and direct detonation.The detonation wave formed in the DDT and unsteady detonation mode is only approximately 75%-85%of the Chapman-Jouguet(C-J)speed;meanwhile,the directly induced detonation wave speed is close to the C-J speed.Transverse waves,which are strong evidence for the existence of detonation waves,are discovered in experiments.The usage of wedge reflectors significantly reduces the initial pressure difference ratio needed for direct detonation ignition.We also provide a practical method for differentiating between detonation and deflagration modes,which involves contrasting the speed of the reflected shock wave with the speed of the theoretically nonreactive reflected shock wave.These findings should serve as a reference for the detonation initiation technique in advanced detonation propulsion engines.

Vertebrate neurons are highly dynamic cells that undergo several alterations in their functioning and physiologies in adaptation to various external stimuli.In particular,how these neurons respond to physical exercise has long been an area of active research.Studies of the vertebrate locomotor system’s adaptability suggest multiple mechanisms are involved in the regulation of neuronal activity and properties during exercise.In this brief review,we highlight recent results and insights from the field with a focus on the following mechanisms:(a)alterations in neuronal excitability during acute exercise;(b)alterations in neuronal excitability after chronic exercise;(c)exercise-induced changes in neuronal membrane properties via modulation of ion channel activity;(d)exercise-enhanced dendritic plasticity;and(e)exercise-induced alterations in neuronal gene expression and protein synthesis.Our hope is to update the community with a cellular and molecular understanding of the recent mechanisms underlying the adaptability of the vertebrate locomotor system in response to both acute and chronic physical exercise.

Separable nonlinear models are widely used in various fields such as time series analysis, system modeling, and machine learning, due to their flexible structures and ability to capture nonlinear behavior of data. However, identifying the parameters of these models is challenging, especially when sparse models with better interpretability are desired by practitioners. Previous theoretical and practical studies have shown that variable projection (VP) is an efficient method for identifying separable nonlinear models, but these are based on \(L_2\) penalty of model parameters, which cannot be directly extended to deal with sparse constraint. Based on the exploration of the structural characteristics of separable models, this paper proposes gradient-based and trust-region-based variable projection algorithms, which mainly solve two key problems: how to eliminate linear parameters under sparse constraint;and how to deal with the coupling relationship between linear and nonlinear parameters in the model. Finally, numerical experiments on synthetic data and real time series data are conducted to verify the effectiveness of the proposed algorithms.

Atherosclerosis(AS)is characterized by impairment and apoptosis of endothelial cells,continuous systemic and focal inflammation and dysfunction of vascular smooth muscle cells,which is documented as the traditional cellular paradigm.However,the mechanisms appear much more complicated than we thought since a bulk of studies on efferocytosis,transdifferentiation and novel cell death forms such as ferroptosis,pyroptosis,and extracellular trap were reported.Discovery of novel pathological cellular landscapes provides a large number of therapeutic targets.On the other side,the unsatisfactory therapeutic effects of current treatment with lipid-lowering drugs as the cornerstone also restricts the efforts to reduce global AS burden.Stem cell-or nanoparticle-based strategies spurred a lot of attention due to the attractive therapeutic effects and minimized adverse effects.Given the complexity of pathological changes of AS,attempts to develop an almighty medicine based on single mechanisms could be theoretically challenging.In this review,the top stories in the cellular landscapes during the initiation and progression of AS and the therapies were summarized in an integrated perspective to facilitate efforts to develop a multi-targets strategy and fill the gap between mechanism research and clinical translation.The future challenges and improvements were also discussed.
