Multi-objective optimal control of tungiasis diseases with terminal demands

In this paper,we aim to minimize the epidemic size of tungiasis disease and economic costs simultaneously,with terminal demands for infected humans.A human-jigger parasite control system with four control schemes for humans and jiggers is *** propose a multi-objective optimal control problem with terminal constraints,in which the accumulated number of infected humans and control costs are *** applying the modified normal boundary intersection algorithm and the interior point scheme,numerical simulations for different combinations of control schemes are carried out,and actual data in Madagascar are *** combination schemes are indicated from the perspectives of disease eradication,cost saving and time *** these effective combinations are properly performed,the disease can be *** only minimizing the epidemic size,the combination of the optimal treatments and adulticiding efforts is the best choice in the rainy season;the combination of the optimal personal protections and treatments is the preferential option in the dry *** only minimizing the economical cost,the combination of the optimal adulticide and larvicide is the better selection in the rainy season;the combination of the optimal personal protections,treatments and adulticiding efforts is the prior choose in the dry ***,there is a trade-off between the two objectives for all the effective combinations,decision-makers may choose an appropriate one to control the disease.

Determination method of mesh size for numerical simulation of blast load in near-ground detonation

In order to improve the overall resilience of the urban infrastructures, it is required to conduct blast resistant design for important building structures in the city. For complex terrain in the city, it is recommended to determine the blast load on the structures via numerical simulation. since the mesh size of the numerical model highly depends on the explosion scenario, there is no generally applicable approach for the mesh size selection. An efficient method to determine the mesh size of the numerical model of near-ground detonation based on explosion scenarios is proposed in this study. The effect of mesh size on the propagation of blast wave under different explosive weights was studied, and the correlations between the mesh size effect and the charge weight or the scaled distance was described. Based on the principle of the finite element method and Hopkinson-Cranz scaling law, a mesh size measurement unit related to the explosive weight was proposed as the criterion for determining the mesh size in the numerical simulation. Finally, the applicability of the method proposed in this paper was verified by comparing the results from numerical simulation and the explosion tests and was verified in AUTODYN.

Snap-through behaviors and nonlinear vibrations of a bistable composite laminated cantilever shell:an experimental and numerical study

The snap-through behaviors and nonlinear vibrations are investigated for a bistable composite laminated cantilever shell subjected to transversal foundation excitation based on experimental and theoretical *** improved experimental specimen is designed in order to satisfy the cantilever support boundary condition,which is composed of an asymmetric region and a symmetric *** symmetric region of the experimental specimen is entirely clamped,which is rigidly connected to an electromagnetic shaker,while the asymmetric region remains free of *** motion paths are realized for the bistable cantilever shell by changing the input signal levels of the electromagnetic shaker,and the displacement responses of the shell are collected by the laser displacement *** numerical simulation is conducted based on the established theoretical model of the bistable composite laminated cantilever shell,and an off-axis three-dimensional dynamic snap-through domain is *** numerical solutions are in good agreement with the experimental *** nonlinear stiffness characteristics,dynamic snap-through domain,and chaos and bifurcation behaviors of the shell are quantitatively *** to the asymmetry of the boundary condition and the shell,the upper stable-state of the shell exhibits an obvious soft spring stiffness characteristic,and the lower stable-state shows a linear stiffness characteristic of the shell.

NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Objective Hepatocellular carcinoma(HCC)is the third leading cause of cancer-associated death *** a first-line drug for advanced HCC treatment,lenvatinib faces a significant hurdle due to the development of both intrinsic and acquired resistance among patients,and the underlying mechanism remains largely *** present study aims to identify the pivotal gene responsible for lenvatinib resistance in HCC,explore the potential molecular mechanism,and propose combinatorial therapeutic targets for HCC *** Cell viability and colony formation assays were conducted to evaluate the sensitivity of cells to lenvatinib and ***-Seq was used to determine the differences in transcriptome between parental cells and lenvatinib-resistant(LR)*** upregulated genes were analyzed by GO and KEGG ***,qPCR and Western blotting were employed to determine the relative gene expression ***,the intracellular reactive oxygen species(ROS)and apoptosis were detected by flow *** PLC-LR and Hep3B-LR were *** was a total of 116 significantly upregulated genes common to both LR cell *** GO and KEGG analyses indicated that these genes were involved in oxidoreductase and dehydrogenase activities,and reactive oxygen species ***,NAD(P)H:quinone oxidoreductase 1(NQO1)was highly expressed in LR cells,and was involved in the lenvatinib *** high expression of NQO1 decreased the production of ROS induced by lenvatinib,and subsequently suppressed the *** combination of lenvatinib and NQO1 inhibitor,dicoumarol,reversed the resistance of LR *** The high NQO1 expression in HCC cells impedes the lenvatinib-induced apoptosis by regulating the ROS levels,thereby promoting lenvatinib resistance in HCC cells.

Development of RPA-Cas12a-fluorescence assay for rapid and reliable detection of human bocavirus 1

Human bocavirus(HBoV)1 is considered an important pathogen that mainly affects infants aged 6–24 months,but preventing viral transmission in resource-limited regions through rapid and affordable on-site diagnosis of individuals with early infection of HBoV1 remains somewhat ***,we present a novel faster,lower cost,reliable method for the detection of HBoV1,which integrates a recombinase polymerase amplification(RPA)assay with the CRISPR/Cas12a system,designated the RPA-Cas12a-fluorescence *** RPA-Cas12a-fluorescence system can specifically detect target gene levels as low as 0.5 copies of HBoV1 plasmid DNA per microliter within 40 min at 37℃without the need for sophisticated *** method also demonstrates excellent specificity without cross-reactivity to non-target ***,the method was appraised using 28 clinical samples,and displayed high accuracy with positive and negative predictive agreement of 90.9%and 100%,***,our proposed rapid and sensitive HBoV1 detection method,the RPA-Cas12a-fluorescence assay,shows promising potential for early on-site diagnosis of HBoV1 infection in the fields of public health and health *** established RPA-Cas12a-fluorescence assay is rapid and reliable method for human bocavirus 1 *** RPA-Cas12a-fluorescence assay can be completed within 40 min with robust specificity and sensitivity of 0.5 copies/μl.

Genetic Analysis of Two Novel GPI Variants Disrupting H Bonds and Localization Characteristics of 55 Gene Variants Associated with Glucose-6-phosphate Isomerase Deficiency

Objective:Glucose-6-phosphate isomerase(GPI)deficiency is a rare hereditary nonspherocytic hemolytic anemia caused by GPI gene *** disorder exhibits wide heterogeneity in its clinical manifestations and molecular characteristics,often posing challenges for precise diagnoses using conventional *** this end,this study aimed to identify the novel variants responsible for GPI deficiency in a Chinese ***:The clinical manifestations of the patient were summarized and analyzed for GPI deficiency phenotype *** compound heterozygous variants of the GPI gene,c.174C>A(***58Lys)and c.1538G>T(***513Leu),were identified using whole-exome and Sanger *** AlphaFold program and Chimera software were used to analyze the effects of compound heterozygous variants on GPI ***:By characterizing 53 GPI missense/nonsense variants from previous literature and two novel missense variants identified in this study,we found that most variants were located in exons 3,4,12,and 18,with a few localized in exons 8,9,and *** study identified novel compound heterozygous variants associated with GPI *** pathogenic variants disrupt hydrogen bonds formed by highly conserved GPI amino ***:Early family-based sequencing analyses,especially for patients with congenital anemia,can help increase diagnostic accuracy for GPI deficiency,improve child healthcare,and enable genetic counseling.

Changes in extreme high temperature warning indicators over China under different global warming levels

High temperature warning indicators play a pivotal role in meteorological departments,serving as crucial criteria for issuing warnings that guide both social production and daily *** their importance,limited studies have explored the relationship between different global warming levels and changes in high temperature warning *** this study,we analyze data from 2,419 meteorological stations over China and utilize the Coupled Model Intercomparison Project Phase 6(CMIP6)models to examine historical changes in high temperature warning indicators used by the China Meteorological *** evaluate model performance and estimate future changes in these indicators using an annual cycle bias correction *** results indicate that since 1961,the number of high temperature days(TX35d and TX40d)and length of season(TX40d and TX40l)with daily maximum temperature reaching or exceeding 35℃ and 40℃ have increased over *** intensity of high temperatures(TXx)has strengthened and the geographical extent affected by high temperatures has *** 2022,the occurrence of 40℃ high temperatures surges,with Eastern China experiencing a two-day increase in TX40d and an extended seasonal length in TX40l by over five *** CMIP6 models have underestimated the high temperature indictors associated with 35℃ during historical periods,notable difference is not observed between the models and observations for TX40d and TX40l,given their rare ***,future projections,after bias correction,indicate that the increasing trends for 35℃ and 40℃ high temperature days and length of season become more pronounced than the raw projection,suggesting a more severe increase than that anticipated *** global warming intensifies,the high temperature days and length of season are projected to increase non-linearly,while the intensity of high temperatures is expected to increase *** every 1℃ increase in global temperature,the intensity is
