BL03HB: a Laue microdiffraction beamline for both protein crystallography and materials science at SSRF

A Laue microdiffraction beamline(BL03HB) was constructed at the Shanghai Synchrotron Radiation Facility(SSRF).This beamline features two consecutive focusing points in two different sectors within its end station, the first dedicated to protein crystallography and the other tailored to materials science applications. Based on a superbend dipole magnet with a magnetic field of 2.29 T, a two-stage focusing design was implemented with two sets of Kirkpatrick-Baez mirrors to achieve a micro white beam as small as 4.2 μ m ×4.3 μ m at the first sector and 0.9 μ m ×1.3 μ m at the second sector in the standard beamline operation mode at SSRF. The X-ray microbeam in the two sectors can be easily switched between monochromatic and white beams by moving a four-bounce monochromator in or out of the light path, respectively. In the protein crystallography sector, white-beam Laue microdiffraction was demonstrated to successfully determine the structure of protein crystals from only a few images of diffraction data collected by a Pilatus 2 M area detector. In the materials science sector,the white-beam Laue diffraction was collected in a reflection geometry using another Pilatus 2 M area detector, which could map the microstructural distribution on the sample surface by scanning the samples. In general, the BL03HB beamline promotes the application of Laue microdiffraction in both protein crystallography and materials science. This paper presents a comprehensive overview of the BL03HB beamline, end station, and the first commission results.

The protein complex crystallographic beamline BL19U1 at the Shanghai Synchrotron Radiation Facility is one of the five beamlines dedicated to protein sciences operated by National Facility for Protein Science(Shanghai,China).The beamline,which features a small-gap invacuum undulator,has been officially open to users since March *** beamline delivers X-ray in the energy range 7–15 *** its high flux,low divergence beam and a large active area detector,BL19U1 is designed for proteins with large molecular weight and large crystallographic unit cell *** performance and stable operation of the beamline have allowed the number of Protein Data Bank(PDB)depositions and the number of articles published based on data collected at this beamline to increase *** date,over 300 research groups have collected data at the *** than 600 PDB entries have been deposited at the PDB(***).More than 300 papers have been published that include data collected at the beamline,including 21 research articles published in the top-level journals Cell,Nature,and Science.

Combined biochar and wheat-derived endophytic bacteria reduces cadmium uptake in wheat grains in a metal-polluted soil

In this study,two wheat-derived cadmium(Cd)-immobilizing endophytic Pseudomonas paralactis M14 and Priestia megaterium R27 were evaluated for their effects on wheat tissue Cd uptake under hydroponic ***,the impacts of the biochar(BC),M14+R27(MR),and BC+MR treatments on wheat Cd uptake and the mechanisms involved were investigated at the jointing,heading,and mature stages of wheat plants under field-plot conditions.A hydroponic experiment showed that the MR treatment significantly decreased the above-ground tissue Cd content compared with theM14 or R27 *** BC+MRtreatment reduced the grain Cd content by 51.5%-67.7%and Cd translocation factor at the mature stage of wheat plants and increased the organic matter-bound Cd content by 31%-75%in the rhizosphere soils compared with the BC or MR *** with the BC or MR treatment,the relative abundances of the biomarkers associated with Gemmatimonas,Altererythrobacter,Gammaproteobacteria,Xanthomonadaceae,Phenylobacterium,and Nocardioides in the BC+MR-treated rhizosphere microbiome decreased and negatively correlated with the organic matter-bound Cd *** the BC+MR-treated root interior microbiome,the relative abundance of the biomarker belonging to Exiguobacterium increased and negatively correlated with the Cd translocation factor,while the relative abundance of the biomarker belonging to Pseudonocardiaceae decreased and positively correlated with the Cd translocation *** findings suggested that the BC+MR treatment reduced Cd availability and Cd transfer through affecting the abundances of these specific biomarkers in the rhizosphere soil and root interior microbiomes,leading to decreased wheat grain Cd uptake in the contaminated soil.

Female-specific vs conventional knee prostheses after total knee arthroplasty:A meta-analysis

AIM： To study whether female-specifc prostheses are superior to conventional prostheses after total knee arthroplasty （TKA） by conducting this meta-analysis to evaluate the effects of the 2 different ***： A systematic electronic search was con-ducted in the databases of PubMed, EMBASE and the Cochrane Library for prospective and retrospective trials. Meta-analysis was performed for the outcomes, includ-ing range of motion （ROM）, Knee Society score, Hospi-tal for Special Surgery scores （HSS） and complications including deep infection, manipulation under anesthesia （MUA）, revisions, anterior knee pain, deep vein throm-bosis and overhang rate. Meta-analysis was conductedwhere applicable. Weighted mean difference （WMD） and odds ratio were calculated according to study ***： Seven studies including 1174 knees wereeligible for data extraction and pooled analysis. The overhang rate of female-specifc prostheses was signif-cantly lower than the conventional ones （WMD, 3.25; 95%CI： 0.00-0.27; P = 0.001）. ROM in the female-spe-cifc prostheses group after TKA tended to be greater than the conventional prostheses group; however, with insignifcant difference （WMD, 2.48; 95%CI： -0.83-5.78; P = 0.14）. HSS （WMD, 0.48; 95%CI： -1.45-0.88; P = 0.63） and complications including deep infection （WMD, 0.39; 95%CI： 0.19-3.08; P =0 .70）, MUA （WMD, 1.53; 95%CI： 0.02-1.61; P = 0.13） and revisions （WMD, 0.55; 95%CI： 0.07-4.34; P = 0.13） were all comparable be-tween the two groups with at least 1 year follow-up. Other indexes were revealed to be similar between the two treatments with a descriptive analytical ***： Although the overhang rate is lower with female-specific prostheses, the current evidence does not support that female-specific prostheses out-weigh conventional ones after TKA.

Least Square Estimation for Multiple Functional Linear Model with Autoregressive Errors

As an extension of linear regression in functional data analysis, functional linear regression has been studied by many researchers and applied in various fields. However, in many cases, data is collected sequentially over time, for example the financial series, so it is necessary to consider the autocorrelated structure of errors in functional regression background. To this end, this paper considers a multiple functional linear model with autoregressive errors. Based on the functional principal component analysis, we apply the least square procedure to estimate the functional coefficients and autoregression coefficients. Under some regular conditions, we establish the asymptotic properties of the proposed estimators. A simulation study is conducted to investigate the finite sample performance of our estimators. A real example on China's weather data is applied to illustrate the validity of our model.

Critical station identification of metro networks based on the integrated topological-functional algorithm:A case study of Chengdu

As a key mode of transportation, urban metro networks have significantly enhanced urban traffic environments and travel efficiency, making the identification of critical stations within these networks increasingly essential. This study presents a novel integrated topological-functional(ITF) algorithm for identifying critical nodes, combining topological metrics such as K-shell decomposition, node information entropy, and neighbor overlapping interaction with the functional attributes of passenger flow operations, while also considering the coupling effects between metro and bus networks. Using the Chengdu metro network as a case study, the effectiveness of the algorithm under different conditions is *** results indicate significant differences in passenger flow patterns between working and non-working days, leading to varying sets of critical nodes across these scenarios. Moreover, the ITF algorithm demonstrates a marked improvement in the accuracy of critical node identification compared to existing methods. This conclusion is supported by the analysis of changes in the overall network structure and relative global operational efficiency following targeted attacks on the identified critical nodes. The findings provide valuable insight into urban transportation planning, offering theoretical and practical guidance for improving metro network safety and resilience.

Factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the patients

BACKGROUND Despite laparoscopic total hysterectomy provided more benefits to patients and has become one of the most commonly used surgical methods for total ***,vaginal cuff dehiscence is a serious complication after laparoscopic total hysterectomy and so far,there are limited studies on vaginal cuff dehiscence after a total *** To investigate the factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the *** This study retrospectively,univariately,and multivariately analyzed the clinical data of patients who experienced vaginal cuff dehiscence after laparoscopic hysterectomy in Yuyao People’s Hospital from January 2015 to December *** regression was utilized to analyze the high-risk factors of vaginal cuff *** Among 1459 cases with laparoscopic hysterectomy,9 reported vaginal cuff dehiscence postoperatively,with an incidence of 0.617%.Univariate logistic regression analysis revealed that preoperative hemoglobin value[odds ratio(OR)=5.12,P=0.016],vaginal cuff suture method(OR=0.26,P=0.048),and postoperative first sexual lifetime(OR=15.86,P=0.002)were associated with vaginal cuff dehiscence following laparoscopic *** logistic regression analysis revealed that preoperative hemoglobin value of<90 g/L(OR=5.17,P=0.015)and time interval between postoperative first sexual life and operation of<3 months(OR=54.00,P=0.004)are independent risk factors for vaginal cuff dehiscence after laparoscopic *** Postoperative hemoglobin and the time interval between postoperative first sexual life and operation were the independent factors of developing vaginal cuff dehiscence after laparoscopic hysterectomy.
