Objective Hepatocellular carcinoma(HCC)is the third leading cause of cancer-associated death worldwide.As a first-line drug for advanced HCC treatment,lenvatinib faces a significant hurdle due to the development of both intrinsic and acquired resistance among patients,and the underlying mechanism remains largely unknown.The present study aims to identify the pivotal gene responsible for lenvatinib resistance in HCC,explore the potential molecular mechanism,and propose combinatorial therapeutic targets for HCC management.Methods Cell viability and colony formation assays were conducted to evaluate the sensitivity of cells to lenvatinib and dicoumarol.RNA-Seq was used to determine the differences in transcriptome between parental cells and lenvatinib-resistant(LR)cells.The upregulated genes were analyzed by GO and KEGG analyses.Then,qPCR and Western blotting were employed to determine the relative gene expression levels.Afterwards,the intracellular reactive oxygen species(ROS)and apoptosis were detected by flow cytometry.Results PLC-LR and Hep3B-LR were established.There was a total of 116 significantly upregulated genes common to both LR cell lines.The GO and KEGG analyses indicated that these genes were involved in oxidoreductase and dehydrogenase activities,and reactive oxygen species pathways.Notably,NAD(P)H:quinone oxidoreductase 1(NQO1)was highly expressed in LR cells,and was involved in the lenvatinib resistance.The high expression of NQO1 decreased the production of ROS induced by lenvatinib,and subsequently suppressed the apoptosis.The combination of lenvatinib and NQO1 inhibitor,dicoumarol,reversed the resistance of LR cells.Conclusion The high NQO1 expression in HCC cells impedes the lenvatinib-induced apoptosis by regulating the ROS levels,thereby promoting lenvatinib resistance in HCC cells.

As a typical candidate of optoelectronic materials,vanadium dioxide(VO_(2))has wide applications in photodetectors(PDs),but is still challenging in largely enhancing the photodetecting performance for low-power human radiation.Herein,high-performance si/VO_(2)nanorods(NRs)heterojunction PDs based on the photothermoelectric(PTE)effect are presented.The uniform VO_(2)-NRs array films were deposited on si by using magnetron sputtering technique,and a si/VO_(2)heterojunctions were fabricated.The device exhibits a four-stage photoresponse and broadband photoresponse from ultraviolet to long-wavelength infrared.Benefited from the unique nanorods structure and the strong PTE effect,the fabricated device exhibits a large enhancement of the photodetecting performance,showing an ultrahigh photodetectivity of 1.6×10^(13)Jones and ultrafast response rates with a risingedge time of~65.0μs,three orders of magnitude higher than other VO_(2)-based devices.Furthermore,the device exhibits unique abilities to detect human radiation even when the human fingers are far away from the device surface up to 10.0 cm.Additionally,the fabricated si/VO_(2)devices can also be applied as breath sensors to distinguish different breathing patterns.These results supply an effective route to design high-performance photodetectors toward detecting human thermal radiation and respiration.

BACKGROUND Preoperative serum tumor markers not only play a role in the auxiliary diagnosis and postoperative monitoring in colorectal cancer(CRC),but also have been found to have potential prognostic value.AIM To analyze whether preoperative serum tumor markers,including carcinoembryonic antigen(CEA)and carbohydrate antigen 19-9(CA19-9),affect the prognosis of CRC.METHODS This was a retrospective study conducted in a single center.Patients with nonmetastatic CRC who underwent initial surgery between January 2011 and January 2020 were enrolled and divided into development site and validation site groups at a ratio of 7:3.The independent prognostic factors were screened by Cox regression analysis,and finally,a prognostic nomogram model was established.The newly developed model was tested by internal validation.RESULTS Eventually,3526 postoperative patients with nonmetastatic CRC were included in the study.There were 2473 patients at the development site and 1056 patients at the validation site.Age(Psis(TNM)classification(Psidered independent prognostic factors for patients with nonmetastatic CRC and were used as variables in the nomogram model.The areas under the curve of the development and validation sites were 0.655 and 0.658,respectively.The calibration plot also showed the significant performance of the newly established nomogram.CONCLUsiON We successfully constructed a nomogram model based on age,TNM stage,preoperative CEA,and CA19-9 levels to evaluate the overall survival of patients with nonmetastatic CRC.

CaCu_(3)Ti_(4)O_(12)(CCTO)ceramic nanocomposites incorporating graphene–carbon black(GRCB)fillers were fabricated by spark plasma sintering process.The percolative effects of conductive GRCB fillers on dielectric response of GRCB/CCTO ternary metacomposites were systematically investigated.The weakly real permittivity(ε′)-negative response(ε′~−1×10^(2))was achieved which originated from weakly low-frequency plasmonic state of free carriers within constructed GRCB networks.With enhancing three-dimensional GRCB network,the plasma frequency of metacomposites increased while the damping factor decreased.Herein,theε′-negative values of metacomposites were tuned from−10^(2) to−10^(4) orders of magnitude andε′-near-zero(ENZ)frequencies from~142 to~340 MHz which substantially benefited from the moderate carrier concentration of GRCB dual fillers.The Drude model and equivalent circuit models were adopted to demonstrate dielectric and electrical characteristics.The obtained metacomposites show strong EM shielding effect along with enhanced plasmonic oscillation and even better achieving perfect EM shielding effect in ENZ media.This work achieves the tunableε′-negative andε′-near-zero response and more importantly clarifies its regulation mechanism in ceramic-based ternary metacomposites,which opens up the possibility of designing high-performance EM shielding materials based on metacomposites.

Tensegrities made of tensile strings and compressed struts possess large strength-to-mass ratio values but tend to experience non-negligible vibration in dynamic environments because of poor structural damping.Here,we introduce particle dampers into a tensegrity prism to attenuate vibration,with the goal of establishing a lightweight and efficient approach of vibration suppression.To integrate the particle dampers and the tensegrity,a novel strut structure formed by assembling a solid strut and a hollow strut is devised,in which granular materials are inserted to develop a particle damper.The vibration attenuation performance of the tensegrity prism is investigated through exciter tests.According to the experimental parametric study,the influences of system parameters including excitation magnitude,the filling height of particles,particle size and the configuration of tensegrity prism on the vibration attenuation performance is analyzed.In addition,the experimental results regarding the dependence of the vibration attenuation on the system parameters are interpreted by the mechanism of collisions and frictions between particles and between particles and struts.The maximal vibration attenuation ratio of 76%can be achieved in the experiments.Thus,this research can provide insights into the design of lightweight tensegrity structures where vibration suppression is important,particularly in some dynamic environments.

High-frequency oscillation(HFO) of gridconnected wind power generation systems(WPGS) is one of the most critical issues in recent years that threaten the safe access of WPGS to the grid. Ensuring the WPGS can damp HFO is becoming more and more vital for the development of wind power. The HFO phenomenon of wind turbines under different scenarios usually has different mechanisms. Hence, engineers need to acquire the working mechanisms of the different HFO damping technologies and select the appropriate one to ensure the effective implementation of oscillation damping in practical engineering. This paper introduces the general assumptions of WPGS when analyzing HFO, systematically summarizes the reasons for the occurrence of HFO in different scenarios, deeply analyses the key points and difficulties of HFO damping under different scenarios, and then compares the technical performances of various types of HFO suppression methods to provide adequate references for engineers in the application of technology. Finally, this paper discusses possible future research difficulties in the problem of HFO, as well as the possible future trends in the demand for HFO damping.

Soft electrochemical actuators that can generate mechanical motions in response to electrical stimuli can be used as artificial muscles in microscale robots.The electrode materials are the key component of the electrochemical actuator because they limit the ion diffusion and accumulation processes and thus affect the deformation in fast response-time and potential engineering applications.Low-dimensional materials,such as one-dimensional(1D)CNT and two-dimensional(2D)vertically stood WS_(2),are great electrode due to their good electrochemical stability and high surface-to-volume ratios,but show limited performances when being used on their own in the actuators because of low reconstruction or low conductivity.A combination of 1D CNT and 2D WS_(2)could form a heterojunction with both high electrochemical activity and great mechanical stability.Herein,by depositing highly disordered graphene nanosheets(HDGNs)on CNT films and then WS_(2)on the formed low-dimensional nanocarbon film,a hybrid WS_(2)/HDGN/CNT film was synthesized through a multistep CVD method.Vertically grown WS_(2)nanosheets were achieved by taking advantage of HDGNs which limits CNTs sliding.So,the volume expansion caused by the transition from WO_(3)to WS-2can force the vertical growth of WS_(2).This hybrid film shows excellent electrochemical properties,such as high specific capacitance and high rate performance.Soft actuators based on the WS_(2)/HDGN/CNT films show good actuating performance,including stability and reconfigurability.

Objective This study aimed to clarify the intervention effect of salidroside(SAL)on lung injury caused by PM_(2.5) in mice and illuminate the function of siRT1-PGC-1ɑaxis.Methods Specific pathogen-free(SPF)grade male C57BL/6 mice were randomly assigned to the following groups:control group,SAL group,PM_(2.5) group,SAL+PM_(2.5) group.On the first day,SAL was given by gavage,and on the second day,PM_(2.5) suspension was given by intratracheal instillation.The whole experiment consist of a total of 10 cycles,lasting 20 days.At the end of treatment,blood samples and lung tissues were collected and analyzed.Observation of pathological changes in lung tissue using inverted microscopy and transmission electron microscopy.The expression of inflammatory,antioxidants,apoptosis,and siRT1-PGC-1ɑproteins were detected by Western blotting.Results Exposure to PM_(2.5) leads to obvious morphological and pathologica changes in the lung of mice.PM_(2.5) caused a decline in levels of antioxidant-related enzymes and protein expressions of HO-1,Nrf2,SOD2,siRT1 and PGC-1ɑ,and an increase in the protein expressions of IL-6,IL-1β,Bax,caspase-9 and cleaved caspase-3.However,SAL reversed the aforementioned changes caused by PM_(2.5) by activating the siRT1-PGC-1α pathway.Conclusion SAL can activate siRT1-PGC-1ɑ to ameliorate PM2.5-induced lung injury.
