This prospective multicenter phase II study evaluated the clinical efficacy of neoadjuvant nivolumab-exclusive(N)and nivolumab–chemotherapy(N/C)combinations based on PD-L1 *** patients exhibited resectable clinical stage IIA–IIIB(AJCC 8th edition)NSCLC without EGFR/ALK *** received either mono-nivolumab(N)or nivolumab+nabpaclitaxel+carboplatin(N/C)for three cycles based on PD-L1 *** primary endpoint was the major pathological response(MPR).Key secondary endpoints included the pathologic complete response(pCR),objective response rate(ORR),and event-free survival(EFS).Baseline PD-L1 expression and perioperative circulating tumor DNA(ctDNA)status were correlated with pCR and ***-two patients were enrolled,with 46 undergoing *** MPR was 50.0%(26/52),with 25.0%(13/52)achieving pCR,and 16.7%and 66.7%for patients with PD-L1≥50%in N and N/C groups,***(25.0%)patients experienced grade 3 or higher immune-related adverse events during neoadjuvant *** with post-neoadjuvant ctDNA negativity was more likely to have pCR(39.1%)compared with those remained positive(6.7%,odds ratio=6.14,95%CI 0.84-Inf,p=0.077).With a median follow-up of 25.1 months,the 18-month EFS rate was 64.8%(95%CI 51.9–81.0%).For patients with ctDNA–***+,the 18m-EFS rate was 93.8%vs 47.3%(HR,0.15;95%CI 0.04,0.94;p=0.005).Immunochemotherapy may serve as an optimal neoadjuvant treatment even for patients with PD-L1 expression≥50%.ctDNA negativity following neoadjuvant treatment and surgery could help identify superior pathological and survival benefits,which requires further confirmation in a prospective clinical trial(NCT04015778).

Objective This study aimed to clarify the intervention effect of salidroside(SAL)on lung injury caused by PM_(2.5) in mice and illuminate the function of siRT1-PGC-1ɑ*** Specific pathogen-free(SPF)grade male C57BL/6 mice were randomly assigned to the following groups:control group,SAL group,PM_(2.5) group,SAL+PM_(2.5) *** the first day,SAL was given by gavage,and on the second day,PM_(2.5) suspension was given by intratracheal *** whole experiment consist of a total of 10 cycles,lasting 20 *** the end of treatment,blood samples and lung tissues were collected and *** of pathological changes in lung tissue using inverted microscopy and transmission electron *** expression of inflammatory,antioxidants,apoptosis,and siRT1-PGC-1ɑproteins were detected by Western *** Exposure to PM_(2.5) leads to obvious morphological and pathologica changes in the lung of ***_(2.5) caused a decline in levels of antioxidant-related enzymes and protein expressions of HO-1,Nrf2,SOD2,siRT1 and PGC-1ɑ,and an increase in the protein expressions of IL-6,IL-1β,Bax,caspase-9 and cleaved ***,SAL reversed the aforementioned changes caused by PM_(2.5) by activating the siRT1-PGC-1α *** SAL can activate siRT1-PGC-1ɑ to ameliorate PM2.5-induced lung injury.

Trial application of the envelope method to the potential ambiguity problem

The optical potential ambiguity is a long-standing problem in the analysis of elastic scattering *** a specific collid-ing system,ambiguous potential families can lead to different behaviors in the nearside and farside scattering *** contrast,the envelope method can decompose the experimental data into two components with negative and positive deflection angles,***,a question arises as to whether the comparison between the calculated nearside(or farside)component and the derived positive-deflection-angle(or negative-deflection-angle)component can help analyze the potential ambiguity *** this study,we conducted a trial application of the envelope method to the potential ambiguity *** envelope method was improved by including uncertainties in the experimental *** colliding systems of 16O+28si at 215.2 MeV and 12C+12C at 1016 MeV were considered in the *** each colliding system,the angular distribution experimental data were described nearly equally well by two potential sets,one of which is“surface transpar-ent”and the other is *** calculated angular distributions were decomposed into nearside and farside scattering *** the improved envelope method,the experimental data were decomposed into the positive-deflection-angle and negative-deflection-angle components,which were then compared with the calculated nearside and farside *** capability of the envelope method to analyze the potential ambiguities was also discussed.

Elasto-inertial microfluidic separation of microspheres with submicron resolution at high-throughput

Elasto-inertial microfluidic separation offers many advantages including high throughput and separation *** though the separation efficiency highly depends on precise control of the flow conditions,no concrete guidelines have been reported yet in elasto-inertial ***,we propose a dimensionless analysis for precise estimation of the microsphere behaviors across the interface of Newtonian and viscoelastic *** number,modified Weissenberg number,and modified elastic number are used to investigate the balance between inertial and elastic lift *** on the findings,we introduce a new dimensionless number defined as the width of the Newtonian fluid stream divided by microsphere *** proposed dimensionless analysis allows us to predict whether the microspheres migrate across the co-flow *** theoretical estimation is found to be in good agreement with the experimental results using 2.1-and 3.2-μm-diameter polystyrene microspheres in a co-flow of water and polyethylene oxide *** on the theoretical estimation,we also realize submicron separation of the microspheres with 2.1 and 2.5μm in diameter at high throughput,high purity(>95%),and high recovery rate(>97%).The applicability of the proposed method was validated by separation of platelets from similar-sized Escherichia coli(***).

Analysis of Aluminum Alloy Double-Pulse MIG Welding Arc signal Characteristics Based on Broadband Mode Decomposition

Welding voltage and current in arc signals are directly related to arc stability and welding *** experiments with different parameters were organized according to the orthogonal experimental design method by constructing an aluminum alloy double-pulse metal inert gas(MIG)welding arc electric signal test *** data acquisition system of the aluminum alloy MIG welding process was established to obtain real-time arc signal information reflecting the welding *** aluminum alloy’s collected double-pulse arc current signals are decomposed adaptively by broadband mode decomposition(BMD).The direct current(DC)signal,pulse signal,distortion signal,ripple signal,and noise signal are separated and extracted,and the composite multiscale fuzzy entropy(CMFE)is calculated for the component set of the electrical *** experimental results show that the current waveform obtained by the double-pulse MIG welding current signal is consistent with the corresponding weld forming ***,the composite multiscale fuzzy entropy is calculated for the arc characteristic *** rationality of matching process parameters and arc stability of aluminum alloy’s double-pulse MIG welding were evaluated.

10H2DA improves PCOS through autophagy-mediated mitochondrial fission dependent on the maintenance of calcium homeostasis between endoplasmic reticulum and mitochondria

Polycystic ovarian syndrome （PCOS） is the most principal reason for infertility in reproductive women， but no versatile treatment is feasible. Although trans-10-Hydroxy-2-decenoic acid （10H2DA） exhibits the anti-inflammatory， anti-cancerous and immunomodulatory properties， its role in improving PCOS remains elusive. This present study examined the effects of 10H2DA on the estrous cycle， ovarian morphology， reproductive hormone， glucose and insulin tolerances of PCOS mice， analyzed the binding between 10H2DA and free fatty acid receptor 4 （FFAR4）， and explored the regulation of 10H2DA on the c-Jun N-terminal kinase （JNK）/JUN （Jun proto-oncogene） signaling， cytosolic calcium ion (Ca2+)， endoplasmic reticulum （ER） Ca2+release， mitochondrial Ca2+， mitochondrial function， autophagy， lysosomal acidification， mitochondrial fission， secretion function and apoptosis of ovarian granulosa cells （GCs） using enzyme-linked immunosorbent assay， flow cytometry. western blotting， molecular docking， various fluorescent probes or plasmid， etc. The results found that 10H2DA alleviated the symptom of PCOS mice through improving the secretion dysfunction of GCs and protecting GCs against apoptosis. After binding to FFAR4 and inactivation of JNK/JUN signaling， 10H2DA restricted the release of ER Ca2+through targeting ryanodine receptors （RYRs） which had been identified as the downstream targets of JUN， diminished cytosolic Ca2+accumulation and prevented Ca2+influx into mitochondria via adjusting the mitochondrial calcium uptake 1 （MICU1） and mitochondrial calcium uniporter （MCU）. Concurrently， 10H2DA kept the integrity of mitochondrial function through maintaining mitochondrial Ca2+homeostasis and suppressed the discharge of mitochondrial reactive oxygen species （mtROS） into the cytosol through blocking the mitochondrial permeability transition pore opening to avoid the lipid peroxidation and ameliorate the secretory function and apoptosis of GCs. Dear Editor,Utilization of heterosis is fundamental to modern agricultural improvement and contributes significantly to food security and agricultural development(Ouyang et al.,2022).However,heterosis of F,hybrids cannot be stably ***,or asexual reproduction through seeds,offers the potential for the fixation of heterosis in F,hybrids and the formation of clonal offspring,which could have considerable implications for plant breeding and agriculture(Schmidt,2020).But naturally occurring apomixis has never been found in major ***,synthetic apomixis has been successfully generated by inducing incomplete meiotic development combined with parthenogenesis development to produce cloned rice(Oryza sativa)seeds(Khanday et al.,2019;Wang et al.,2019;Vernet et al.,2022;Wei et al.,2023).
