早期营养护理干预对食管癌根治术后患者免疫功能及营养指标的影响

目的分析食管癌根治术后患者行早期营养护理干预的效果。方法便利选取2021年6月—2023年6月徐州市中心医院收治的98例食管癌根治术患者作为研究对象,采用随机数表法分为两组,各49例。对照组实施常规护理,观察组实施早期营养护理。对比两组营养风险筛查量表(Nutrition Risk Screening-2002,NRS-2002)评分、微型营养评定简表(Mini Nutritional Assessment Short Form Version,MNASF)评分、免疫功能、营养不耐受发生率。结果护理后,观察组NRS-2002评分较对照组更低,差异有统计学意义(P<0.05);观察组MNASF评分为(11.27±1.56)分,较对照组的(9.02±1.14)分更高,差异有统计学意义(t=8.152,P<0.05);观察组免疫功能指标均优于对照组,差异有统计学意义(P均<0.05);观察组营养不耐受发生率(6.12%)较对照组(20.41%)更低,差异有统计学意义(χ^(2)=4.346,P<0.05)。结论食管癌根治术后行早期营养护理可改善患者营养状态,提升其营养水平与免疫功能,且营养不耐受发生率较低。

With the rapid development of 5G technology,it has become fast and easy for people to transmit information on the *** images can express information more intuitively,so transmitting information through images has excellent *** paper uses a new chaotic system called 1D-Sin-Logistic-Map(1D-SLM).1D-SLM has two control parameters,which can provide larger parameter space,and the parameter space in the chaotic state is *** Lyapunov exponent analysis(LE),bifurcation diagrams analysis,spectral entropy analysis(SE),and 0-1 test,it is verified that 1D-SLM has complex dynamic behavior and is very suitable for *** with other 1D chaotic systems,the 1D-SLM has a larger Lyapunov exponent(LE)and spectral entropy(SE).For color image encryption algorithms,only relying on chaotic mapping is not enough to ensure *** combined with 1D-SLM,we design a color image encryption algorithm,which is implemented by plane expansion,which reduces the correlation between the three channels of color *** experimental results show that the proposed cross-plane color image encryption algorithm is safe and resistant to common attack methods.

为评估一种添加了骨碎补的功能性奶粉对生长期大鼠骨健康的影响。将50只断乳SPF级雌性SD大鼠随机分为低钙对照组、碳酸钙对照组和奶粉低中高3个剂量组,灌胃90 d,于干预期结束前3 d进行钙代谢实验。干预结束后取双侧股骨进行股骨常规指标、骨密度、骨钙测定及抗酒石酸碱性磷酸酶免疫组化染色,并进行Micro-CT扫描骨微观结构,同时取血清测定I型胶原C端交联肽(C-telopeptide of typeⅠcollagen,CTX-Ⅰ)、Ⅰ型前胶原氨基端原肽(procollagen typeⅠamino-terminal peptide,PINP)和骨钙素(osteocalcin,BGP)的水平。结果表明,与低钙对照组相比,碳酸钙对照组和奶粉各剂量组的股骨干重和股骨长均显著升高(P<0.05,P<0.01),骨密度和骨钙含量均显著升高(P<0.01),CTX-Ⅰ、PINP及BGP水平均显著升高(P<0.01),其中高剂量组PINP/CTX-I比例显著降低(P<0.05);除奶粉低剂量组外,破骨细胞数量均显著降低(P<0.05,P<0.01);奶粉低和中剂量组的钙吸收率和钙储留率显著升高(P<0.01)。Micro-CT提示各奶粉剂量组的骨微观结构均优于低钙对照组。因此,受试奶粉具有有助于改善骨密度的作用,其机制可能与促进骨形成有关。

目的:对脊柱后路手术患者发生术中压力性损伤的危险因素进行Meta分析,为临床预防术中压力性损伤提供依据。方法:检索中英文数据库,检索时限为建库至2022年12月31日,收集有关脊柱后路手术患者发生术中压力性损伤危险因素的文献,由2名研究者独立进行文献筛选、资料提取和质量评价后,采用RevMan 5.4软件进行Meta分析。结果:共纳入10篇文献,共6402名患者,结果显示,脊柱后路手术患者术中压力性损伤的发生率为13%。BMI≥28 kg/m^(2)[OR=1.91,95%CI(1.06~3.41)]、合并糖尿病[OR=2.48,95%CI(1.56~3.95)]、麻醉持续时间≥4 h[OR=13.60,95%CI(6.30~29.34)]、手术时间≥4 h[OR=2.96,95%CI(1.97~4.45)]是脊柱后路手术患者发生术中压力性损伤的危险因素。结论:脊柱后路手术患者术中压力性损伤的发生率高,但对其危险因素的研究仍较局限,需要针对危险因素进行全面调查,为今后采取预防措施提供干预靶点,以纠正可控影响因素,做好医生、护士和麻醉师三方协同配合,以减少术中压力性损伤的发生。

A Profile of Native Integration Sites Used by φC31 Integrase in the Bovine Genome

The Streptomyces phage φC31 integrase can efficiently target attB-bearing transgenes to endogenous pseudo attP sites within mammalian genomes. To better understand the activity of φC31 integrase in the bovine genome, DNA sequences of 44 integration events were analyzed, and 32 pseudo attP sites were identified. The majority of these sites share a sequence motif that contains inverted repeats and has similarities to wild-type attP site. Genomic DNA flanking these sites typically contained repetitive sequence elements, such as short and long interspersed repetitive elements. These sequence features indicate that DNA sequence recognition plays an important role in guiding φC31-mediated site-specific integration. In addition, BF27 integration hotspot sites were identified in the bovine genome, which accounted for 13.6% of all isolated integration events and mapped to an intron of the deleted in liver cancer 1 （DLC1） gene. Also we found that the pseudo attP sites in the bovine genome had other features in common with those in the human genome. This study represents the first time that the sequence features of pseudo attP sites specific integrase system has great potential for applied modifications in the bovine genome were analyzed. We conclude that this site- of the bovine genome.

Effects of donor cells on in vitro development of cloned bovine embryos

The donor cells from different individuals and with different foreign genes introduced were investigated to determine their effects on the efficiency of somatic cell nuclear transfer （SCNT）. The bovine ear fibroblast from different individuals was isolated, cultured, and then transfected with foreign genes to establish the stable cell lines, which were used as donor cells for nuclear transfer. The oocytes were obtained through ovum pick up operation. After in vitro maturation, the M II phase oocytes were selected as receptors for nuclear transfer. The reconstructed embryos were cultured in vitro and observed at 2 h, 48 h, and 7 days after transfer to assess the rate of fusion using cleaved and blastocyst as the parameters of SCNT efficiency. The donor cells from different individuals （04036, 06081, 06088, and 06129） had no obvious effect on the fusion and cleaved rate, whereas there was significant difference in the blastocyst rate （P〈0.05）, and the rate was 62.3%, 37.0%, 35.1%, and 15.6%, respectively. There was no significant difference among the rate of fusion, cleaved and blastocyst in donor cells with different foreign genes （P〉0.05）. It was concluded that the genetic background of the donor cells could affect the efficiency of SCNT, while the introduction of foreign genes into the donor cells had no obvious effect on the efficiency. This study provides useful information for the SCNT and would benefit in promoting the efficiency.

Reconstructed embryos derived from intersubspecies somatic cell nuclear transfer （SCNT） have poorer developmental potential than those from intrasubspecies SCNT. Based on our previous study that Holstein dairy bovine （HD） mitochondrial DNA （mtDNA） haplotype compatibility between donor karyoplast and recipient cytoplast is crucial for SCNT embryo development, we performed intersubspecies SCNT using HD as donor karyoplast and Luxi yellow heifer （LY） as recipient cytoplast according to mtDNA haplotypes determined by polymerase chain reaction- restriction fragment length polymorphism （PCR-RFLP） analysis. The results demonstrated that intersubspecies mtDNA homotype SCNT embryos had higher pre- and post-implantation developmental competence than intrasubspecies mtDNA heterotype embryos as well as improved blastocyst reprogramming status, including normal H3K9 dimethylation pattern and promoter hypomethylation of pluripotent genes such as Oct4 and Sox2, suggesting that intersubspecies SCNT using LY oocytes maintains HD cloning efficiency and may reprogram HD nuclei to develop into a normal cloned animal ultimately. Our results indicated that karyoplast-cytoplast interactions and mtDNA haplotype compatibility may affect bovine intersubspecies SCNT efficiency. This study on bovine intersubspecies SCNT is valuable for understanding the mechanisms of mtDNA haplotype compatibility between karyoplast and cytoplast impacting the bovine SCNT efficiency, and provides an alternative and economic resource for HD cloning.

Hemophilia A （HA） is an inherited X-linked recessive bleeding disorder caused by coagulant factor VIII （FVIII） deficiency. Previous studies showed that introduction of mesenchymal stem cells （MSCs） modified by FVIll-expressing retrovims may result in phenotypic correction of HA animals. This study aimed at the investigation of an alternative gene therapy strategy that may lead to sustained FVIII transgene expression in HA mice. B-domain-de/eted human FVIll （hFVHIBD） vector was microinjected into single-cell embryos of wild-type mice to generate a transgenic mouse line, from which hFVIIIBD-MSCs were isolated, followed by transplantation into HA mice. RT-PCR and real-time PCR analysis demonstrated the expression of hFVlllBD in multi-organs of recipient HA mice. Immunohistochemistry showed the presence of hFVIIIBD positive staining in multi-organs of recipient HA mice. ELISA indicated that plasma hFVIIIBD level in recipient mice reached its peak （77 ng/ mL） at the 3rd week after implantation, and achieved sustained expression during the 5-week observation period. Plasma FVIII activities of recipient HA mice increased from 0% to 32% after hFVIIIBD-MSCs transplantation. APTT （activated partial thromboplastin time） value decreased in hFVIIIBD-MSCs transplanted HA mice compared with untreated HA mice （45.5 s vs. 91.3 s）. Our study demonstrated an effective phenotypic correction in HA mice using genetically modified MSCs from hFVIIIBD transgenic mice.

Amelioration of β^(654)-thalassemia in mouse model with the knockdown of aberrantly spliced β-globin mRNA

Large amounts of aberrantly spliced mRNA from the β^654 allele was present in erythroid cells, which might impair the erythropoiesis. A therapeutic strategy for β-thalassemia was explored by knocking down the aberrantly spliced mRNA of β-globin. Lentiviral vector with siRNA fragment targets on the specific portion of β^654-globin aberrantly spliced pre-mRNA was constructed. In HeLa β^654 cells, the siRNA vector could reduce approximately 60% of aberrantly spliced mRNA, which was assessed by RT-PCR and qRT-PCR. Furthermore, a disease model of β^654 thalassemia mice with lentiviral-mediated siRNA was produced by subzonal injection （named Hβi-Hbb^th-4/Hbb^＋ transgenic mice）. Our results showed that the hemotological parameters were improved in Hβi-Hbb^th-4/Hbb^＋ transgenic mice. This study provides a potential way for β^654-thalassemia therapy by knocking down the aberrantly spliced β-globin mRNA, whilst supporting that the aberrantly spliced β-globin mRNA may aggravate the disease.
