Immune checkpoint inhibitors:breakthroughs in cancer treatment

Over the past two decades, immunotherapies have increasingly been considered as first-line treatments for most cancers. One such treatment is immune checkpoint blockade(ICB), which has demonstrated promising results against various solid tumors in clinical trials. Monoclonal antibodies(mAbs) are currently available as immune checkpoint inhibitors(ICIs). These ICIs target specific immune checkpoints, including cytotoxic T-lymphocyte-associated antigen-4(CTLA-4) and programmed cell death protein 1(PD-1). Clinical trial results strongly support the feasibility of this immunotherapeutic approach. However, a substantial proportion of patients with cancer develop resistance or tolerance to treatment, owing to tumor immune evasion mechanisms that counteract the host immune response.Consequently, substantial research focus has been aimed at identifying additional ICIs or synergistic inhibitory receptors to enhance the effectiveness of anti-PD-1, anti-programmed cell death ligand 1(anti-PD-L1), and anti-CTLA-4 treatments. Recently, several immune checkpoint molecular targets have been identified, such as T cell immunoreceptor with Ig and ITIM domains(TIGIT), mucin domain containing-3(TIM-3), lymphocyte activation gene-3(LAG-3), V-domain immunoglobulin suppressor of T cell activation(VISTA), B and T lymphocyte attenuator(BTLA), and signal-regulatory protein α(SIRPα). Functional m Abs targeting these molecules are under development. CTLA-4, PD-1/PD-L1, and other recently discovered immune checkpoint proteins with distinct structures are at the forefront of research. This review discusses these structures, as well as clinical progress in m Abs targeting these immune checkpoint molecules and their potential applications.

Turbo-shaft engine adaptive neural network control based on nonlinear state space equation

Intelligent Adaptive Control(AC) has remarkable advantages in the control system design of aero-engine which has strong nonlinearity and uncertainty. Inspired by the Nonlinear Autoregressive Moving Average(NARMA)-L2 adaptive control, a novel Nonlinear State Space Equation(NSSE) based Adaptive neural network Control(NSSE-AC) method is proposed for the turbo-shaft engine control system design. The proposed NSSE model is derived from a special neural network with an extra layer, and the rotor speed of the gas turbine is taken as the main state variable which makes the NSSE model be able to capture the system dynamic better than the NARMA-L2 model. A hybrid Recursive Least-Square and Levenberg-Marquardt(RLS-LM) algorithm is advanced to perform the online learning of the neural network, which further enhances both the accuracy of the NSSE model and the performance of the adaptive controller. The feedback correction is also utilized in the NSSE-AC system to eliminate the steady-state tracking error. Simulation results show that, compared with the NARMA-L2 model, the NSSE model of the turboshaft engine is more accurate. The maximum modeling error is decreased from 5.92% to 0.97%when the LM algorithm is introduced to optimize the neural network parameters. The NSSE-AC method can not only achieve a better main control loop performance than the traditional controller but also limit all the constraint parameters efficiently with quick and accurate switching responses even if component degradation exists. Thus, the effectiveness of the NSSE-AC method is validated.

Electric Vehicle Charging Load Optimization Strategy Based on Dynamic Time-of-Use Tariff

Electric vehicle(EV)is an ideal solution to resolve the carbon emission issue and the fossil fuels scarcity problem in the future.However,a large number of EVs will be concentrated on charging during the valley hours leading to new load peaks under the guidance of static time-of-use tariff.Therefore,this paper proposes a dynamic time-of-use tariff mechanism,which redefines the peak and valley time periods according to the predicted loads using the fuzzy C-mean(FCM)clustering algorithm,and then dynamically adjusts the peak and valley tariffs according to the actual load of each time period.Based on the proposed tariff mechanism,an EV charging optimization model with the lowest cost to the users and the lowest variance of the grid-side load as the objective function is established.Then,a weight selection principle with an equal loss rate of the two objectives is proposed to transform the multi-objective optimization problem into a single-objective optimization problem.Finally,the EV charging load optimization model under three tariff strategies is set up and solved with the mathematical solver GROUBI.The results show that the EV charging load optimization strategy based on the dynamic time-of-use tariff can better balance the benefits between charging stations and users under different numbers and proportions of EVs connected to the grid,and can effectively reduce the grid load variance and improve the grid load curve.

Mechanism and control factors of hydrate plugging in multiphase liquid-rich pipeline flow systems: a review

There is nothing illogical in the concept that hydrates are easily formed in oil and gas pipelines owing to the low-temperature and high-pressure environment,although requiring the cooperation of flow rate,water content,gas-liquid ratio,and other specific factors.Therefore,hydrate plugging is a major concern for the hydrate slurry pipeline transportation technology.In order to further examine potential mechanisms underlying these processes,the present paper listed and analyzed the significant research efforts specializing in the mechanisms of hydrate blockages in the liquid-rich system,including oil-based,water-based,and partially dispersed systems(PD systems),in gathering and transportation pipelines.In addition,it summarized the influences of fluid flow and water content on the risk of hydrate blockage and discussed.In general,flow rate was implicated in the regulation of blockage risk through its characteristic to affect sedimentation tendencies and flow patterns.Increasing water content can potentiate the growth of hydrates and change the oil-water dispersion degree,which causes a transition from completely dispersed systems to PD systems with a higher risk of clogging.Reasons of diversity of hydrate plugging mechanism in oil-based system ought to be studied in-depth by combining the discrepancy of water content and the microscopic characteristics of hydrate particles.At present,it is increasingly necessary to expand the application of the hydrate blockage formation prediction model in order to ensure that hydrate slurry mixed transportation technology can be more maturely applied to the natural gas industry transportation field.

The HDAC inhibitor GCJ-490A suppresses c-Met expression through IKKα and overcomes gefitinib resistance in non-small cell lung cancer

Objective:The novel compound GCJ-490A has been discovered as a pan-histone deacetylase(HDAC)inhibitor that exerts potent inhibitory activity against HDAC1,HDAC3,and HDAC6.Because of the important roles of HDACs in lung cancer development and the high distribution of GCJ-490A in lung tissue,we explored the anti-tumor potency of GCJ-490A against non-small cell lung cancer(NSCLC)in vitro and in vivo in this study.Methods:The in vitro effects of GCJ-490A alone or combined with the EGFR inhibitor gefitinib against NSCLC were measured with proliferation,apoptosis,and colony formation assays.NSCLC xenograft models were used to investigate the efficacy of GCJ-490A combined with gefitinib for the treatment of NSCLC in vivo.Western blot assays,luciferase reporter assays,chromatin immunoprecipitation assays,quantitative real time-PCR,immunohistochemistry,and transcription factor activity assays were used to elucidate possible mechanisms.Results:GCJ-490A effectively inhibited NSCLC cell proliferation and induced apoptosis in vitro and in vivo.Interestingly,inhibition of HDAC1 and HDAC6 by GCJ-490A increased histone acetylation at the IKKαpromoter and enhanced IKKαtranscription,thus decreasing c-Met.Moreover,this c-Met downregulation was found to be essential for the synergistic anti-tumor activity of GCJ-490A and gefitinib.Conclusions:These findings highlight the promising potential of HDAC inhibitors in NSCLC treatment and provide a rational basis for the application of HDAC inhibitors in combination with EGFR inhibitors in clinical trials.

Degradation of black tea theaflavin through C-ring cleavage by gut microbiota

Increasing evidence has shown that gut microbiota plays important roles in metabolizing large molecular polyphenols to bioavailable and bioactive microbial metabolites.Theaflavin(TF)is one of the major color compounds in black tea and has demonstrated anti-inflammation,antioxidant,and anticancer effects properties.However,little is known about the metabolism of TF by gut microbiota in vivo.In this study,following the administration of TF to mice,the C-ring cleavage metabolites,dihydro-and tetrahydro-theaflavin(DH-TF and TH-TF)were detected in mouse feces by LC-MS and validated by authentic standards from in situ chemical reaction.The observation of the C-ring cleavage metabolites in TF-treated conventionalized mice but not in germ-free(GF)mice confirmed the role of gut microbiota in cleaving the C-rings of TF.The detection of DH-TF from the anaerobic incubation of TF with catechin-converting gut bacteria,Eggerthella lenta(Eggerth),suggested that the microbes with the capacity to cleave the C-ring of catechins were able to metabolize TF following the same mechanism.Additionally,three small phenolic metabolites were detected in mouse feces,and one of them was primarily detected in SPF mice not GF mice,which revealed that TF,subsequent to the cleaved C-ring,can be further metabolized into smaller phenolic metabolites by gut microbiota.Dose-dependent production of these metabolites were observed from the administration of 100 mg/kg to 400 mg/kg body weight of TF.In conclusion,gut microbiota can metabolize TF to the open-ring metabolites and the phenolic metabolites through the C-ring cleavage in mice.

In a genome-wide association study,we identified a rice UDP-glycosyltransferase gene,OsUGT706D2,whose transcription was activated in response to cold and submergence stress and to exogenous abscisic acid(ABA).OsUGT706D2 positively regulated the biosynthesis of tricin-4’-O-(syringyl alcohol)ether-7-O-glucoside at both the transcriptional and metabolic levels.OsUGT706D2 mediated cold and submergence tolerance by modulating the expression of stress-responsive genes as well as the abscisic acid(ABA)signaling pathway.Gain of function of OsUGT706D2 increased cold and submergence tolerance and loss of function of OsUGT706D2 reduced cold tolerance.ABA positively regulated OsUGT706D2-mediated cold tolerance but reduced submergence tolerance.These findings suggest the potential use of OsUGT706D2 for improving abiotic stress tolerance in rice.

Effects of dietary phosphorus level on growth,body composition,liver histology and lipid metabolism of spotted seabass(Lateolabrax maculatus)reared in freshwater

The present study was conducted to determine the effects of dietary phosphorus(P)levels on growth performance,body composition,liver histology and enzymatic activity,and expression of lipid metabolism-related genes in spotted seabass(Lateolabrax maculatus).Seven diets were prepared to contain available P levels of 0.48%(the control group),0.69%,0.89%,1.10%,1.28%,1.51%and 1.77%and feed fish(4.26±0.03 g)to satiety twice daily for 10 weeks.Significantly higher weight gain and specific growth rate were recorded at P levels of 0.69%-1.51%compared to the control group.Feed conversion ratio decreased with increasing P levels up to 0.89%and increased thereafter.The lowest liver lipid content,viscerosomatic index and lipid content of whole-body were obtained in the 0.89%-P group among dietary treatments.P and calcium(Ca)contents in whole body were increased,while liver triglyceride and cholesterol contents were decreased with increasing dietary P levels from 0.48%to 1.77%.The highest activity of hepatic lipase was recorded in the 1.10%-P group among dietary treatments.Compared to the control group,1.10%P enhanced the proportion of HUFA and reduced the proportion of SFA and MUFA.The histological observations showed that P deficiency(0.48%)led to the vacuolization of hepatocytes and increased number of lipid droplets.Meanwhile,overall liver tissue structure was improved when P level increased to 1.28%.Compared to the control group,expression of lipid metabolism-related genes such as FAS,ACC-2 and SREBP-1 was decreased at 0.89%-1.10%P group while an opposite trend was observed in the expression of PPARa2 and CPT-1 genes.The current study showed that 0.89%dietary P levels could promote growth performance of spotted seabass and reduce lipid accumulation in the liver.A broken-line regression analysis based on weight gain showed that the optimum dietary P level(available P)for juvenile spotted seabass reared in freshwater was 0.72%.

Transcriptome Profiling of Barley Cultivar Hua 30 MDEC in Response to Agrobacterium Infection

Agrobacterium-mediated transformation has been widely used in plants.However,the mechanism in plant cells’response to Agrobacterium infection was very complex.The mechanism of the determinants in host cell remains obscure,especially in barley,which is recalcitrant for Agrobacterium-mediated transformation.In the present study,microspore-derived embryogenic calli(MDEC)from barley elite cultivar were employed as unique subjects to characterize the mechanisms during the Agrobacterium infection process.Hua 30 MDEC can be successfully infected by Agrobacterium.RNA-sequencing at different infection points(0,2,6,12,24 hpi)was performed.The average expressional intensity of the whole genomics increased from 0 to 2 hpi,and then decreased subsequently.More upregulated than downregulated differentially expressed genes(DEGs)were counted at the same time.GO enrichment analysis showed that protein modification was significantly overrepresented in upregulated DEGs.Chromosome-related biological processes,gene expression and cellular metabolic processes were significantly overrepresented in downregulated DEGs.KEGG analysis showed that plant defense responses,phenylpropanoid biosynthesis and biosynthesis of amino acids were significantly enriched across the infection time course.Nine DEGs related to defense responses were identified.All DEGs were upregulated from 2 to 24 hpi.We speculate that these genes are possibly related to Agrobacterium infection.These findings will provide deep insights into the molecular events occurring during the process of Agrobacterium-mediated transformation.

Background:The calcineurin inhibitor(CNI)-based immune maintenance regimen that is commonly used after renal transplantation has greatly improved early graft survival after transplantation;however,the long-term prognosis of grafts has not been significantly improved.The nephrotoxicity of CNI drugs is one of the main risk factors for the poor long-term prognosis of grafts.Sirolimus(SRL)has been employed as an immunosuppressant in clinical practice for over 20 years and has been found to have no nephrotoxic effects on grafts.Presently,the regimen and timing of SRL application after renal transplantation vary,and clinical data are scarce.Multicenter prospective randomized controlled studies are particularly rare.This study aims to investigate the effects of early conversion to a low-dose CNI combined with SRL on the long-term prognosis of renal transplantation.Methods:Patients who receive four weeks of a standard regimen with CNI+mycophenolic acid(MPA)+glucocorticoid after renal transplantation in multiple transplant centers across China will be included in this study.At week 5,after the operation,patients in the experimental group will receive an additional administration of SRL,a reduction in the CNI drug doses,withdrawal of MPA medication,and maintenance of glucocorticoids.In addition,patients in the control group will receive the maintained standard of care.The patients’vital signs,routine blood tests,routine urine tests,blood biochemistry,serum creatinine,BK virus(BKV)/cytomegalovirus(CMV),and trough concentrations of CNI drugs and SRL at the baseline and weeks 12,24,36,48,72,and 104 after conversion will be recorded.Patient survival,graft survival,and estimated glomerular filtration rate will be calculated,and concomitant medications and adverse events will also be recorded.Conclusion:The study data will be utilized to evaluate the efficacy and safety of early conversion to low-dose CNIs combined with SRL in renal transplant patients.
