The effect of cylindrospermopsin on the bacterioplankton community:a microcosm experiment on water from Dishui Lake,Shanghai,China

Eutrophication and climate warming have intensified the global expansion of invasive cyanobacteria such as Cylindrospermopsis *** Chrysosporum ***(CYN)produced by species of the latter two genera may harm phytoplankton,zooplankton,and ***,effects of CYN on the bacterioplankton community have not been *** on high-throughput sequencing,we explored the effect of CYN on the structure and function of the bacterioplankton community by adding pure CYN to in-situ water collected from a brackish coastal shallow lake:Dishui Lake,*** found that most bacterioplankton taxa had a certain tolerance potential to CYN,but that high concentrations of CYN(40μg/L)caused a significant decrease in microbial abundance and functional *** the dominant phyla,Actinobacteria had the strongest tolerance to *** analysis indicated that CYN caused shifts in the community structure of the bacterioplankton,reducing community stability and structural *** CYN concentrations also reduced the correlation between the different bacterioplankton groups,and the abundances of some bacterial taxa associated with the denitrification function and the process of carbon transfer in the microbial food web were *** study provide s new insight into the response of the bacterioplankton communities to harmful algal toxins produced by cyanobacteria.

We prepared porous Fe-doped nickel cobaltate(Fe-NiCo_(2)O_(4))hollow hierarchical nanospheres through a facile self-templated synthetic *** to the porous hollow structure and composition,the Fe-NiCo_(2)O_(4)presented vastly superior electrocatalytic activity for the oxygen evolution reaction(OER),compared with NiCo_(2)O_(4)and the majority of other OER *** an aim of stimulating a current density of 10 mA$cm-2,the Fe-NiCo_(2)O_(4)catalyst needs an overpotential of 210 mV,which is on a par with the general properties of commercial *** addition,the Fe-NiCo_(2)O_(4)catalyst performed stably in long-term *** excellent activity and long-term stability showed that such catalysts have great promise for widespread application in the field of water splitting.

Smurf participates in Helicoverpa armigera diapause by regulating the transforming growth factor-βsignaling pathway

Diapause,an important strategy used by insects to avoid adverse environments,is regulated by various cell signaling *** results of our previous studies demonstrated that the transforming growth factor-β(TGF-β)signaling pathway regulated pupal diapause in Helicoverpa armigera,which was accompanied by downregulation of proteins in TGF-β***,to date the mechanism underlying this phenomenon remains ***,we cloned the E3 ubiquitin ligases gene *** vitro experiments showed that Smurf directly bound to TGF-βreceptor type I(TGFβRI)and *** Smurf promoted ubiquitination of TGFβRI and Smad2,thereby downregulating their protein ***,silencing of the Smurf gene suppressed ubiquitination of TGFβRI and Smad2 thereby increasing their protein *** from in vivo co-immunoprecipitation assays revealed that the binding of Smurf to TGFβRI or Smad2 was stronger in diapause pupae than in nondiapause *** of Smurf inhibitor A01 into diapause pupae markedly upregulated expression of TGFβRI and Smad2 proteins,leading to resumption of development in diapause *** together,these findings suggested that ubiquitin ligase E3 Smurf participated in *** diapause by regulating TGF-βsignaling,and thus could be playing a crucial role in insect diapause.

Application of Nanopore Single Molecule Detection Technology in Analysis of Xylan Dissolved in Ionic Liquid

Xylan is the most abundant hemicellulose in nature. As a new type of green organic solvent, ionic liquid shows good preservation ability for the functional groups of hemicellulose. In this paper, a single molecule detection technology based on glass nanopore was established to analyze xylan dissolved in ionic liquid. Arabino-xylan (AX) and beech xylan (BX) are respectively taken as the representatives of heterogeneous xylan and homogeneous xylan. Firstly, unmodified glass nanopore was used to detect the dissolved xylan in ionic liquid, and then poly(ethylene imine) (PEI) was used to modify the nanopore to change the surface charge in the nanopore and further enhance the interaction between the nanopore and the xylan molecule. It was found that before and after nanopore modification, at negative voltage and low positive voltage, AX didn't generate current blocking signal. On the contrary, BX didn't generate current blocking signal at positive voltage. This phenomenon may be due to the current disturbance driven by electrophoresis and electroosmosis of xylan molecules with weak negative charge. After statistics analysis, the current blocking signal of AX showed that the modified nanopore showed multiple peaks. It indicates that heterogeneous xylan and PEI modified nanopore had stronger interaction. The results show that the nanopore detection technology can show the structural difference of heterogenous branched chain and homogeneous straight chain based on the single characteristic current blocking signal and statistical information, providing a research basis for the structural analysis of water insoluble polysaccharides.

Multi-proxy reconstructions of hydrological changes from continental shelf sediments in the northern South China Sea during the interval 9200–6200 cal a BP

Past hydroclimatic conditions in southern China are poorly constrained owing to the lack of high-resolution marine-sediment records. In this study, we present high-resolution geochemical and grain-size records of marine sediments from the coastal shelf of the northern South China Sea to investigate regional hydrological *** suggest a warm and humid climate during the interval 9 200–7 600 cal a BP, followed by a cold and dry climate from 7 600 cal a BP to 6 500 cal a BP, and progressive humidification during the period 6 500–6 200 cal a BP.A prominent hydrological anomaly occurred during 7 600–6 500 cal a BP. This abrupt event corresponds closely to tropical Pacific and interhemispheric temperature gradients, suggesting that moisture variations in southern China may have been driven by interhemispheric and zonal Pacific temperature gradients via modulation of the intensity and location of the West Pacific subtropical high.

A G-type lectin receptor-like kinase regulates the perception of oomycete apoplastic expansin-like proteins

Phytophthora capsici is one of the most harmful pathogens in agriculture, which threatens the safe production of multiple crops and causes serious economic losses worldwide. Here, we identified a P. capsici expansin-like protein, Pc EXLX1, by liquid chromatography-tandem mass spectrometry from Nicotiana benthamiana apoplastic fluid infected with P. capsici. Clustered regularly interspaced short palindromic repeats/crispr associated protein9(CRISPR/Cas9)-mediated Pc EXLX1 knockout mutants exhibited significantly enhanced virulence,while the overexpression of Pc EXLX1 impaired the virulence. Prokaryotically expressed Pc EXLX1 activated multiple plant immune responses, which were BRI1-associated kinase 1(BAK1)-and suppressor of BIR1-1(SOBIR1)-dependent. Furthermore, overexpression of Pc EXLX1 homologs in N. benthamiana could also increase plant resistance to P. capsici. A G-type lectin receptor-like kinase from N. benthamiana, expansin-regulating kinase 1(ERK1), was shown to regulate the perception of Pc EXLX1 and positively mediate the plant resistance to P. capsici. These results reveal that the expansin-like protein, Pc EXLX1, is a novel apoplastic effector with plant immunity-inducing activity of oomycetes, perception of which is regulated by the receptor-like kinase, ERK1.

Sirt1-ROS-TRAF6 Signaling-Induced Pyroptosis Contributes to Early Injury in Ischemic Mice

Stroke is an acute cerebro-vascular disease with high incidence and poor prognosis,most commonly ischemic in *** recent years,increasing attention has been paid to inflammatory reactions as symptoms of a ***,the role of inflammation in stroke and its underlying mechanisms require *** this study,we evaluated the inflammatory reactions induced by acute ischemia and found that pyroptosis occurred after acute ischemia both in vivo and in vitro,as determined by interleukin-1β,apoptosis-associated speck-like protein,and *** early inflammation resulted in irreversible ischemic injury,indicating that it deserves thorough ***,acute ischemia decreased the Sirtuin 1(Sirtl)protein levels,and increased the TRAF6(TNF receptor associated factor 6)protein and reactive oxygen species(ROS)*** further exploration,both Sirtl suppression and TRAF6 activation were found to contribute to this *** Sirtl levels were responsible for the production of ROS and increased TRAF6 protein levels after ischemic ***,N-acetyl-L-cysteine,an ROS scavenger,suppressed the TRAF6 accumulation induced by oxygen-glucose deprivation via suppression of ROS *** phenomena indicate that Sirtl is upstream of ROS,and ROS bursts result in increased TRAF6 ***,the activation of Sirtl during the period of ischemia reduced ischemiainduced injury after 72 h of reperfusion in mice with middle cerebral artery *** sum,these results indicate that pyroptosis-dependent machinery contributes to the neural injury during acute ischemia via the Sirt1-ROS-TRAF6 signaling *** propose that inflammatory reactions occur soon after oxidative stress and are detrimental to neuronal survival;this provides a promising therapeutic target against ischemic injuries such as a stroke.
