Liraglutide directly protects cardiomyocytes against reperfusion injury possibly via modulation of intracellular calcium homeostasis

Background Liraglutide is glucagon-like peptide-1 receptor agonist for treating patients with type 2 diabetes mellitus. Our previous studies have demonstrated that liraglutide protects cardiac function through improving endothelial function in patients with acute myocardial infarction undergoing percutaneous coronary intervention. The present study will investigate whether liraglntide can perform direct protective effects on cardiomyocytes against reperfusion injury. Methods In vitro experiments were performed using H9C2 cells and neonatal rat ventricular cadiomyocytes undergoing simulative hypoxia/reoxygenation （H/R） induction. Cardiomyocytes apoptosis was detected by fluorescence TUNEL. Mitochondrial membrane potential （AWm） and intracellular reactive oxygen species （ROS） was assessed by JC-1 and DHE, respectively. Fura-2/AM was used to measure intracellular Ca2＋ concentration and calcium transient. Immtmofluorescence staining was used to assess the expression level of sarcoplasmic reticulum Ca2＋-ATPase （SERCA2a）. In vivo experiments, myocardial apoptosis and expression of SERCA2a were detected by colorimetric TUNEL and by immunofluorescence staining, respectively. Results In vitro liraglutide inhibited cardiomyotes apoptosis against H/R. △mψ of cardiomyocytes was higher in liraglntide group than H/R group. H/R increased ROS production in H9C2 cells which was attenuated by liraglutide. Liraglutide significantly lowered Ca2＋ overload and improved calcium transient compared with H/R group, lmmunofluorescence staining results showed liraglutide promoted SERCA2a expression which was decreased in H/R group. In ischemia/reperfusion rat hearts, apoptosis was significantly attenuated and SERCA2a expression was increased by liraglutide compared with H/R group. Conclusions Liraglutide can directly protect cardiomyocytes against reperfusion injury which is possibly through modulation of intracellular calcium homeostasis.

Inflammation-based different association between anatomical severity of coronary artery disease and lung cancer

BACKGROUND Coronary artery disease(CAD)is associated with *** role of inflammation in the association of CAD with cancer remains *** study investigated whether inflammation could impact the relationship between CAD and lung *** The study involved 96 newly diagnosed lung cancer patients without receiving anti-cancer therapy and 288 matched non-cancer *** the patients underwent coronary angiography and were free from previous percutaneous coronary inter-vention or coronary artery bypass *** score(SXscore)were used to assess severity of *** SXscore(SXhigh)grade was defined as SXscore>16(highest quartile).Neutrophil-to-lymphocyte ratio(NLR)served as an inflammatory ***-high grade referred to NLR>2.221(median).RESULTS Among 384 study patients,380 patients(98.96%)had NLR value(median:2.221,interquartile range:1.637-3.040).Com-pared to non-cancer patients,lung cancer patients had higher rate of SXhigh among total study patients(P=0.014)and among pat-ients with NLR-high(P=0.006),but had not significantly higher rate of SXhigh among patients with NLR-low(P=0.839).Multiv-ariate logistic regression analysis showed that SXhigh was associated with lung cancer[odds ratio(OR)=1.834,95%CI:1.063-3.162,P=0.029].Subgroup analysis showed that SXhigh was associated with lung cancer among patients with NLR-high(OR=2.801,95%CI:1.355-5.794,P=0.005),however,the association between SXhigh and lung cancer was not significant among pati-ents with NLR-low(OR=0.897,95%CI:0.346-2.232,P=0.823).CONCLUSIONS Inflammation could lead different association between anatomical severity of CAD and lung *** of CAD was significantly associated with increased risk of lung cancer among patients with high inflammation rather than am-ong patients with low inflammation.

Lowering the synthesis temperature of boron nitride nanotubes(BNNTs)is crucial for their *** primary reason for adopting a high temperature is to enable the effective activation of highmelting-point solid *** this study,we developed a novel approach for efficiently activating boron by introducing alkali metal compounds into the conventional MgO–B *** approach can be adopted to form various low-melting-point AM–Mg–B–O growth *** growth systems have improved catalytic capability and reactivity even under low-temperature conditions,facilitating the synthesis of BNNTs at temperatures as low as 850℃.In addition,molecular dynamics simulations based on density functional theory theoretically demonstrate that the systems maintain a liquid state at low temperatures and interact with N atoms to form BN *** findings offer novel insights into the design of boron activation and are expected to facilitate research on the low-temperature synthesis of BNNTs.

Photo‑Energized MoS_(2)/CNT Cathode for High‑Performance Li–CO_(2)Batteries in a Wide‑Temperature Range

Li–CO_(2) batteries are considered promising energy storage systems in extreme environments such as Mars;however,severe performance degradation will occur at a subzero temperature owning to the sluggish reaction ***,a photo-energized strategy adopting sustainable solar energy in wide working temperature range Li–CO_(2) battery was achieved with a binder-free MoS_(2)/carbon nanotube(CNT)photo-electrode as *** unique layered structure and excellent photoelectric properties of MoS_(2) facilitate the abundant generation and rapid transfer of photo-excited carriers,which accelerate the CO_(2) reduction and Li_(2)CO_(3) decomposition upon *** illuminated battery at room temperature exhibited high discharge voltage of 2.95 V and mitigated charge voltage of 3.27 V,attaining superior energy efficiency of 90.2%and excellent cycling stability of over 120 *** at an extremely low temperature of−30℃,the battery with same electrolyte can still deliver a small polarization of 0.45 V by the photoelectric and photothermal synergistic mechanism of MoS_(2)/CNT *** work demonstrates the promising potential of the photo-energized wide working temperature range Li–CO_(2) battery in addressing the obstacle of charge overpotential and energy efficiency.

To prepare a conductive polymer actuator with decent performance,a self-built experimental platform for the preparation of polypyrrole film is *** of the essential goals is to examine the mechanical characteristics of the actuator in the presence of various combinations of process parameters,combined with the orthogonal test method of"four factors and three levels".The bending and sensing characteristics of actuators of various sizes are methodically examined using a self-made bending polypyrrole *** functional relationship between the bending displacement and the output voltage signal is established by studying the characteristics of the actuator sensor subjected to various degrees of *** experimental results reveal that the bending displacement of the actuator tip almost exhibits a linear variation as a function of length and *** the voltage reaches 0.8 V,the bending speed of the actuator tends to be ***,the mechanical properties of the self-assembled polypyrrole actuator are verified by the design and fabrication of the microgripper.

Effect of Nb-V microalloying on the hot deformation behavior of medium Mn steels

The influence of Nb-V microalloying on the hot deformation behavior and microstructures of medium Mn steel(MMS)was investigated by uniaxial hot compression *** establishing the constitutive equations for simulating the measured flow curves,we successfully constructed deformation activation energy(Q)maps and processing maps for identifying the region of flow *** concluded the following consequences of Nb-V alloying for MMS.(i)The critical strain increases and the increment diminishes with the increasing deformation temperature,suggesting that NbC precipitates more efficiently retard dynamic recrystallization(DRX)in MMS compared with solute Nb.(ii)The deformation activation energy of MMS is significantly increased and even higher than that of some reported high Mn steels,suggesting that its ability to retard DRX is greater than that of the high Mn content.(iii)The hot workability of MMS is improved by narrowing the hot processing window for the unstable flow stress,in which fine recrystallized and coarse unrecrystallized grains are present.

Tumor inflammation-specific self-amplifying fluorescent molecular probes induce an anti-tumor immune response

Enhancing the concentration of exogenous molecular drugs within the tumor microenvironment through enzyme-catalyzed polymerization presents a novel strategy for cancer therapy. Nonetheless, the optimization of the catalytic efficiency is often impeded by the inefficient expression of enzymes. Herein, we reported a self-amplifying fluorescent molecular probe, Bis-HTPICG, for photodynamic therapy(PDT) and subsequent PDT-induced immunoreaction. The Bis-HTP-ICG probe possesses a noticeable enzyme-catalyzed polymerization facilitated by myeloperoxidase(MPO), a crucial enzyme secreted by neutrophils at inflammation sites. Upon exposure to laser irradiation, Bis-HTP-ICG showed a high PDT efficacy, inducing an acute inflammatory response that stimulates further recruitment of neutrophils and then elevated MPO secretion. The heightened level of MPO enhances the accumulation of the Bis-HTP-ICG via self-polymerization or binding with intratumoral proteins following MPO enzyme catalysis, instigating a self-amplifying chain reaction cycle involving Bis-HTP-ICG, neutrophils and ***, PDT efficiently incites immunogenic cell death(ICD) in tumor cells, initiating an anti-tumor immune response including dendritic cells(DCs) maturation, T cell proliferation and reprogramming of tumor-associated neutrophils(TANs). This work portrays a promising strategy for self-amplification of fluorescent molecular probes through adjustable enzyme levels,potentially offering a unique avenue to enhance the tumor accumulation of molecular drugs for improved tumor therapy.

将监护定位于职责或权利,监护关系的外部效力都具有绝对性,对其侵犯需适用《民法典》侵权责任编规则。侵权人包括令被监护人从监护状态中脱离的行为人,以及延续其脱离状态的收买人等在后行为人。即便其他行为也可能延续被监护人脱离监护的状态,这一假设原因也不减免在后行为人的损害赔偿责任。无过失的受托监护人并非侵权人。除监护人的身份利益之外,脱离监护关系同时有损被监护人的人格利益,因此被亲生父母出卖的子女可主张侵害人格权的损害赔偿。脱离监护型侵权的损害赔偿范围包括被监护人成年后持续产生的损害,损害赔偿请求权适用普通诉讼时效。精神损害赔偿请求权的主体不应限于父母和其他近亲属,非近亲属监护人等也可主张。
