Skeletal muscle plays a significant role in both local and systemic energy metabolism. The current investigation aims to explore the role of the Bambi gene in skeletal muscle, focusing on its implications for muscle hypertrophy and systemic metabolism. We hypothesize that skeletal muscle-specific deletion of Bambi induces muscle hypertrophy, improves metabolic performance, and activates thermogenic adipocytes via the reprogramming of progenitor of i WAT, offering potential therapeutic strategies for metabolic syndromes. Leveraging the Chromatin immunoprecipitation(ChIP)-seq and bioinformatics analysis, Bambi gene is shown to be a direct target of HIF2α, which is further confirmed by ChIP-qPCR and promoter luciferase assay. Skeletal muscle-specific Bambi deletion led to significant muscle hypertrophy and improved metabolic parameters, even under high-fat diet conditions. This deletion induced metabolic reprogramming of stromal vascular fractions(SVFs) into thermogenic adipocytes, contributing to systemic metabolic improvements, potentially through the secretory ***, mice with skeletal muscle-specific Bambi deletion demonstrate resistance to high-fat diet-induced metabolic disorders, highlighting a potential therapeutic pathway for metabolic syndrome management. Thus, skeletal muscle-specific deletion of Bambi triggers muscle growth, enhances metabolic performance, and activates thermogenic adipocytes. These findings suggest Bambi as a novel therapeutic target for metabolic syndromes, providing new insights into the interaction between muscle hypertrophy and systemic metabolic improvement. The study underscores the potential of manipulating muscle physiology to regulate whole-body metabolism, offering a novel perspective on treating metabolic disorders.

An Integration Testing Framework and Evaluation Metric for Vulnerability Mining Methods

Software an important way to vulnerability mining is detect whether there are some loopholes existing in the software, and also is an important way to ensure the secu- rity of information systems. With the rapid development of information technology and software industry, most of the software has not been rigorously tested before being put in use, so that the hidden vulnerabilities in software will be exploited by the attackers. Therefore, it is of great significance for us to actively de- tect the software vulnerabilities in the security maintenance of information systems. In this paper, we firstly studied some of the common- ly used vulnerability detection methods and detection tools, and analyzed the advantages and disadvantages of each method in different scenarios. Secondly, we designed a set of eval- uation criteria for different mining methods in the loopholes evaluation. Thirdly, we also proposed and designed an integration testing framework, on which we can test the typical static analysis methods and dynamic mining methods as well as make the comparison, so that we can obtain an intuitive comparative analysis for the experimental results. Final- ly, we reported the experimental analysis to verify the feasibility and effectiveness of the proposed evaluation method and the testingframework, with the results showing that the final test results will serve as a form of guid- ance to aid the selection of the most appropri- ate and effective method or tools in vulnera- bility detection activity.

A Method for Software Vulnerability Detection Based on Improved Control Flow Graph

With the rapid development of software technology, software vulnerability has become a major threat to computer security. The timely detection and repair of potential vulnerabilities in software, are of great significance in reducing system crashes and maintaining system security and integrity. This paper focuses on detecting three common types of vulnerabilities: Unused_Variable, Use_of_Uninitialized_Variable, and Use_After_ Free. We propose a method for software vulnerability detection based on an improved control flow graph(ICFG) and several predicates of vulnerability properties for each type of vulnerability. We also define a set of grammar rules for analyzing and deriving the three mentioned types of vulnerabilities, and design three vulnerability detection algorithms to guide the process of vulnerability detection. In addition, we conduct cases studies of the three mentioned types of vulnerabilities with real vulnerability program segments from Common Weakness Enumeration(CWE). The results of the studies show that the proposed method can detect the vulnerability in the tested program segments. Finally, we conduct manual analysis and experiments on detecting the three types of vulnerability program segments(30 examples for each type) from CWE, to compare the vulnerability detection effectiveness of the proposed method with that of the existing detection tool Cpp Check. The results show that the proposed method performs better. In summary, the method proposed in this paper has certain feasibility and effectiveness in detecting the three mentioned types of vulnerabilities, and it will also have guiding significance for the detection of other common vulnerabilities.

Background The interplay between gut microbiota and tumor microenvironment(TME)in the pathogenesis of colorectal cancer(CRC)is not well ***,we elucidated the functional role of Bifidobacterium adolescentis(B.a)on CRC and investigated its possible mechanism on the manipulation of cancer-associated fibroblasts(CAFs)in *** Different CRC animal models and various cell line models were established to explore the function of B.a on *** single-cell RNA sequencing(scRNA-seq)or flow cytometry was used to detect the cell subsets in the TME of *** blot,quantitative real-time polymerase chain reaction(qRT-PCR),or immunofluorescence staining were performed to examine the activation of Wnt signaling and growth arrest specific 1(GAS1)on CD143+*** immunoprecipitation quantitative real-time PCR(CHIP-qPCR)was performed to investigate the regulation of transcription factor 4(TCF4)on ***-immunofluorescence assay examined the expression level of CD143 and GAS1 on tissue *** We found that B.a abundance was significantly reduced in CRC patients from two independent cohorts and the bacteria database of *** with B.a suppressed ApcMin/+spontaneous or AOM/DSS-induced tumorigenesis in ***-seq revealed that B.a facilitated a subset of CD143+CAFs by inhibiting the infiltration of Th2 cells,while promoting the TNF-alpha+B cells in ***143+CAFs highly expressed GAS1 and exhibited tumor suppressive ***,GAS1 was activated by the Wnt/β-catenin signaling in CD143+CAFs.B.a abundance was correlated with the expression level of CD143 and *** level of CD143+CAFs predicted the better survival outcome in CRC *** These results highlighted that B.a induced a new subset of CD143+CAFs by Wnt signaling-regulated GAS1 to suppress tumorigenesis and provided a novel therapeutic target for probiotic-based modulation of TME in CRC.

The gut microbiota plays a pivotal role in systemic metabolic processes and in particular functions,such as developing and preserving the skeletal muscle ***,the interplay between gut microbiota/metabolites and the regulation of satellite cell(SC)homeostasis,particularly during aging,remains *** propose that gut microbiota and its metabolites modulate SC physiology and homeostasis throughout skeletal muscle development,regeneration,and aging *** investigation reveals that microbial dysbiosis manipulated by either antibiotic treatment or fecal microbiota transplantation from aged to adult mice,leads to the activation of SCs or a significant reduction in the total ***,employing multi-omics(e.g.,RNA-seq,16S r RNA gene sequencing,and metabolomics)and bioinformatic analysis,we demonstrate that the reduced butyrate levels,alongside the gut microbial dysbiosis,could be the primary factor contributing to the reduction in the number of SCs and subsequent impairments during skeletal muscle ***,butyrate supplementation can mitigate the antibiotics-induced SC activation irrespective of gut microbiota,potentially by inhibiting the proliferation and differentiation of SCs/*** butyrate effect is likely facilitated through the monocarboxylate transporter 1(Mct1),a lactate transporter enriched on membranes of SCs and *** a result,butyrate could serve as an alternative strategy to enhance SC homeostasis and function during skeletal muscle *** findings shed light on the potential application of microbial metabolites in maintaining SC homeostasis and preventing skeletal muscle aging.
