Nicotinamide adenine dinucleotide(NAD+)plays an essential role in cellular metabolism,mitochondrial homeostasis,inflammation,and ***,the role of NAD+-regulated genes,including coding and long non-coding genes in cancer development is poorly *** constructed a prediction model based on the expression level of NAD+metabolism-related genes(NMRGs).Furthermore,we validated the expression of NMRGs in gastric cancer(GC)tissues and cell lines;additionally,β-nicotinamide mononucleotide(NMN),a precursor of NAD+,was used to treat the GC cell lines to analyze its effects on the expression level of NMRGs lncRNAs and cellular proliferation,cell cycle,apoptosis,and senescence-associated secretory phenotype(SASP).A total of 13 NMRGs-related lncRNAs were selected to construct prognostic risk signatures,and patients with high-risk scores had a poor *** immune checkpoint genes were upregulated in the high-risk *** addition,cell cycle,epigenetics,and senescence were significantly downregulated in the high-risk ***,we found that the levels of immune cell infiltration,including CD8 T cells,CD4 naïve T cells,CD4 memory-activated T cells,B memory cells,and naïve B cells,were significantly associated with risk ***,the treatment of NMN showed increased proliferation of AGS and MKN45 *** addition,the expression of SASP factors(IL6,IL8,IL10,TGF-β,and TNF-α)was significantly decreased after NMN *** conclude that the lncRNAs associated with NAD+metabolism can potentially be used as biomarkers for predicting clinical outcomes of GC patients.

Objective:To observe the therapeutic effects and underlying mechanism of baicalin against colon ***:The effects of baicalin on the proliferation and growth of colon cancer cells MC38 and *** were observed and predicted potential molecular targets of baicalin for colon cancer therapy were studied by network ***,molecular docking and drug affinity responsive target stability（DARTS）analysis were performed to confirm the interaction between potential targets and ***,the mechanisms predicted by in silico analyses were experimentally verified in-vitro and ***:Baicalin significantly inhibited proliferation,invasion,migration,and induced apoptosis in MC38 and CT26 cells（all P<0.01）.Additionally,baicalin caused cell cycle arrest at the S phase,while the G0/G1phase was detected in the tiny portion of the *** network pharmacology analysis identified 6 therapeutic targets associated with baicalin,which potentially affect various pathways including 39 biological processes and 99 sugnaling *** addition,molecular docking and DARTS predicted the potential binding of baicalin with cyclin dependent kinase inhibitor 2A（CDKN2A）,protein kinase B（AKT）,caspase 3,and mitogen-activated protein kinase（MAPK）.In vitro,the expressions of CDKN2A,MAPK,and p-AKT were suppressed by baicalin in MC38 and CT26 *** vivo,baicalin significantly reduced the tumor size and weight（all P<0.01） in the colon cancer mouse model via inactivating p-AKT,CDKN2A,cyclin dependent kinase 4,cyclin dependent kinase 2,interleukin-1,tumor necrosis factor α,and activating caspase 3 and mouse double minute 2 homolog signaling（all P<0.05）.Conclusion:Baicalin suppressed the CDKN2A protein level to prevent colon cancer and could be used as a therapeutic target for colon cancer.

Background Chitinase is an enzyme that hydrolyzes chitin,a major component of the exoskeleton of insects,including plant pests like *** present study aimed to investigate the expression of chemically synthesized barley ch1 and chi2 genes in cotton(Gossypium hirsutum)through Agrobacterium-mediated ***-five putative transgenic cotton plants were obtained,out of which fifteen plants successfully survived and were shifted to the *** gene-specific primers,amplification of 447 bp and 401 bp fragments confirmed the presence of the ch1 and chi2 genes in five transgenic cotton plants of the T0 *** five plants were further evalu-ated for their mRNA expression *** T0 transgenic cotton plants with the highest mRNA expression level and better yield performance in field,were selected to raise their subsequent *** The T1 cotton plants showed the highest mRNA expression levels of 3.5-fold in P10(2)for the ch1 gene and 3.7-fold in P2(1)for the chi2 *** in situ hybridization(FISH)confirmed a single copy number of ch1 and chi2(hemizygous)on chromosome ***,the efficacy of transgenes on whitefly was evaluated through an insect bioassay,where after 96 h of infestation,mortality rates of whitefly were calculated to be 78%–80%in transgenic cotton *** number of eggs on transgenic cotton plants were calculated to be 0.1%–0.12 per plant compared with the non-transgenic plants where egg number was calculated to be 0.90–1.00 per *** Based on these findings,it can be concluded that the chemically synthesized barley chitinase genes(ch1 and chi2)have the potential to be effective against insects with chitin exoskeletons,including *** transgenic cotton plants expressing these genes showed increased resistance to whiteflies,resulting in reduced egg numbers and higher mortality rates.

Knowledge, Attitude, and Practice of Using Helmets in Children amongst Parents to Prevent Head Injuries: A Cross-Sectional Study in Riyadh, Saudi Arabia

Objectives: The primary objective was to characterize the range of Knowledge, Attitude, and Practice (KAP) of Helmet use in children amongst parents to prevent head injuries and death. Methods: This is a cross-sectional study, done by online survey using a snowball sampling technique, the number of included responses were 386 parents (Male and female) living in Riyadh Aged 21 - 60 years old or above. Results: The study showed that there is a difference in Parents’ belief in the importance of helmet use while riding a Bicycle vs Motorcycle/Quad bike and that was affected by parents’ education level, almost all the people who answered the survey (76.7%) agree that it is important for their children to wear a helmet when riding both a Bicycle and a Motorcycle or Quadbike with a cumulative percentage of (93.8%). And almost all agreed on multiple approaches to help increase helmet use be it by forcing rental shops to give out helmets, forcing sellers to recommend the use of helmets, increasing awareness campaigns, and imposing fines for not wearing helmets. Conclusions: This study is the first to explore Family helmet use while riding Bicycles and Motorcycles/Quad bikes. Although Parent’s belief in the importance of helmet use for their children was high, it is clear that the level of practice is low. With that the risk of head injuries might be high, our findings suggest that safety interventions for increasing pediatric helmet use are needed to increase helmet use and reduce the risk of head injury and hospitalization.

Cardiac amyloidosis(CA)is caused by deposition of amyloid fibrils in the myocardium and has two main sub-types,transthyretin cardiac amyloidosis(ATTR)and immunoglobulin light chain cardiac amyloidosis(AL).ATTR is further dif-ferentiated into wild-type(wtATTR)and hereditary(hATTR),depending on the absence or presence of mutation in the trans-thyretin *** increased recognition of disease with the improvement in diagnostic armamentarium and serendipitous ad-vancements in the therapeutic landscape have changed the status of CA from being a rare and untreatable disease to being a not-so-rare and treatable *** ATTR and AL have certain clinical aspects that can provide early clues for the *** electrocardiography followed by echocardiography and subsequently cardiac magnetic resonance can raise suspicion for CA,the definitive diagnosis of ATTR is non-invasively established by bone scintigraphy while that of AL always needs histological *** of CA can be gauged by serum biomarker-based staging of both ATTR and *** therapies work by silen-cing or stabilizing TTR or by degrading amyloid fibrils,while AL is managed with anti-plasma cell therapies and autologous stem cell transplant.

The linear characteristics of the unstable mode of ion-acoustic waves are examined in an electrostatic electron-ion plasma composed of streaming hot electrons,non-streaming cold electrons and dynamical positive *** plasma under consideration is modeled by using a non-gyrotropic nonextensive q-distribution function in which the free energy source for wave excitation is provided by the relative directed motion of streaming hot electrons with respect to the other plasma *** the frame work of kinetic model,a linearized set of Vlasov-Poisson's equations are solved to obtain the analytical expressions for dispersion relation and Landau damping *** threshold condition for the unstable ion-acoustic wave is derived to assess the stability of the wave in the presence of nonextensive *** in the wave spectrum and nontrivial effects of q-nonextensive parameter on the ion-acoustic waves can be of interest for the readers in the regions of Saturns's magnetosphere.

We have measured the dielectric constant for NdMn2O5 in an external magnetic field to map out the magnetoelectric phase diagram. The phase diagram corresponds well with the previously reported data of neutron diffraction and magnetic susceptibility. Our main finding is the observation of a dielectric anomaly in the low temperature phase with a strong magnetoelectric effect, which is attributed to the independent Nd^3＋ ordering. Moreover, the absence of the dielectric anomaly in the paramagnetic phase is discussed, keeping in view the exchange interaction and its dependence on the rareearth R^3＋ ionic radius.

Innovations on the Internet of Everything(IoE)enabled systems are driving a change in the settings where we interact in smart units,recognized globally as smart city ***,intelligent video-surveillance systems are critical to increasing the security of these smart *** precisely,in today’s world of smart video surveillance,person re-identification(Re-ID)has gained increased consideration by *** researchers have designed deep learningbased algorithms for person Re-ID because they have achieved substantial breakthroughs in computer vision *** this line of research,we designed an adaptive feature refinementbased deep learning architecture to conduct person *** the proposed architecture,the inter-channel and inter-spatial relationship of features between the images of the same individual taken from nonidentical camera viewpoints are focused on learning spatial and channel *** addition,the spatial pyramid pooling layer is inserted to extract the multiscale and fixed-dimension feature vectors irrespective of the size of the feature ***,the model’s effectiveness is validated on the CUHK01 and CUHK02 *** compared with existing approaches,the approach presented in this paper achieves encouraging Rank 1 and 5 scores of 24.6% and 54.8%,respectively.

The main theme of the current article is to investigate the heat transfer in the pulsatile flow of an electrically conducting viscous fluid in a constricted channel under the effect of the magnetic field and thermal *** unsteady governing equations simplified for low conducting fluids are solved numerically by finite difference method using stream-vorticity function *** influence of the flow parameters such as the Hartmann number(magnetic parameter),Strouhal number(flow pulsation parameter),Prandtl number,and radiation parameter is studied on the relevant flow *** influence of different emerging parameters on the skin friction coefficient and Nusselt number are examined,as *** general,the profiles are observed to exhibit a relatively more regular pattern upstream of the construction than that downstream of the constriction.
