Antisense expression of PKCα improved sensitivity of SGC7901/VCR cells to doxorubicin

AIM:To explore whether antisense blocking of protein kinase C alpha(PKCα)would reverse multi-drug resistance(MDR)in the vincristine(VCR)-resistant human gastric cancer cell line SGC7901/VCR. METHODS:SGC7901/VCR cells expressing antisense PKCα,SGC7901/VCR/aPKC,were established by transfection with a recombinant plasmid reversely inserted with PKCα*** vector(PCI-neo)transfected cell clones,SGC7901/VCR/neo,served as the *** blot method was used to detect PKCαcontent in SGC7901,SGC7901/VCR,SGC7901/ VCR/neo and SGC7901/VCR/aPKC cells,using PKCα-specific *** sensitivity of SGC7901,SGC7901/ VCR,SGC7901/VCR/neo and SGC7901/VCR/aPKC cells to doxorubicin(DOX)in vitro was determined by MTT *** uptake of DOX in these cells was detected with fluorescence ***:Western blot analysis showed that the PKCαprotein level was about 8.7-fold higher in SGC7901/ VCR cells than that in SGC7901 cells,whereas the protein expression of PKCαwas reduced by 78%in SGC7901/VCR/aPKC cells when compared with the SGC7901/VCR ***7901/VCR/aPKC cells had a 4.2-fold increase in DOX cytotoxicity,accompanied by a 1.7-fold increase of DOX accumulation in comparison with SGC7901/VCR cells. CONCLUSION:PKCαpositively regulates MDR in SGC7901 cells,and inhibition of PKCαcan partially attenuate MDR in human gastric cancer cells.

Neuromyelitis optica spectrum disorders are neuroinflammatory demyelinating disorders that lead to permanent visual loss and motor *** date,no effective treatment exists as the exact causative mechanism remains ***,experimental models of neuromyelitis optica spectrum disorders are essential for exploring its pathogenesis and in screening for therapeutic *** most patients with neuromyelitis optica spectrum disorders are seropositive for IgG autoantibodies against aquaporin-4,which is highly expressed on the membrane of astrocyte endfeet,most current experimental models are based on aquaporin-4-IgG that initially targets *** experimental models have successfully simulated many pathological features of neuromyelitis optica spectrum disorders,such as aquaporin-4 loss,astrocytopathy,granulocyte and macrophage infiltration,complement activation,demyelination,and neuronal loss;however,they do not fully capture the pathological process of human neuromyelitis optica spectrum *** this review,we summarize the currently known pathogenic mechanisms and the development of associated experimental models in vitro,ex vivo,and in vivo for neuromyelitis optica spectrum disorders,suggest potential pathogenic mechanisms for further investigation,and provide guidance on experimental model *** addition,this review summarizes the latest information on pathologies and therapies for neuromyelitis optica spectrum disorders based on experimental models of aquaporin-4-IgG-seropositive neuromyelitis optica spectrum disorders,offering further therapeutic targets and a theoretical basis for clinical trials.

BACKGROUND There is a lack of clinical evidence on the efficacy and safety of transitioning from a thrice-daily pre-mixed insulin or basal-prandial regimen to insulin degludec/aspart(IDegAsp)therapy,with insufficient data from the Chinese *** To demonstrate the efficacy,safety,and treatment satisfaction associated with the transition to IDegAsp in type 2 diabetes mellitus(T2DM).METHODS In this 12-week open-label,non-randomized,single-center,pilot study,patients with T2DM receiving thrice-daily insulin or intensive insulin treatment were transitioned to twice-daily injections of insulin *** doses,hemoglobin A1c(HbA1c)levels,fasting blood glucose(FBG),hypoglycemic events,a Diabetes Treatment Satisfaction Questionnaire,and other parameters were assessed at baseline and *** This study included 21 participants.A marked enhancement was observed in the FBG level(P=0.02),daily total insulin dose(P=0.03),and overall diabetes treatment satisfaction(P<0.01)in the participants who switched to *** was a decrease in HbA1c levels(7.6±1.1 vs 7.4±0.9,P=0.31)and the frequency of hypoglycemic events of those who switched to IDegAsp decreased,however,there was no statistically significant *** The present findings suggest that treatment with IDegAsp enhances clinical outcomes,particularly FBG levels,daily cumulative insulin dose,and overall satisfaction with diabetes treatment.

Bullous pemphigoid associated with acquired hemophilia A: A case report

BACKGROUND Acquired hemophilia A (AHA) is a rare and potentially severe bleeding disordercaused by circulating autoantibodies against factor Ⅷ (FⅧ). In approximately50% of the patients, the condition is associated with autoimmune diseases,cancers, medication use, pregnancy, and the post-partum period. Bullous pemphigoid(BP) is a chronic autoimmune subepidermal blistering disease associatedwith tissue-bound and circulating autoantibodies against BP antigens 180 (BP180)and 230 (BP230). AHA-associated BP has a high mortality rate;hence, the understandingof this disease must *** SUMMARY A 69-year-old man presented with erythema, blisters, blood blisters, and crustsaccompanied by severe pruritus for more than 20 days, and ecchymosis andswelling on his left upper arm for 3 days. Pathological examination revealed asubepidermal blister that contained eosinophils. Laboratory tests showed that theBP180 autoantibody levels had increased, isolated activated partial thromboplastintime was notably prolonged (115.6 s), and coagulation FⅧ activity wasextremely low (< 1.0%). Furthermore, the FⅧ inhibitor titer had greatlyincreased (59.2 Bethesda units). Therefore, the patient was diagnosed as having BP associated with AHA, prescribed 0.05% topical halometasone cream, and transferred to a higher-level hospitalfor effective treatment;however, he died after 2 *** AHA associated BP is rare, dangerous, and has a high mortality rate. Therefore, its timely diagnosis and effectivetreatment are necessary.

The Gamma-Dirichlet algebra corresponds to the decomposition of the gamma process into the independent product of a gamma random variable and a Dirichlet process. This structure allows us to study the properties of the Dirichlet process through the gamma process and vice versa. In this article, we begin with a brief survey of several existing results concerning this structure. New results are then obtained for the large deviations of the jump sizes of the gamma process and the quasi-invariance of the two-parameter Poisson-Dirichlet distribution. We finish the paper with the derivation of the transition function of the Fleming-Viot process with parent independent mutation from the transition function of the measure-valued branching diffusion with immigration by exploring the Gamma-Dirichlet algebra embedded in these processes. A Comparison of Corrosion Behavior in Saline Environment: Rare Earth Metals (Y, Nd, Gd, Dy) for Alloying of Biodegradable Magnesium Alloys

Rare earth （RE） metals are widely used as the alloying implants. However, corrosion behavior of pure RE elements in biodegradable magnesium alloys as medical metals not only in physiological media but also in chlorinated saline environment is not well understood. In the present work, the RE metals Y, Nd, Gd and Dy are selected to investigate their corrosion behavior in 0.1 mol/L NaCI solution with immersion and electrochemistry techniques. As indicated, corrosion of the currently investigated RE metals is promoted in the order of Dy, Y, Gd and Nd. In terms of electrochemical response, such a sequence correlates with the increased impedance and the decreased corrosion rate （CR）. These RE metals manifest weak ability for passivation in the native surface. Then, reaction with aqueous solution easily happens through the anodic dissolution and cathodic hydrogen evolution. The corrosion products, RE（OH）3, adhered on the surface of RE metals, do not have an appreciable power to resist the reaction proceeding with corrosive chloride ions. In contrast to pure Mg, the RE metals, including Y, Nd, Gd and Dy, exhibit significantly fragile corrosion resistance in saline media. Therefore, with the curr correlation of corrosion resistance between RE-contain ent findings, it is impossible to reveal a well-defined ng Mg alloy and RE metal itself
