Objectives: To determine if the use of high frequency linear transducer (HFLT) in addition to conventional curvilinear transducer (CCT) aids assessment of fetal cardiac and non-cardiac anatomy in the first trimester. Methods: Transabdominal CCT (4 - 8 MHz) followed by HFLT (9 MHz) was used to study prospectively the visualization rate of basic and optional anatomical structures according to international guidelines. McNemar and Chi-square test were used to compare correlated and independent proportions respectively. Postnatal outcomes were traced. Results: Comparing with CCT alone, additional use of HFLT did not improve the completion rate of basic anomaly screen (95.0% vs. 97.0%, p = 0.5) in the 101 women studied, but it improved the visualization rate of some optional structures including lens (57.4% vs. 73.3%), three-vessel view (3 VV) (17.8% vs. 48.5%), left ventricular outflow tract (17.8% vs. 51.5%), kidneys (8.9% vs. 47.5%), and umbilical artery (86.1% vs. 93.1%) (all p < 0.05). Favourable fetal position was associated with a better visualisation of four-chamber view and 3VV (p < 0.05). Conclusions: Our findings supported that the use of HFLT in addition to CCT does not improve the completion of basic anomaly screen, nor does it achieve a high visualisation rate of different cardiac views in the first trimester.

Mass spectrometry(MS)-based omics technologies are now widely used to profile small molecules in multiple matrices to confer comprehensive snapshots of cellular metabolic *** metabolomes of cells,tissues,and organisms comprise a variety of molecules including lipids,amino acids,sugars,organic acids,and so *** mainly focus on the hydrophilic classes,while lipidomics has emerged as an independent omics owing to the complexities of the organismal *** potential roles of lipids and small metabolites in disease pathogenesis have been widely investigated in various human diseases,but system-level understanding is largely lacking,which could be partly attributed to the insufficiency in terms of metabolite coverage and quantitation accuracy in current analytical *** scientists are continuously striving to develop high-coverage omics approaches,integration of metabolomics and lipidomics is becoming an emerging approach to mechanistic *** of metabolome and lipidome offers a complete atlas of the metabolic landscape,enabling comprehensive network analysis to identify critical metabolic drivers in disease pathology,facilitating the study of interconnection between lipids and other metabolites in disease *** this review,we summarize omics-based findings on the roles of lipids and metabolites in the pathogenesis of selected major diseases threatening public *** also discuss the advantages of integrating lipidomics and metabolomics for in-depth understanding of molecular mechanism in disease pathogenesis.

Ferroptosis,a regulated and iron-dependent form of cell death characterized by peroxidation of membrane phospholipids,has tremendous potential for the therapy of human *** causal link between phospholipid homeostasis and ferroptosis is incompletely ***,we reveal that spin-4,a previously identified regulator of the“B12-one-carbon cycle-phosphatidylcholine(PC)”pathway,sustains germline development and fertility by ensuring PC sufficiency in the nematode Caenorhabditis ***,SPIN-4 regulates lysosomal activity which is required for B12-associated PC *** deficiency-induced sterility can be rescued by reducing the levels of polyunsaturated fatty acids,reactive oxygen species,and redox-active iron,which indicates that the sterility is mediated by germline *** results highlight the critical role of PC homeostasis in ferroptosis susceptibility and offer a new target for pharmacological approaches.

With the explosive growth of surveillance video data,browsing videos quickly and effectively has become an urgent *** key frame extraction has received widespread attention as an effective ***,accurately capturing the local motion state changes of moving objects in the video is still challenging in key frame *** target center offset can reflect the change of its motion *** observation proposed a novel key frame extraction method based on moving objects center offset in this *** proposed method utilizes the center offset to obtain the global and local motion state information of moving objects,and meanwhile,selects the video frame where the center offset curve changes suddenly as the key *** processing effectively overcomes the inaccuracy of traditional key frame extraction ***,extracting the center point of each ***,calculating the center point offset of each frame and forming the center offset curve by connecting the center offset of each ***,extracting candidate key frames and optimizing them to generate final key *** experimental results demonstrate that the proposed method outperforms contrast methods to capturing the local motion state changes of moving objects.

Lipidomics, which targets at the construction of a comprehensive map of lipidome comprising the entire lipid pool within a cell or tissue, is currently emerging as an independent discipline at the interface of lipid biology, technology and medicine. The diversity and complexity of the biological lipidomes call for technical innovation and improvement to meet the needs of various biomedical studies. The recent wave of expansion in the field of lipidomic research is mainly attributed to advances in analytical technologies, in particular, the development of new mass spectrometric and chromatographic tools for the characterization and quantification of the wide array of diverse lipid species in the cellular lipidome. Here, we review some of the key technical advances in lipidome analysis and put forth the applications of lipidomics in addressing the biological roles of lipids in numerous disease models including the metabolic syndrome, neurodegenerative diseases and infectious diseases, as well as the increasing urgency to construct the lipidome inventory for various mammalian/organism models useful for biomedical research.

Lipidome Atlas of the Developing Heart Uncovers Dynamic Membrane Lipid Attributes Underlying Cardiac Structural and Metabolic Maturation

Precise metabolic rewiring during heart organogenesis underlies normal cardiac ***,we utilized high-coverage,quantitative lipidomic approaches to construct lipidomic atlases of whole hearts(861 lipids;31 classes)and mitochondria(587 lipids;27 classes)across prenatal and postnatal developmental stages in *** uncovered the progressive formation of docosahexaenoyl-phospholipids and enhanced remodeling of C18:2,C20:3,and C20:4 fatty acyl moieties into cardiolipins as cardiac development progresses.A preferential flow of ceramides toward sphingomyelin biosynthesis over complex glycosphingolipid formation was also *** maSigPro and GPclust algorithms,we identified a repertoire of 448 developmentally dynamic lipids and mapped their expression patterns to a library of 550 biologically relevant developmentally dynamic *** combinatorial transcriptomics and lipidomics approaches identified Hadha,Lclat1,and Lpcat3 as candidate molecular drivers governing the dynamic remodeling of cardiolipins and phospholipids,respectively,in heart *** analyses revealed that postnatal cardiolipin remodeling in the heart constitutes a biphasic process,which first accumulates polyunsaturated C78-cardiolipins prior to tetralinoleoyl cardiolipin forming the predominant *** analyses supplemented with transmission electron microscopy imaging uncovered enhanced mitochondria–lipid droplet contacts mediated by *** combinatorial analyses of multiomics data uncovered an association between mitochondrial-resident,docosahexaenoic acid-phospholipids and messenger RNA levels of proton-transporting adenosine triphosphate synthases on inner mitochondrial membranes,which adds credence to the membrane pacemaker theory of *** current findings offer lipid-centric biological insights potentially important to understanding the molecular basis of cardiac metabolic flexibility and disease pathology.

Pulmonary surfactant is a lipid-protein complex secreted by alveolar typeⅡepithelial cells and is essential for the maintenance of the delicate structure of mammalian alveoli to promote efficient gas exchange across the air-liquid *** Golgi apparatus plays an important role in pulmonary surfactant modification and secretory ***,the physiological function of the Golgi apparatus in the transport of pulmonary surfactants is *** the present study,deletion of GM130,which encodes for a matrix protein of the cis-Golgi cisternae,was shown to induce the disruption of the Golgi structure leading to impaired secretion of lung surfactant proteins and ***,the results of in vitro and in vivo analysis indicated that the loss of GM130 resulted in trapping of Sftpa in the endoplasmic reticulum,Sftpb and Sftpc accumulation in the Golgi apparatus,and an increase in the compensatory secretion of ***,global and epithelial-specific GM130 knockout in mice resulted in an enlargement of alveolar airspace and an increase in alveolar epithelial autophagy;however,surfactant repletion partially rescued the enlarged airspace defects in GM130-deficient ***,our results demonstrate that GM130 and the mammalian Golgi apparatus play a critical role in the control of surfactant protein secretion in pulmonary epithelial cells.
