Despite the precise controllability of droplet samples in digital microfluidic(DMF)systems,their capability in isolating single cells for long-time culture is still limited:typically,only a few cells can be captured on an electrode.Although fabricating small-sized hydrophilic micropatches on an electrode aids single-cell capture,the actuation voltage for droplet transportation has to be significantly raised,resulting in a shorter lifetime for the DMF chip and a larger risk of damaging the cells.In this work,a DMF system with 3D microstructures engineered on-chip is proposed to form semi-closed micro-wells for efficient single-cell isolation and long-time culture.Our optimum results showed that approximately 20%of the micro-wells over a 30×30 array were occupied by isolated single cells.In addition,low-evaporation-temperature oil and surfactant aided the system in achieving a low droplet actuation voltage of 36V,which was 4 times lower than the typical 150 V,minimizing the potential damage to the cells in the droplets and to the DMF chip.To exemplify the technological advances,drug sensitivity tests were run in our DMF system to investigate the cell response of breast cancer cells(MDA-MB-231)and breast normal cells(MCF-10A)to a widely used chemotherapeutic drug,Cisplatin(Cis).The results on-chip were consistent with those screened in conventional 96-well plates.This novel,simple and robust single-cell trapping method has great potential in biological research at the single cell level.

【目的】利用拐点法量化公园的降温效应,并确定具有高效降温效应的公园面积和周长阈值范围,基于多源数据综合考虑城市公园的内部和外部特征对其降温效应的影响。【方法】基于2019年9月13日Landsat-8遥感影像,采用辐射传输法反演地表温度;基于高分二号遥感影像,采用随机森林法进行土地覆被分类;基于百度地图兴趣面(area of interest,AOI)获取83个公园边界。利用拐点法得到公园的降温强度和降温距离,利用相关性分析和SHAP分析,从公园内景观特征、公园内环境特征和公园外环境特征3个方面探讨影响因子与公园降温效应之间的关系。【结果】(1)公园的降温面积阈值为18~19 hm^(2),周长阈值在1.9~2.0 km。(2)公园内部的建设用地高占比和较高的斑块密度会削弱公园降温效应,而水体和绿地的占比则可增强公园降温效应;面积加权周长比越小的公园越有助于公园降温效应的发挥。(3)公园内植被结构参数和生理特征对降温效应有显著影响,植被冠层越高、长势越好,降温距离越远;土壤含水量对降温效应至关重要。(4)公园外部较高的建筑高度和高道路密度会削弱公园降温效应。【结论】公园的降温效应受到公园内外景观特征和环境特征的影响。可以通过提高公园景观的完整性、合理配置绿地水体和培育优质植被等措施来改善公园降温效应,从而缓解城市热岛效应。

目的探讨劈离式供肝儿童肝移植门静脉并发症的诊断及治疗策略。方法回顾性分析接受劈离式肝移植的88例儿童受者的临床资料。术中根据受者门静脉内径、发育情况,利用门静脉左右分支处进行吻合或间置供者髂静脉搭桥吻合,围手术期采用规范化的门静脉血流监测,术后按肝素钠桥接华法林的方案进行抗凝治疗。经增强CT或门静脉造影确诊门静脉狭窄或血栓形成后,予切开取栓、全身抗凝、介入下取栓、球囊扩张和(或)支架置入等处理。结果88例受者中共10例患儿确诊门静脉并发症,其中4例门静脉狭窄,确诊时间分别为术后1 d、2个月、8个月、11个月,6例门静脉血栓形成,确诊时间分别为术中、术后2 d、术后3 d(2例)、术后6 d、术后11个月。1例门静脉狭窄者和1例门静脉血栓形成者于围手术期死亡,门静脉并发症相关病死率为2%(2/88)。其余8例患者中,1例行全身抗凝治疗、2例行门静脉切开取栓术、1例行介入下球囊扩张、4例行介入下球囊扩张及支架置入,术后均长期随访,未再出现门静脉相关症状,复查门静脉血流参数正常。结论规范化的术中及术后门静脉血流监测有助于早期发现门静脉并发症,及时采取术中门静脉切开取栓,术后介入下球囊扩张、支架置入等手段可有效治疗门静脉并发症,减少门静脉并发症导致的移植物丢失和受者死亡。

In order to obtain stable and high-precision motion tracking in mixed reality(MR)simulation of the complex system,an optical-inertial hybrid tracking method is proposed,which combines the accuracy of optical motion capture equipment and the robustness of inertial motion capture equipment,and can reduce the occlusion problem of entity scene on the premise of ensuring the localization accuracy.The method has been applied to the ergonomic evaluation of large civil aircraft development processes,and the effectiveness and usability of the method have been verified.

The purpose of this study is to reduce the uncertainty in the calculation process on hesitant fuzzy sets(HFSs).The innovation of this study is to unify the cardinal numbers of hesitant fuzzy elements(HFEs)in a special way.Firstly,a probability density function is assigned for any given HFE.Thereafter,equal-probability transformation is introduced to transform HFEs with different cardinal numbers on the condition into the same probability density function.The characteristic of this transformation is that the higher the consistency of the membership degrees in HFEs,the higher the credibility of the mentioned membership degrees is,then,the bigger the probability density values for them are.According to this transformation technique,a set of novel distance measures on HFSs is provided.Finally,an illustrative example of intersection traffic control is introduced to show the usefulness of the given distance measures.The example also shows that this study is a good complement to operation theories on HFSs.

In order to analyze the proposed method,this paper takes the cockpit as the complex system,since the traditional cockpit design evaluation method can set up an actual engineering prototype.Although this method can realize the accurate evaluation of the internal structure layout,control devices,and interface display of the cockpit,it is dif-ficult to make modifications once the production is completed,which is not suitable for multi-scheme comparison and rapid iteration in the early stage of civil aircraft design.In this regard,this paper establishes the civil aircraft cockpit physical environment and the corresponding virtual cockpit environment based on the virtual-real matching tech-nology,then proposes a mixed reality simulation evaluation method so as to realize the real-time ergonomic evaluation of the digital human body followed by a real person.Experimental results show that this mixed reality simulation evaluation method can effectively improve the evaluation accuracy of cockpit design,ensure the efficiency of multi-scheme comparison and optimization iteration in the early stage of cockpit design.

Health assessment of plantations based on LiDAR canopy spatial structure parameters

The Yellow River Delta(YRD)has China's largest artificial Robinia pseudoacacia forest,which was planted in the late 1970s and suffered extensive dieback in the 1990s.The health grade of the R.pseudoacacia forest(named canopy vigor grade,CVG)could be achieved by using high-resolution images and canopy vigor indicators(CVIs).However,a previous study showed that there was no significant correlation between CVG and the field-estimated aboveground biomass(AGB)of R.pseudoacacia forest.Therefore,this study aims to construct forest health indicators(FHIs)based on canopy spatial structure parameters extracted from LiDAR.The FHIs included Weibull_α(the scale parameter of the Weibull density function that reflects the shape of the tree canopy),VCI(vertical complexity index),sdCC(the standard deviation of canopy cover),H99(the 99th percentile height)and cvLAD(the coefficient of variation of leaf area density),and could significantly distinguish three forest health grades(FHG)(p<0.05).The FHG was positively correlated with forest AGB(rs=0.51,p=0.004),and the similarity value with CVG was 63.33%.The results of this study confirmed that the FHIs can reflect both canopy vigor and tree productivity,and distinguish forest health status without prior classification information.

With the dawn of organ donation after a citizen's death in China,the use of split-liver transplantation(SLT)can effectively increase the source of donor liver,reduce the waiting time for organ transplantation in patients,and particularly solve the problem of organ shortage in children.In recent years,many transplantation centers have been performing SLT to varying degrees and efficacy.At the current stage,the experiences of countries with advanced transplantation techniques should be used to establish an SLT consensus that is suitable for China to further increase the ratio and efficacy of SLT.In this paper,we combined expert experiences to generate an SLT expert consensus that included donor and donor liver evaluation,recipient selection criteria,donor and recipient matching,selection of splitting form and tools,blood vessels and bile ducts dissection and allocation,perioperative management of SLT,and organ allocation.

A patient with steroid-resistant acute rejection 50 days after ABO-compatible orthotopic liver transplantation(LT)received regular infusion of allogeneic mesenchymal stem cells(MSCs)after three sessions of steroid pulse therapy which failed to control the pathogenetic condition as shown by biopsy.Liver function improved gradually after intravenous injection of MSCs once weekly for 10 weeks(as confirmed by biopsy)and remained stable under administration of conventional immunosuppressive agents.There was no evidence of neoplasms 5 years after treatment.MSCs infusion appears to successfully reverse resistance to immunosuppressive agents and may be a useful treatment for post-liver transplant steroid-resistant rejection.
