Structural variants(SVs),such as deletions(DELs)and insertions(INSs),contribute substantially to pig genetic diversity and phenotypic variation.Using a library of SVs discovered from long-read primary assemblies and short-read sequenced genomes,we map pig genomic SVs with a graph-based method for re-genotyping SVs in 402 genomes.Our results demonstrate that those SVs harboring specific trait-associated genes may greatly shape pig domestication and local adaptation.Further characterization of SVs reveals that some population-stratified SVs may alter the transcription of genes by affecting regulatory elements.We identify that the genotypes of two DELs(296-bp DEL,chr7:52,172,101e52,172,397;278-bp DEL,chr18:23,840,143 e23,840,421)located in muscle-specific enhancers are associated with the expression of target genes related to meat quality(FSD2)and muscle fiber hypertrophy(LMOD2 and WASL)in pigs.Our results highlight the role of SVs in domestic porcine evolution,and the identified candidate functional genes and SVs are valuable resources for future genomic research and breeding programs in pigs.

Lung cancer is a malignant tumor with the highest incidence and mortality in China,which seriously affects the quality of life of patients.Traditional Chinese medicine(TCM)plays a unique advantage in the treatment and rehabilitation of lung cancer.In order to standardize TCM diagnosis and treatment and disease management of clinical medical workers,the Chinese Association of Integrative Medicine issued the“Expert Consensus on the Diagnosis and Treatment of Lung Cancer by Integrated Traditional Chinese and Western Medicine”in August 2021.Experts’recommendations were made on the clinical staging and syndrome differentiation and classification of lung cancer,characteristic TCM therapies,and nursing care of integrated traditional Chinese and Western medicine.This article interpreted the Consensus on the above aspects from a nursing perspective,hoping to provide references for clinical medical workers in the treatment and nursing care of lung cancer.

Background Growth differentiation factor-15(GDF-15)is a stress response protein and is related to cardiovascular diseases(CVD).This study aimed to investigate the association between GDF-15 and pre-eclampsia(PE).Method The study involved 299 pregnant women,out of which 236 had normal pregnancies,while 63 participants had PE.Maternal serum levels of GDF-15 were measured by using enzyme-linked immunosorbent assay kits and then translated into multiple of median(MOM)to avoid the influence of gestational week at blood sampling.Logistic models were performed to estimate the association between GDF-15 MOM and PE,presenting as odd ratios(ORs)and 95%confidence intervals(CIs).Results MOM of GDF-15 in PE participants was higher compared with controls(1.588 vs.1.000,p1)had a 4.74-fold(95%CI=2.23–10.08,p<0.001)increased risk of PE,adjusted by age,preconceptional body mass index,gravidity,and parity.Conclusions These results demonstrated that higher levels of serum GDF-15 were associated with PE.GDF-15 may serve as a biomarker for diagnosing PE.

Background Hepatic inflammation is the major risk factor of hepatocellular carcinoma(HCC).However,the underlying mechanism by which hepatic inflammation progresses to HCC is poorly understood.This study was designed to investigate the role of ETS translocation variant 4(ETV4)in linking hepatic inflammation to HCC.Methods Quantitative real-time PCR and immunoblotting were used to detect the expression of ETV4 in HCC tissues and cell lines.RNA sequencing and luciferase reporter assays were performed to identify the target genes of ETV4.Hepatocyte-specific ETV4-knockout(ETV4fl/fl,alb-cre)and transgenic(ETV4Hep-TG)mice and diethylnitrosamine-carbon tetrachloride(DEN-CCL4)treatment experiments were applied to investigate the function of ETV4 in vivo.The Cancer Genome Atlas(TCGA)database mining and pathological analysis were carried out to determine the correlation of ETV4 with tumor necrosis factor-alpha(TNF-α)and mitogen-activated protein kinase 11(MAPK11).Results We revealed that ETV4 was highly expressed in HCC.High levels of ETV4 predicted a poor survival rate of HCC patients.Then we identified ETV4 as a transcription activator of TNF-αand MAPK11.ETV4 was positively correlated with TNF-αand MAPK11 in HCC patients.As expected,an increase in hepatic TNF-αsecretion and macrophage accumulation were observed in the livers of ETV4Hep-TG mice.The protein levels of TNF-α,MAPK11,and CD68 were significantly higher in the livers of ETV4Hep-TG mice compared with wild type mice but lower in ETV4fl/fl,alb-cre mice compared with ETV4fl/fl mice as treated with DEN-CCL4,indicating that ETV4 functioned as a driver of TNF-α/MAPK11 expression and macrophage accumulation during hepatic inflammation.Hepatocyte-specific knockout of ETV4 significantly prevented development of DEN-CCL4-induced HCC,while transgenic expression of ETV4 promoted growth of HCC.Conclusions ETV4 promoted hepatic inflammation and HCC by activating transcription of TNF-αand MAPK11.Both the ETV4/TNF-αand ETV4/MAPK11 axes represented two potential therapeutic target

The stability of pump-turbines is of great importance to the operation of pumped storage power (PSP) stations. Both hydraulic instabilities and operational instabilities have been reported in PSP stations in China. In order to provide a reference to the engineers and scientists working on pump-turbines, this paper summarizes the hydraulic instabilities and performance characteristics that promote the operational instabilities encountered in pump-turbine operations in China. Definitions, analytical methods, numerical and experimental studies, and main results are clarified. Precautions and countermeasures are also provided based on a literature review. The gaps between present studies and the need for engineering practice are pointed out.

Phase equilibria in the Al-rich corner of the Al-Er-Y ternary system from 65 at.% to 100 at.% Al at 673 and873 K were investigated,and the two isothermal sections were established by X-ray diffraction(XRD),scanning electron microscope(SEM) and electron probe micro analysis(EPMA).Three ternary-phase regions are observed and the phase relations are similar at both isothermal sections.There are two ternary compounds,and a complete crystal structures and composition ranges of the two ternary compounds are determined as τ_(1)(Al_(75-76)Er_(15-22)Y_(2-10),R-3 m) and τ_(2)(Al_(75-76)Er_(11-17)Y_(7-14),P6_(3)/mmc) via XRD,focused ion beam coupled with transmission electron microscope(FIB-TEM) and EPMA.Meanwhile,the solubilities of the third element in the binary compounds at both temperatures are measured.The solubility of Er inβ-Al_(3) Y phase reaches to 6.6 at.% and that of Y in Al_(3) Er phase reaches to 1.5 at.% after annealing at 873 K.The temperature effect is not obvious for the phase equilibria at 673 and 873 K,but it increases the solubility of Er in β-Al_(3) Y,while it is not obvious for both the solubility of Y in Al_(3) Er and that of Er and Y in(Al).The phase equilibria of the Al-Er-Y ternary system determined by the present work have provided new phase diagram data for a future thermodynamic assessment of this system.

Gastric cancer is a common and highly malignant tumor of the digestive system,which seriously threatens human health.The benefit of surgical treatment is limited.The effect of radiotherapy and chemotherapy is not significant,and the toxic and side effects are large.Although the diagnosis and treatment modes of molecular targeting and multidisciplinary cooperation have also been further developed and applied,a relatively unified comprehensive treatment standard scheme has not been formed.Traditional Chinese medicine(TCM),as complementary and alternative medicine,has unique advantages in the development of diagnosis and treatment of diseases in China.Therefore,the Chinese Association of Integrative Medicine formed“Expert Consensus for Diagnosis and Treatment of Gastric Cancer with Integrated Traditional Chinese and Western Medicine”on the basis of clinical practice and experience.In this article,the consensus is interpreted from the perspective of nursing,focusing on the pathogenesis,high‑risk factors,characteristic therapies of TCM,and nursing of integrated traditional Chinese and Western medicine for gastric cancer,to provide a reference for front‑line medical personnel in the roots in the future nursing care of gastric cancer.

As one of the most common malignant tumors in the world,colorectal cancer(CRC)is accompanied by a variety of symptoms in the process of disease development and treatment,which seriously affect the functional state and quality of life of patients.In China,comprehensive intervention measures of traditional Chinese medicine(TCM)play an indispensable role in the treatment and rehabilitation of CRC.In order to further standardize and improve the treatment and management of CRC by medical staff,the Chinese Association of Integrative Medicine issued the“Expert Consensus for Diagnosis and Treatment of Colorectal Cancer with Integrated Traditional Chinese and Western Medicine”in August 2021.This paper interprets this consensus in detail from the perspective of nursing,focusing on the common understanding of the pathogenesis of CRC,TCM diagnosis and syndrome differentiation,integrated traditional Chinese and Western medicine nursing,TCM characteristic therapies,etc.,with a view to deepening the understanding and practical application of the consensus among clinical doctors and nurses and improving the level of diagnosis,treatment,and nursing of CRC.

Viscous liquid sloshing damping in cylindrical container using a volume of fluid method

Liquid sloshing is a kind of very complicated free surface flow and exists widely in many fields.In order to calculate liquid sloshing damping precisely a volume of fluid method based on finite volume scheme is used to simulate free surface flows in partly filled cylindrical containers.A numerical method is pre-sented to simulate the movement of the free surface flow,in which a piecewise linear interface con-struction scheme and an unsplit Lagrangian advection scheme instead of Eulerian advection scheme are used.The damping performance of liquid sloshing in cylindrical containers under fundamental sloshing mode is investigated.There are four factors determining the surface-wave damping:free surface,boundary-layer,interior fluid and contact line.In order to study different contributions from these four factors to whole damping,several examples are simulated.No-slip and slip wall boundary conditions on both side wall and bottom wall of the cylindrical containers are studied to compare with the published results obtained by solving Stokes equations.In the present method the first three main factors can be considered.The simulation results show that the boundary-layer damping contribution increases while the interior fluid damping contribution decreases with increase of Reynolds number.

Because the inhibition of Nogo proteins can promote neurite growth and nerve cell differentiation, a cell-scaffold complex seeded with Nogo receptor （NgR）-silenced neural stem cells and Schwann cells may be able to improve the microenvironment for spinal cord injury repair. Previous studies have found that mild hypothermia helps to attenuate secondary damage in the spinal cord and exerts a neuroprotective effect. Here, we constructed a cell-scaffold complex consisting of a poly（D,L-lactide-co-glycolic acid）（PLGA） scaffold seeded with NgR-silenced neural stem cells and Schwann cells, and determined the effects of mild hypothermia combined with the cell-scaffold complexes on the spinal cord hemi-transection injury in the T9 segment in rats. Compared with the PLGA group and the NgR-silencing cells ＋ PLGA group, hindlimb motor function and nerve electrophysiological function were dearly improved, pathological changes in the injured spinal cord were attenuated, and the number of surviving cells and nerve fibers were increased in the group treated with the NgR-silenced cell scaffold ＋ mild hypothermia at 34℃ for 6 hours. Furthermore, fewer pathological changes to the injured spinal cord and more surviving cells and nerve fibers were found after mild hypothermia therapy than in injuries not treated with mild hypothermia. These experimental results indicate that mild hypothermia combined with NgR gene-silenced cells in a PLGA scaffold may be an effective therapy for treating spinal cord injury.
