The nonreceptor tyrosine phosphatase SHP2(PTPN11)is a ubiquitously expressed molecule which regulatescellular proliferation,survival,migration,and differentiationduring *** of shp2 null embryos died shortly after implantation due to failed trophoblast ***,the genetic role of Shp2 during placentation is still largely *** the present study,we find that Shp2 is expressed by multiple trophoblast cell subtypes in the developing *** deletion of Shp2 in placental trophoblast results in reduced trophoblast invasion into the spiral arterioles and a poorly branched labyrinth,leading to substantial embryonic lethality at midgestation(11.5).Further studies showed that Shp2 acts via active Rho A signaling to regulate the migration and invasion of trophoblast giant cells(TGCs),which is required for infiltrating and remodeling the maternal blood *** in the labyrinth,Shp2 functions through Ras-MAPK signaling pathway to sustain the proliferation of labyrinth trophoblast progenitors(La TP),which ensuring Syn T terminal differentiation and placental exchange interface *** sum,these results demonstrate that Shp2 is essential for placental development through regulating TGC invasion and sustaining La TP proliferation in a cell-type specific manner.
