Edaravone-loaded poly(amino acid) nanogel inhibits ferroptosis for neuroprotection in cerebral ischemia injury

Neurological injury caused by ischemic stroke is a major cause of permanent disability and death. The currently available neuroprotective drugs fail to achieve desired therapeutic efficacy mainly due to short circulation half-life and poor blood−brain barrier (BBB) permeability. For that, an edaravone-loaded pH/glutathione (pH/GSH) dual-responsive poly(amino acid) nanogel (NG/EDA) was developed to improve the neuroprotection of EDA. The nanogel was triggered by acidic and EDA-induced high-level GSH microenvironments, which enabled the selective and sustained release of EDA at the site of ischemic injury. NG/EDA exhibited a uniform sub-spherical morphology with a mean hydrodynamic diameter of 112.3 ± 8.2 nm. NG/EDA efficiently accumulated at the cerebral ischemic injury site of permanent middle cerebral artery occlusion (pMCAO) mice, showing an efficient BBB crossing feature. Notably, NG/EDA with 50 µM EDA significantly increased neuron survival (29.3%) following oxygen and glucose deprivation by inhibiting ferroptosis. in addition, administering NG/EDA for 7 d significantly reduced infarct volume to 22.2% ± 7.2% and decreased neurobehavioral scores from 9.0 ± 0.6 to 2.0 ± 0.8. Such a pH/GSH dual-responsive nanoplatform might provide a unique and promising modality for neuroprotection in ischemic stroke and other central nervous system diseases.

To the Editor:Copper is a crucial trace element in the body that plays a key role in metabolism and biological *** ions are found in two forms,Cu^(1+)and Cu^(2+),of which Cu^(1+)is the main form stored in *** studies have shown that intracellular copper accumulation can cause oxidative stress and disrupt cellular function.[1]Different from apoptosis,autophagy,pyroptosis,and ferroptosis,cuproptosis is a newly discovered type of cell death in 2022.[2]it is mediated by protein lipoylation with ferredoxin 1(FDX1)in *** most common pathological type of kidney cancer in adults is clear cell renal cell carcinoma(ccRCC),which accounts for 70-80%of all cases of renal cell carcinoma(RCC).[3]Currently,surgical resection is the preferred treatment for RCC,as radiotherapy,chemotherapy,and immunotherapy have shown limited effectiveness in treating ***,cuproptosis induction may offer a new approach for ccRCC treatment.

Multimorbidity and mortality among older patients with coronary heart disease in Shenzhen,China

BACKGROUND The current understanding of the magnitude and consequences of multimorbidity in Chinese older adults with coronary heart disease(CHD)is *** aimed to assess the association and population-attributable fractions(PAFs)between multimorbidity and mortality among hospitalized older patients who were diagnosed with CHD in Shenzhen,*** We conducted a retrospective cohort study of older Chinese patients(aged≥65 years)who were diagnosed with *** proportional hazards models were used to estimate the associations between multimorbidity and all-cause and cardiovascular disease(CVD)*** also calculated the *** The study comprised 76,455 older hospitalized patients who were diagnosed with CHD between January 1,2016,and August 31,*** them,70,217(91.9%)had multimorbidity,defined as the presence of at least one of the predefined 14 chronic *** with cancer,hemorrhagic stroke and chronic liver disease had the worst overall death risk,with adjusted HRs(95%Cis)of 4.05(3.77,4.38),2.22(1.94,2.53),and 1.85(1.63,2.11),*** CVD mortality,the highest risk was observed for hemorrhagic stroke,ischemic stroke,and chronic kidney disease;the corresponding adjusted HRs(95%Cis)were 3.24(2.77,3.79),1.91(1.79,2.04),and 1.81(1.64,1.99),***-cause mortality was mostly attributable to cancer,heart failure and ischemic stroke,with PAFs of 11.8,10.2,and 9.1,*** for CVD mortality,the leading PAFs were heart failure,ischemic stroke and diabetes;the corresponding PAFs were 18.0,15.7,and 6.1,*** Multimorbidity was common and had a significant impact on mortality among older patients with CHD in Shenzhen,***,heart failure,ischemic stroke and diabetes are the primary contributors to ***,prioritizing improved treatment and management of these comorbidities is essential for the survival prognosis of CHD patients from a holistic public health perspective.

A role for Retinoblastoma 1 in hindbrain morphogenesis by regulating GBX family

Nitrogen deposition mediates more stochastic processes in structuring plant community than soil microbial community in the Eurasian steppe

Anthropogenic environmental changes may affect community assembly through mediating both deterministic(e.g.,competitive exclusion and environmental filtering)and stochastic processes(e.g.,birth/death and dispersal/colonization).it is traditionally thought that environmental changes have a larger mediation effect on stochastic processes in structuring soil microbial community than aboveground plant community;however,this hypothesis remains largely *** we report an unexpected pattern that nitrogen(N)deposition has a larger mediation effect on stochastic processes in structuring plant community than soil microbial community(thosein diameter,including archaea,bacteria,fungi,and protists)in the Eurasian *** performed a ten-year nitrogen deposition experiment in a semiarid grassland ecosystem in inner Mongolia,manipulating nine rates(0–50 g N m^(-2)per year)at two frequencies(nitrogen added twice or 12 times per year)under two grassland management strategies(fencing or mowing).We separated the compositional variation of plant and soil microbial communities caused by each treatment into the deterministic and stochastic components with a recently-developed *** nitrogen addition rate increased,the relative importance of stochastic component of plant community first increased and then decreased,while that of soil microbial community first decreased and then *** the whole,the relative importance of stochastic component was significantly larger in plant community(0.552±0.035;mean±standard error)than in microbial community(0.427±0.035).Consistently,the proportion of compositional variation explained by the deterministic soil and community indices was smaller for plant community(0.172–0.186)than microbial community(0.240–0.767).Meanwhile,as nitrogen addition rate increased,the linkage between plant and microbial community composition first became weaker and then became *** larger stochasticity in plant community relative to microbial

Sex pheromones are considered to play critical roles in partner communication of most parasitic ***,the identification of sex pheromone components remains limited to a few families of parasitoid *** this study,we functionally characterized a candidate sex pheromone component in Microplitis mediator(Hymenoptera:Braconidae),a solitary parasitoid of Noctuidae *** found that the body surface extract from female wasps could significantly stimulate courtship behavior of *** chromatography-electroantennographic detection(GC-EAD)analysis revealed that a candidate semiochemical from extract triggered significant electrophysiological response of antennae of *** performing gas chromatography-mass spectrometer(GC-MS)measurement,GC-EAD active compound was identified as n-octyl acrylate,a candidate sex pheromone component in female *** electroantennogram(EAG)tests,antennae of male wasps showed significantly higher electrophysiological responses to n-octyl acrylate than those of females.Y-tube olfactometer assays indicated that male wasps significantly chose n-octyl acrylate compared with the ***,male wasps showed a remarkable preference for n-octyl acrylate in a simulated field condition behavioral trial;simultaneously,n-octyl acrylate standard could also trigger significant courtship behavior in *** propose that n-octyl acrylate,as a candidate vital sex pheromone component,could be utilized to design behavioral regulators of *** to implement the protection and utilization of natural enemies.

Molecular crowding electrolytes for stabilizing Zn metal anode in rechargeable aqueous batteries

A solid electrolyte interphase(SEi)with a robust mechanical property and a high ionic conductivity is imperative for high-performance zinc metal ***,it is difficult to form such a SEi directly from an *** this work,a molecular crowding effect is based on the introduction of Zn(OTF)_(2)and Zn(ClO_(4))_(2)to 2 mol/L ZnSO_(4)*** and experiments indicate that the Zn(OTF)_(2)and Zn(ClO_(4))_(2)not only create a molecularly crowded electrolyte environment to promote the interaction of Zn^(2+)and OTF^(-),but also participate in the reduction to construct a robust and high ionic-conductive SEi,thus promoting metal zinc deposition to the(002)crystal *** this molecular crowding electrolyte,a high current density of 1 mA/cm^(2)can be obtained by assembling symmetric batteries with Zn as the anode for over 1000 *** in a temperature environment of-10℃,a current density of 1 mA/cm^(2)can be obtained by assembling symmetric batteries with Zn for over 200 ***//Bi_(2)S_(3)/VS4@C cells achieve a CE rate of up to 99.81%over 1000 ***,the utilization of a molecular crowding electrolyte is deemed a highly effective approach to fabricating a sophisticated SEi for a zinc anode.

Hepatocellular carcinoma(HCC)is a highly heterogeneous tumor,with dynamic equilibrium and complex interplay between its intricate tumor nature and ambient tumor immune microenvironment(TiME).1 Elegant research has indicated that cancer stem cells,a small subset of neoplastic cells confined within dedicated niches,display stem cell-like properties and interact with cells in TiME,thereby imparting an indelible impact on stemness regulation,tumor heterogeneity,and cancer cell plasticity.2 Previous taxonomies solely from the perspective of stemness or TiME may introduce some degree of bias in the comprehension of HCC carcinogenesis,3,4 and thus it is of paramount importance to systematically consider tumor stemness and TiME as a whole to truly portray the biological landscape of HCC.

Achievement of a‘clinical cure’in chronic hepatitis B(CHB)implies sustained virological suppression and immunological control over the infection,which is the ideal treatment goal according to domestic and international CHB management *** practice has shown encouraging results for specific patient cohorts using tailored treatment *** regimens incorporate either nucleos(t)ide analogs,immunomodulatory agents such as pegylated interferonα,or a strategic combination of both,sequentially or concurrently *** these advancements in the clinical handling of hepatitis B,achieving a clinical cure remains elusive for a considerable subset of patients due to the number of challenges that preclude the realization of optimal treatment *** include,but are not limited to,the emergence of antiviral resistance,incomplete immune recovery,and the persistence of covalently closed circular ***,the variance in response to interferon therapy and the lack of definitive biomarkers for treatment cessation also contribute to the complexity of achieving a clinical *** article briefly overviews the current research progress and existing issues in pursuing a clinical cure for hepatitis B.

Facile identification of fluorosurfactant category in aqueous film-forming foam concentrates via optimized^(19)F NMR

Aqueous film-forming foams(AFFFs)are the primary source of toxic perfluoroalkyl and polyfluoroalkyl substances(PFAS)in ***,it is urgent to develop a facile and fast method for identifying fluorosurfactants in commercially available *** this work,fluorine nuclear magnetic resonance(^(19)F NMR)spectroscopy was optimized to measure AFFFs directly with the extra addition of 5%D_(2)O as the locking reagent,and high-quality spectra could be acquired within 4 min(0.1%fluorosurfactant content).Recovery experiments demonstrated that the use of different AFFFs had no marked influence on the quantitative analysis of *** method works with low-field NMR spectroscopy(1.4 T)as ***-dimensional(2D)^(19)F COSY NMR was used to make signal assignments for different fluorosurfactant *** optimized ^(19)F NMR could quantify the commercially available fluorosurfactants in different AFFFs,identify them being in either the perfluorooctane sulfonate(PFOS)or fluorotelomer sulfonic acid(FTS)categories,and distinguish the head-group of PFOS and FTS derivatives,which exhibits great potentials in the developments of relevant commercial detections.
