AIM: To investigate whether the major kavalactone kavain imposes adverse effects on the liver ultrastructure and function by affecting vascular and microvascular architecture and altering hepatocellular morphology. METHODS: Kavain solution (10 μg/mL or 43.5 μmol/L) was perfused for 2 h in isolated rat livers. After standard fi xation and tissue preparation,the samples were examined by scanning electron microscopy (SEM),transmission electron microscopy (TEM),and light microscopy (LM). RESULTS: LM,SEM,and TEM examinations indicated kavain-treated rat livers (n = 4) displayed severe vascular and endothelial damage compared to control livers (n = 4). CONCLUSION: The data so far support the hypothesis that kavain induces adverse effects on liver; additional investigations with other kavalactones and their effects on liver are urgently needed.

Spatiotemporal variations of ambient air pollutants and meteorological influences over typical urban agglomerations in China during the COVID-19 lockdown

To investigate the air quality change during the COVID-19 pandemic,we analyzed spatiotemporal variations of six criteria pollutants in nine typical urban agglomerations in China using ground-based data and examined meteorological influences through correlation analysis and backward trajectory analysis under different *** of PM2.5,PM10,NO2,SO2 and CO in urban agglomerations respectively decreased by 18%–45%(30%–62%),17%–53%(22%–39%),47%-64%(14%–41%),9%–34%(0%–53%)and 16%-52%(23%–56%)during Lockdown(Post-lockdown)period relative to Pre-lockdown ***2.5 pollution events occurred during Lockdown in Beijing-Tianjin-Hebe(BTH)and Middle and South Liaoning(MSL),and daily O3 concentration rose to gradeⅡstandard in Post-lockdown *** from the nationwide slump of NO2 during Lockdown period,a rebound(~40%)in Post-lockdown period was observed in Cheng-Yu(CY),Yangtze River Middle-Reach(YRMR),Yangtze River Delta(YRD)and Pearl River Delta(PRD).With slightly higher wind speed compared with 2019,the reduction of PM2.5(51%–62%)in Post-lockdown period is more than2019(15%–46%)in HC(Harbin-Changchun),MSL,BTH,CP(Central Plain)and SP(ShandongPeninsula),suggesting lockdown measures are effective to PM2.5 *** O3 concentrations generally increased during the lockdown,its increment rate declined compared with 2019 under similar sunlight duration and ***,unlike HC,MSL and BTH,which suffered from additional(>30%)air masses from surrounding areas after the lockdown,the polluted air masses reaching YRD and PRD mostly originated from the long-distance transport,highlighting the importance of joint regional governance.

Hydrogenated nanocrystalline silicon carbide （SIC） thin films were deposited on the single-crystal silicon substrate using the helicon wave plasma enhanced chemical vapor deposition （HW-PECVD） technique. The influences of magnetic field and hydrogen dilution ratio on the structures of SiC thin film were investigated with the atomic force microscopy （AFM）, the Fourier transform infrared absorption （FTIR） and the transmission electron microscopy （TEM）. The results indicate that the high plasma activity of the helicon wave mode proves to be a key factor to grow crystalline SiC thin films at a relative low substrate temperature. Also, the decrease in the grain sizes from the level of microcrystalline to that of nanocrystalline can be achieved by increasing the hydrogen dilution ratios. Transmission electron microscopy measurements reveal that the size of most nanocrystals in the film deposited under the higher hydrogen dilution ratios is smaller than the doubled Bohr radius of 3C-SiC （approximately 5.4 nm）, and the light emission measurements also show a strong blue photoluminescence at the room temperature, which is considered to be caused by the quantum confinement effect of small-sized SiC nanocrystals.

A 2D heterostructure with proximity coupling of magnetism and topology can provide enthralling prospects for hosting new quantum states and exotic properties that are relevant to next-generation spintronic ***,we synthesize a delicate van der Waals(vdW)heterostructure of CrTe_(2)/Bi_(2)Te_(3) at the atomic scale via molecular beam ***-temperature scanning tunneling microscopy/spectroscopy measurements are utilized to characterize the geometric and electronic properties of the CrTe_(2)/Bi_(2)Te_(3) heterostructure with a compressed vdW *** structural analysis reveals complex interfacial structures with diversiform step heights and intriguing moiré*** formation of the interface is ascribed to the embedded characteristics of CrTe_(2) and Bi_(2)Te_(3) by sharing Te atomic layer upon interfacing,showing intercoupled features of electronic structure for CrTe_(2) and Bi_(2)Te_(3).Our study demonstrates a possible approach to construct artificial heterostructures with different types of ordered states,which may be of use for achieving tunable interfacial Dzyaloshinsky–Moriya interactions and tailoring the functional building blocks in low dimensions.

Complete Pathway Elucidation and Heterologous Reconstitution of Rhodiola Salidroside Biosynthesis

Salidroside is a bioactive tyrosine-derived phenolic natural product found in medicinal plants under the Rhodiola genus. In addition to their anti-fatigue and anti-anoxia roles in traditional medicine, Rhodiola total extract and salidroside have also displayed medicinal properties as anti-cardiovascular diseases and anticancer agents. The resulting surge in global demand of Rhodiola plants and salidroside has driven some species close to extinction. Here, we report the full elucidation of the Rhodiola salidroside biosynthetic pathway utilizing the first comprehensive transcriptomics and metabolomics datasets for Rhodiola rosea. Unlike the previously proposed pathway involving separate decarboxylation and deamination enzymatic steps from tyrosine to the key intermediate 4-hydroxyphenylacetaldehyde （4-HPAA）, Rhodiola contains a pyridoxal phosphate-dependent 4-HPAA synthase that directly converts tyrosine to 4-HPAA. We further identified genes encoding the subsequent 4-HPAA reductase and tyrosohUDP-glucose 8-O-glucosyltransferase, respectively, to complete salidroside biosynthesis in Rhodiola. We show that heterologous production of salidroside can be achieved in the yeast Saccharomyces cerevisiae as well as the plant Nicotiana benthamiana through transgenic expression of Rhodiola salidroside biosynthetic genes. This study provides new tools for engineering sustainable production of salidroside in heterologous hosts.

Vertical Short Crack Initiation in Medium Carbon Bainitic Steel Under Mild Tractive Rolling Contact

To improve the current grinding procedure of the back-up roll of CVC hot rolling mills so that the back-up roll service life can be extended, the crack initiation and propagation behavior of medium carbon bainitic back-up roll steel was investigated, a kind of asperity-scale, surface originated vertical short cracks occurred at 5 × 10^2 -1 × 10^4 cycles. Theoretical analysis indicated that the maximum tensile stress occurring at the back edge of the contact of asperities keeps at above 1 347. 97 MPa, and ratcheting and cyclic plastic deformation take place at such sites within 1 × 10^4 cycles. The early initiation of the vertical short cracks is caused by the asperity contact. According to the crack initiation mechanism, short crack behavior and preventive grinding strategy, steel consumption can be reduced considerably by decreasing the surface roughness and removing the asperity influenced surface thin layer at about 70%-80% of the surface distress life.

Improvements in early screening,new diagnostic techniques,and surgical treatment have led to continuous downward trends in hepatocellular carcinoma(HCC)morbidity and mortality ***,high recurrence and refractory cancer after hepatectomy remain important factors affecting the long-term prognosis of *** clinical characteristics and prognosis of recurrent HCC are heterogeneous,and guidelines on treatment strategies for recurrent HCC are *** such as surgical resection,radiofrequency ablation,and transhepatic arterial chemoembolization are effective for tumors confined to the liver,and targeted therapy is a very important treatment for unresectable recurrent HCC with systemic *** the deepening of the understanding of the immune microenvironment of HCC,blocking immune checkpoints to enhance the antitumor immune response has become a new direction for the treatment of *** addition,improvements in the tumor immune microenvironment caused by local treatment may provide an opportunity to improve the therapeutic effect of HCC *** and future clinical trial data of combined therapy may develop the new treatment scheme for recurrent *** paper reviews the pattern of recurrent HCC and the characteristics of the immune microenvironment,demonstrates the basis for combining local treatment and systemic treatment,and reports current evidence to better understand current progress and future approaches in the treatment of recurrent HCC.
