Intensive management enhances mycorrhizal respiration but decreases free-living microbial respiration by affecting microbial abundance and community structure in Moso bamboo forest soils

Intensive management is known to markedly alter soil carbon(C)storage and turnover in Moso bamboo forests compared with extensive management.However,the effects of intensive management on soil respiration(RS)components remain unclear.This study aimed to evaluate the changes in different RScomponents(root,mycorrhizal,and free-living microorganism respiration)in Moso bamboo forests under extensive and intensive management practices.A1-year in-situ microcosm experiment was conducted to quantify the RScomponents in Moso bamboo forests under the two management practices using mesh screens of varying sizes.The results showed that the total RSand its components exhibited similar seasonal variability between the two management practices.Compared with extensive management,intensive management significantly increased cumulative respiration from mycorrhizal fungi by 36.73%,while decreased cumulative respiration from free-living soil microorganisms by 8.97%.Moreover,the abundance of arbuscular mycorrhizal fungi(AMF)increased by 43.38%,but bacterial and fungal abundances decreased by 21.65%and 33.30%,respectively,under intensive management.Both management practices significantly changed the bacterial community composition,which could be mainly explained by soil pH and available potassium.Mycorrhizal fungi and intensive management affected the interrelationships between bacterial members.Structural equation modeling indicated that intensive management changed the cumulative RSby elevating AMF abundance and lowering bacterial abundance.We concluded that intensive management reduced the microbial respiration-derived C loss,but increased mycorrhizal respiration-derived C loss.

Rationally tailored lipid nanoparticles(LNPs)with efficient and tunable delivery of mRNA in vivo are crucial for mRNA vaccines.Selective expression of antigenic protein in lymphoid tissues/organs could improve the immunostimulatory efficacy and safety of LNPs-based mRNA vaccines.Inspired by the metabolic behavior that long-chain saturated fatty acids tending to enter lymphoid tissue rather than the liver,we developed fatty acid-doped LNPs capable of mediating differential protein expressions in the liver and spleen when administered intravenously.When the molar ratio of saturated fatty acid located 60%–70%,the doped LNPs achieved the spleen selective mRNA translation.The mechanism could be attributed to the different cellular uptake behaviors of saturated fatty acids in hepatocytes.Immunization with a model antigen(ovalbumin)mRNA-loaded spleen selective LNPs,we observed enhanced antigen-specific T cell immune responses,and potent immunotherapeutic and immunoprophylactic efficacy in the mouse lymphoma model.Our natural long-chain saturated fatty acids metabolic characteristics-inspired design of LNPs for spleen-selective mRNA vaccines delivery will provide references for designing mRNA vaccines with high efficacy and safety for tumor immunotherapy.

Objective The latest perspective suggests that elevated levels of inflammation and cytokines are implicated in atonic postpartum hemorrhage.Lipopolysaccharide(LPS)has been widely used to induce inflammation in animal models.Therefore,this study aimed to induce uterine inflammation using LPS to investigate whether local inflammation triggers dysfunction and atrophy in the myometrium,as well as the potential underlying molecular mechanisms involved.Methods In vivo,an animal model was established by intraperitoneal injection of 300μg/kg LPS in rats on gestational day 21.Hematoxylin-eosin(H&E)staining and Masson staining were employed to determine morphological changes in the rat uterine smooth muscle.Enzyme-linked immunosorbent assay(ELISA)was used to detect inflammatory cytokines.Immunohistochemistry,tissue fluorescence,and Western blotting were conducted to assess the expression levels of the uterine contraction-related proteins Toll-like receptor 4(TLR4)and the nuclear factor kappa-B(NF-κB)signaling pathway.In vitro,human uterine smooth muscle cells(HUtSMCs)were exposed to 2μg/mL LPS to further elucidate the involvement of the TLR4/NF-κB signaling pathway in LPS-mediated inflammation.Results In this study,LPS induced uterine myometrial dysfunction in rats,leading to a disorganized arrangement,a significant increase in collagen fiber deposition,and widespread infiltration of inflammatory cells.In both in vivo animal models and in vitro HUtSMCs,LPS elevated IL-6,IL-1β,and TNF-αlevels while concurrently suppressing the expression of connexin 43(Cx43)and oxytocin receptor(OXTR).Mechanistically,the LPS-treated group exhibited TLR4 activation,and the phosphorylation levels of p65 and IκBαwere notably increased.Conclusion LPS triggered the TLR4/NF-κB signaling pathway,inducing an inflammatory response in the myometrium and leading to uterine myometrial dysfunction and uterine atony.

Investigations on lubrication characteristics of high-speed electric multiple unit gearbox by oil volume adjusting device

In this paper,a numerical simulation model of the flow field in a gearbox with an oil volume adjusting device is established for the first time to study its influence on the lubrication characteristics of a high-speed electric multiple unit(EMU)gearbox.The moving particle semi-implicit(MPS)method is used to numerically simulate the internal flow field of the gearbox of the high-speed EMU under working conditions.The effects of the velocity of the high-speed EMU,the immersion depth,and the oil sump temperature on the power loss of the gears and the lubricant quantity of each bearing are studied and provide an effective tool for the quantitative evaluation of the lubrication characteristics of the gearbox.The lubrication characteristics of the gearbox under different working conditions are studied when the oil volume adjusting device is closed and opened.The results show that the oil volume adjusting device mainly changes the amount of lubricant stirred by the output gear by changing the flow rate of lubricant from the cavity pinion(Cavity P)to the cavity gear(Cavity G),and thus affects the power loss of gears and the lubricant quantity of each bearing.

JAK inhibition ameliorated experimental autoimmune encephalomyelitis by blocking GM-CSF-driven inflammatory signature of monocytes

Monocytes are key effectors in autoimmunity-related diseases in the central nervous system(CNS)due to the critical roles of these cells in the production of proinflammatory cytokines,differentiation of T-helper(Th)cells,and antigen presentation.The JAK-STAT signaling is crucial for initiating monocytes induced immune responses by relaying cytokines signaling.However,the role of this pathway in modulating the communication between monocytes and Th cells in the pathogenesis of multiple sclerosis(MS)is unclear.Here,we show that the JAK1/2/3 and STAT1/3/5/6 subtypes involved in the demyelination mediated by the differentiation of pathological Th1 and Th17 and the CNS-infiltrating inflammatory monocytes in experimental autoimmune encephalomyelitis(EAE),a model for MS.JAK inhibition prevented the CNS-infiltrating CCR2-dependent Ly6^(Chi)monocytes and monocyte-derived dendritic cells in EAE mice.In parallel,the proportion of GM-CSF^(+)CD4^(+)T cells and GM-CSF secretion were decreased in pathological Th17 cells by JAK inhibition,which in turns converted CNS-invading monocytes into antigen-presenting cells to mediate tissue damage.Together,our data highlight the therapeutic potential of JAK inhibition in treating EAE by blocking the GM-CSF-driven inflammatory signature of monocytes.

The central environmental protection inspection (CEPI) system in China is a significant institutional innova‐tion in national environmental governance. The CEPI applies a joint supervision strategy to address salient en‐vironmental issues and strictly enforce the environmental responsibilities of local governments. This study col‐lects and organizes CEPI inspection reports covering three stages that encompass the first round, the “look back”, and the second round, applying text analysis to obtain sample data and conduct statistical quantifica‐tion of word frequency in inspection reports and identify notable changes. The study explores the allocation of CEPI attention between policy objectives and the intensity of policy instruments. We determine that in con‐junction with public opinion feedback, the CEPI conducts targeted inspections and focuses more on pollutant governance, which has high severity and can be addressed quickly. The CEPI fills the gap of normalized gover‐nance with a campaign-style governance approach. Regarding the intensity of policy measures, the CEPI pri‐marily uses economic incentive policy instruments, supplemented by command-and-control and public guid‐ance approaches, advancing the sustainability of regulatory effectiveness through economic, social, and politi‐cal activities. This study extends knowledge in the field of CEPI policy priorities and implementation, expand‐ing the literature related to outcomes of environmental policy in developing countries.

With the increasing oil demand, the construction of oil energy reserves in China needs to be further strengthened. However, given that there has been no research on the main influencing factors of crude oil temperature drop in storage tanks under actual dynamically changing environments, this paper considers the influence of dynamic thermal environment and internal crude oil physical properties on the fluctuating changes in crude oil temperature. A theoretical model of the unsteady-state temperature drop heat transfer process is developed from a three-dimensional perspective. According to the temperature drop variation law of crude oil storage tank under the coupling effect of various heat transfer modes such as external forced convection, thermal radiation, and internal natural convection, the external dynamic thermal environment influence zone, the internal crude oil physical property influence zone, and the intermediate transition zone of the tank are proposed. And the multiple non-linear regression method is used to quantitatively characterize the influence of external ambient temperature, solar radiation, wind speed, internal crude oil density, viscosity, and specific heat capacity on the temperature drop of crude oil in each influencing zone. The results of this paper not only quantitatively explain the main influencing factors of the oil temperature drop in the top, wall, and bottom regions of the tank, but also provide a theoretical reference for oil security reserves under a dynamic thermal environment.
