Calcitonin gene related peptide modified mesenchymal stem cells reduce restenosis after carotid balloon injury in rats

This work aimed to investigate the effects of calcitonin gene-related peptide(CGRP)-modified mesenchymal stem cells(MSCs)on vascular stenosis in carotid balloon-injured rats.The CGRP gene labeled with EGFP was transfected into bone marrow MSCs,and CGRP protein expression in MSCs was confirmed by immunofluorescence assays.A rat carotid balloon injury model was established using a surgicalmethod.CGRP-modifiedMSCs were orthotopically transplanted into the injured area of the rats.At 28 days after cell transplantation,EGFP and CD31 expression was detected by immunofluorescence staining.Hematoxylin-eosin(H&E)staining was used to detect the intima/media area of the injured carotid artery stenosis site,and the expression of proliferating cell nuclear antigen(PCNA)was detected by immunohistochemistry.MSCs from rat bone marrow positively expressed CD29 and negatively expressed CD45.In vivo immunofluorescence staining showed that EGFP expression was significantly increased at the vascular injury site of rats transplanted with MSC_CGRP compared with the control group on the 28th day after cell transplantation and that the damaged vessels exhibited continuous CD31 expression.H&E staining showed that the vascular intimal proliferation area of rats transplanted with MSC_CGRP was significantly reduced compared with that of other groups.Furthermore,the immunohistochemistry results showed that PCNA expression in the endothelium of the MSC_CGRP group was lower than that of the other groups.Therefore,MSCs transfected with the CGRP gene can express the CGRP protein,and the implantation of CGRPmodified MSCs at the damaged site after carotid balloon-induced injury can reduce the neointimal area.

Tailpipe emission characterizations of diesel-fueled forklifts under real-world operations using a portable emission measurement system

Non-road equipment is one of the key contributing sources to air pollution.Thus,an accurate development of emission inventory from non-road equipment is imperative for air quality management,especially for equipment with a large population such as diesel-fueled forklifts.The objective of this paper is to characterize duty-cycle based emissions from diesel-fueled forklifts using a portable emission measurement system(PEMS).Three dutycycles were defined in this study,including idling,moving,and working(active duty operation)and used to characterize in-use emissions for diesel-fueled forklifts.A total of twelve diesel-fueled forklifts were selected for real-world emission measurements.Results showed that fuel-based emission factors appear to have smaller variability compared to time-based ones.For example,the time-based emission factors for CO,HC,NO,and PM 2.5 for forklifts were estimated to be 16.6-43.9,5.3-15.1,26.2-49.9,5.5-11.1 g/hr with the fuel-based emission factors being 12.1-20.3,4.1-8.3,19.1-32.4,3.5-6.5 g/kg-fuel,respectively.NO emissions appear to be the biggest concern for emissions control.Furthermore,most of the emissions factors estimated from this study are significantly different from those in both National Guideline for Emission Inventory Development for Non-Road Equipment in China and welldeveloped emission factor models such as NONROAD by US EPA.This implies that localized,preferably fuel-based emission factors should be adjusted based on real-world emission measurements in order to develop a representative emission inventory for non-road equipment.

Evaluation of the applicability of multiple drought indices in the core zone of “westerlies-dominated climatic regime”

Drought indices are frequently used to measure the intensity,start and end of droughts.However,the performances of these indices depend on regions and the type of droughts.Therefore,it is necessary to evaluate whether these indices are applicable for a given region.This study evaluated the ability of the self-calibrating Palmer Drought Severity Index(sc PDSI),the Standardized Precipitation Evapotranspiration Index(SPEI),and the Standardized Moisture Anomaly Index(SZI) to describe the dry-wet conditions and drought events in the core zone of the “westerlies-dominated climatic regime”.The results showed that the 12-month SZI(SZI12) and SPEI(SPEI12) have good correlations with precipitation and PET respectively,while the sc PDSI can capture the changes in precipitation and soil moisture.In Xinjiang region of China,the SZI12 and sc PDSI showed that this region gradually became wetter during 1961–2014,which was consistent with increased precipitation,decreased PET,and improved vegetation.However,the SPEI12 showed significant drying due to the strong influence of the PET,suggesting that this index exaggerated the drought conditions in this region.Precipitation in Kazakhstan and the southern Central Asian regions has increased slightly over the past 50 years,but the PET has greatly increased,altogether,all three indices suggested a drying trend in Central Asia.The evaluation of the ability of these drought indices to identify typical drought events in Xinjiang region of China suggested that the sc PDSI can better detect the typical drought events compared with the SZI12.In conclusion,the sc PDSI is the most suitable index for characterizing the long-term trend of hydroclimate conditions and drought events in the core zone of the “westerlies-dominated climatic regime”.This study provides a theoretical basis for the rational utilization and improvement of the drought index,which is critical for monitoring,attributing,and predicting of drought events in arid areas.

Temperature variations along the Silk Road over the past 2000 years:Integration and perspectives

Reconstructing temperature changes along the Silk Road(SR)over the last two millennia can provide insights into past global changes and their impact on the rise and fall of ancient civilizations in this region.Numerous high-quality single-site paleotemperature records have been produced for the eastern part of the SR(mainly for the Xinjiang region and its surrounding areas),which provide the data basis for a comprehensive synthesis.In this study,we used objective criteria to select 10 highquality ones from 30 temperature reconstructions derived from various geological archives including lacustrine sediments,ice cores,and tree rings in this region.Our aims are to summarize the pattern of temperature change over the past 2000 years,to provide a long-term viewpoint on the present warming,and to evaluate the impact of climate change on civilizations along the SR.The principal results are as follows:(1)The temperature variations over the last millennium are mutually consistent within these records.The study area experienced typical climate anomalies during the Medieval Warm Period(MWP,AD 1000-1250),the Little Ice Age(LIA,AD 1450-1850),and the Current Warm Period(CWP,AD 1850 to present);however,contrary to previous knowledge,the amplitude of climatic warming during the CWP did not exceed that during the MWP.(2)Fewer temperature records were available for the interval AD 1–1000,and there were large differences between them.For example,the reconstructed climate during both the Han Dynasty and the Sui-Tang Dynasties was either warm or cold,without prevailing consensus.(3)The warming during the MWP favored the rapid development of the SR route along the northern slopes of the Tianshan Mountains,and the cooling during the LIA contributed to the decline of the SR marked by the closure of the Jiayuguan Pass.Notably,the scarcity of temperature records and the discrepancies between them during AD 1-1000 in the eastern part of the SR have hindered our understanding of the hydroclimatic changes and their influence on the developm

0 INTRODUCTION.The intensification of global warming is generating increasing risks to human society and sustainable development,which are of growing concern to the international scientific community and policymakers(Sun,2023).The ecosystems of the Asian drylands are fragile and highly sensitive to global changes;moreover,while these drylands are the most important areas of oasis agriculture on Earth,their water resources are being adversely impacted by ongoing climate change(Zhang et al.,2023).Hence,an improved understanding of the patterns of climate change in this region is important and attracting increasing research attention.

Vector meson effects on multi-Skyrmion states from the rational map ansatz

The roles of the lightest vector mesons ρ and ω in the multi-Skyrmion states are studied using the hidden local symmetry approach up to the next-to-leading order,including the homogeneous Wess-Zumino terms.The low-energy constants in the effective field theory are determined using the Sakai-Sugimoto model and the flat-space five-dimensional Yang-Mills action.With only two inputs,m_(ρ) and f_(π),it is possible to determine all low-energy constants without ambiguity.The vector meson effects can be investigated by sequentially integrating vector mesons,and their geometry can be elucidated by comparing the results using the low-energy constants estimated from the Sakai-Sugimoto model and the flat-space five-dimensional Yang-Mills action.We found that theρmeson reduces the masses of the multi-Skyrmion states and increases the overlaps of their constituents,whereas theωmeson repulses the constituents of the multi-Skyrmion states and increases their masses.Therefore,these vector mesons are crucial in the Skyrme model approach to nuclei.We also found that the warping factor,an essential element in the holographic model of QCD,affects the properties of the multi-Skyrmion states and cannot be ignored.

Ant Pupae from Myanmar Amber Reveal the Evolutionary Process of Cocoon-Spinning Behavior in Formicidae

many extant species spin cocoons before their pupal stage,which can protect them from predators,pathogens,and fluctuant environmental factors in nests.However,owing to the scarcity of ant cocoon fossils,little is known about the origin and evolution of this behavior in Formicidae.Here,we describe two ant pupae specimens from Myanmar amber.The presence of cocoons in these Mesozoic fossils suggests the ancestral origin of spinning cocoon behavior;based on our phylogenetic analysis and ancestral state reconstruction,this behavior was secondarily lost at least six times in Formicidae,which could be ascribed to the transition of nesting and feeding habits,as well as the development of eusociality.Moreover,the first specimen preserved worker ants with empty cocoons simultaneously,indicating garbage removal or brood care behavior,which improves our understanding of their eusociality in the Cretaceous.

Preserving details in semantics-aware context for scene parsing

Great success of scene parsing(also known as, semantic segmentation) has been achieved with the pipeline of fully convolutional networks(FCNs). Nevertheless, there are a lot of segmentation failures caused by large similarities between local appearances. To alleviate the problem, most of existing methods attempt to improve the global view of FCNs by introducing different contextual modules. Though the reconstructed high resolution output of these methods is of rich semantics, it cannot faithfully recover the fine image details owing to lack of desired precise low-level information. To overcome the problem, we propose to improve the spatial decoding process through embedding possibly lost low-level information in a principled way. To this end, we make the following three contributions. First, we propose a semantics conformity module to make low-level features variations agnostic. Second, we introduce semantics into the conformed low level features through guidance from semantically aware features. Finally, we institute the availability of various possible contextual features at feature fusion to enrich context information. The proposed approach demonstrates competitive performance on challenging PASCAL VOC 2012, Cityscapes,and ADE20K benchmarks in comparison to the state-of-the-art methods.

Mutations in the Ras oncogene are the most frequently cancer alterations, occurring in more than 30% of all human cancers. The failures in the development of successful clinical inhibitors against Ras have made Ras as a “undruggable” target. The major reason is lack of a systematic understanding of oncogenic cooperation between Ras activation and mutation of related tumor suppressor genes.1 The genetic techniques available in the Drosophila melanogaster allow analysis of the behavior of cells with distinct mutations, making this the ideal model organism to dissect oncogenic cooperation induced tumorigenesis.2 The exact same mutation, e.g., RasV12, was mimicked in the Drosophila, which enables us to perform large scale genetic screens, aiming to unearth novel tumor suppressors that can synergistically enhance RasV12-related tumor growth.

Effect of grain size on ferroelectric and dielectric properties of Bi_(3.25)La_(0.75)Ti_(3)O_(12) thin films prepared by rf-magnetron sputtering

Bi_(3.25)La_(0.75)Ti_(3)O_(12)(BLT)thin films are promising materials used in non-volatile memories.In this work,BLT films were deposited on Pt(111)/Ti/SiO_(2)/Si substrates by rf-magnetron sputtering method followed by annealing treatments.The microstructures of BLT thin films were investigated via X-ray diffraction(XRD),scanning electron microscopy(SEM)and atomic force microscopy(AFM).With the increase in annealing temperature,the grain size increased significantly and the preferred crystalline orientation changed.A well-saturated hysteresis loop with a superior remnant polarization of 15.4μC/cm^(2) was obtained for BLT thin films annealed at 700°C.The results show that the dielectric constant decreased with the increase in grain sizes.
