Osteosarcoma is a very serious primary bone cancer with a high death rate and a dismal *** there is no permanent therapy for this condition,it is necessary to develop a ***,this investigation was carried out to assess the impacts and biological functions of hydroxysafflor yellow A(HYSA)in osteosarcoma cell lines(MG63).In this investigational study,MG63 cells were *** experiments,quantitative polymerase chain reaction(qPCR),immunofluorescent staining,extracellular acidification rate(ECAR),oxygen consumption rate(OCR),glucose consumption,lactate production,and ATP levels,proliferation assay,5-Ethynyl-2′-deoxyuridine(EDU)staining,and Western blot were *** MG63 cells,HYSA lowered cell proliferation and metastasis rates,suppressed EDU cell number,and enhanced caspase-3/9 activity *** reduced the Warburg effect and induced ferroptosis(FPT)in MG63 *** ferroptosis diminished HYSA’s anti-cancer activities in MG63 *** stimulation of the HIF-1α/SLC7A11 pathway decreased HYSA’s anti-cancer activities in MG63 ***-1αis one target spot for HYSA in a model of osteosarcoma cancer(OC).HYSA altered HIF-1α’s thermophoretic activity;following binding with HYSA,HIF-1α’s melting point increased from~55°C to~60°*** significantly enhanced the thermal stability of exogenous WT HIF-1αwhile not affecting Mut HIF-1α,suggesting that ARG-311,GLY-312,GLN-347,and GLN-387 may be involved in the interaction between HIF-1αand ***,our study revealed that HYSA induced FPT and reduced the Warburg effect of OC through mitochondrial damage by HIF-1α/HK2/SLC7A11 *** is a possible therapeutic option for OC or other cancers.

With the theoretical and technological developments related to cratonic strike-slip faults,the shuntuoguole Low Uplift in the Tarim Basin has attracted considerable attention *** by multi-stage tectonic movements,the strike-slip faults have controlled the distribution of hydrocarbon resources owing to the special fault characteristics and fault-related *** contrast,the kinematics and formation mechanism of strike-slip faults in buried sedimentary basins are difficult to investigate,limiting the discussion of these faults and hydrocarbon *** this study,we identified the characteristics of massive sigmoidal tension gashes(STGs)that formed in the shunnan area of the Tarim ***-resolution three-dimensional seismic data and attribute analyses were used to investigate their geometric and kinematic ***,the stress state of each point of the STGs was calculated using seismic curvature ***,the formation mechanism of the STGs and their roles in controlling hydrocarbon migration and accumulation were *** results suggest that:(1)the STGs developed in the shunnan area have a wide distribution,with a tensile fault arranged in an enéchelon pattern,showing an S-shaped *** STGs formed in multiple stages,and differential rotation occurred along the direction of strike-slip stress during formation.(2)Near the principal displacement zone of the strike-slip faults,the stress value of the STGs was higher,gradually decreasing at both *** shallow layer deformation was greater than the deep layer deformation.(3)STGs are critical for connecting source rocks,migrating oil and gas,sealing horizontally,and developing efficient *** study not only provides seismic evidence for the formation and evolution of super large STGs,but also provides certain guidance for oil and gas exploration in this area.

Numerical simulation of inlet placement on sewage characteristics in the rounded square aquaculture tank with single inlet

To improve the self-cleaning ability of aquaculture tank and the efficiency of circulating water,physical and numerical experiments were conducted on the influence of inlet structure on sewage discharge in a rounded square aquaculture tank with a single *** on the physical model of the tank,analysis of how inlet structure adjustment affects sewage discharge efficiency and flow field characteristics was conducted to provide suitable flow field conditions for sinkable solid particle *** addition,an internal flow field simulation was conducted using the RNG k-εturbulence model in hydraulic drive *** a solid-fluid multiphase model was created to investigate how the inlet structure affects sewage collection in the rounded square aquaculture tank with single inlet and *** finding revealed that the impact of inlet structure is considerably affecting sewage *** conditions of C/B=0.07-0.11(the ratio of horizontal distance between the center of the inlet pipe and the tank wall(C)to length of the tank(B))andα=25°(αis the angle between the direction of the jet and the tangential direction of the arc angle)resulted in optimal sewage collection,which is similar to the flow field experiment in the rounded square aquaculture tank with single inlet and *** excellent correlation was revealed between sewage collection and fluid circulation stability in the aquaculture *** present study provided a reference for design and optimization of circulating aquaculture tanks in aquaculture industry.

BACKGROUND Pulmonary fibrosis(PF)is a chronic interstitial lung disease characterized by fibroblast proliferation and extracellular matrix formation,causing structural damage and lung *** cell therapy and mesenchymal stem cells-extracellular vesicles(MSC-EVs)offer new hope for PF *** To investigate the therapeutic potential of MSC-EVs in alleviating fibrosis,oxidative stress,and immune inflammation in A549 cells and bleomycin(BLM)-induced mouse *** The effect of MSC-EVs on A549 cells was assessed by fibrosis markers[collagen I andα-smooth muscle actin(α-SMA),oxidative stress regulators[nuclear factor E2-related factor 2(Nrf2)and heme oxygenase-1(HO-1),and inflammatory regu-lators[nuclear factor-kappaB(NF-κB)p65,interleukin(IL)-1β,and IL-2].Similarly,they were assessed in the lungs of mice where PF was induced by BLM after MSC-EV ***-EVs ion PF mice were detected by pathological staining and western ***-cell RNA sequencing was performed to investigate the effects of the MSC-EVs on gene expression profiles of macrophages after modeling in *** Transforming growth factor(TGF)-β1 enhanced fibrosis in A549 cells,significantly increasing collagen I andα-SMA ***,treatment with MSC-EVs demonstrated a remarkable alleviation of these ***,the expression of oxidative stress regulators,such as Nrf2 and HO-1,along with inflammatory regulators,including NF-κB p65 and IL-1β,were mitigated by MSC-EV ***,in a parallel manner,MSC-EVs exhibited a downregulatory impact on collagen deposition,oxidative stress injuries,and inflammatory-related cytokines in the lungs of mice with ***,the mRNA sequencing results suggested that BLM may induce PF in mice by upregulating pulmonary collagen fiber deposition and triggering an immune inflammatory *** findings collectively highlight the potential therapeutic efficacy of MSC-EVs in ameliorating fibrotic processes,oxidative stres

Proteomics analysis of coronary atherosclerotic heart disease with different Traditional Chinese Medicine syndrome types before and after percutaneous coronary intervention

OBJECTIVE: To investigate the underlying protein molecular mechanisms of "Qi stagnation and blood stasis syndrome"(QS) and "Qi deficiency and blood stasis syndrome"(QD), as two subtypes of coronary artery disease(CAD) in Traditional Chinese Medicine(TCM),following percutaneous coronary intervention(PCI).METHODS: In this study, a total of 227 CAD patients with QS and 211 CAD patients with QD were enrolled; all participants underwent PCI. Label-free quantification proteomics were employed to analyze the changes in serum in two subtypes of CAD patients before and 6 months after PCI, aiming to elucidate the intervention mechanism of PCI in treating CAD characterized by two different TCM ***: Biochemical analysis revealed significant changes in tumor necrosis factor-α, high density lipoprotein cholesterol, blood stasis clinical symptoms observation, and Gensini levels in both patient groups post-PCI; Proteomic analysis identified 79 and 95 differentially expressed proteins in the QS and QD patient groups, respectively, compared to their control *** C8 alpha chain, complement factor H,apolipoprotein H, apolipoprotein B, plasminogen,carbonic anhydrase 2, and complement factor Ⅰ were altered in both comparison groups. Furthermore,enrichment analysis demonstrated that cell adhesion and connectivity-related processes underwent changes in QS patients post-PCI, whereas lipid metabolism-related pathways, including the peroxisome proliferator-activated receptor signaling pathway and extracellular matrix receptor interaction, underwent changes in the QD *** protein-protein interaction network analysis further enriched 52 node proteins, including apolipoprotein B,lipoprotein(a), complement C5, apolipoprotein A4,complement C8 alpha chain, complement C8 beta chain,complement C8 gamma chain, apolipoprotein H,apolipoprotein A-Ⅱ, albumin, complement C4-B,apolipoprotein C3, among others. Fe/S oxidation-coupled arsenic speciation transformation mediated by AMD enrichment culture under different pH conditions

Arsenic(As)speciation transformation in acid mine drainage(AMD)is comprehensively affected by biological and abiotic factors,such as microbially mediated Fe/S redox reactions and changes in environmental conditions(pH and oxidation-reduction potential).However,their combined impacts on arsenic speciation transformation remain poorly ***,we explored arsenic transformation and immobilization during pyrite dissolution mediated by AMD enrichment culture under different acidic pH *** results for incubation and mineralogical transformation of solid residues show that in the presence of AMD enrichment culture,pH 2.0,2.5,and 3.0 are more conducive to the formation of jarosites and ferric arsenate,which could immobilize high quantities of dissolved arsenic by adsorption and *** pH conditions significantly affect the initial adsorption of microbial cells to the minerals and the evolution of microbial community structure,further infuencing the biodissolution of pyrite and the release and oxidation process of Fe/*** results of Fe/S/As speciation transformation of the solid residues show that the transformation of Fe,S,and As in solution is mainly regulated by pH and potential values,which imposed significantly different effects on the formation of secondary minerals and thus arsenic oxidation and *** above results indicated that arsenic transformation is closely related to the Fe/S oxidation associated with pyrite bio-oxidation,and this correlation is critically regulated by the pH conditions of the system.
