Loss of LBP triggers lipid metabolic disorder through H3K27 acetylation-mediated C/EBPβ-SCD activation in non-alcoholic fatty liver disease

Non-alcoholic fatty liver disease(NAFLD)is associated with mutations in lipopolysaccharide-binding protein(LBP),but the underlying epigenetic mechanisms remain understudied.Herein,LBP^(-/-)rats with NAFLD were established and used to conduct integrative targetingactive enhancer histone H3 lysine 27 acetylation(H3K27ac)chromatin immunoprecipitation coupled with high-throughput and transcriptomic sequencing analysis to explore the potential epigenetic pathomechanisms of active enhancers of NAFLD exacerbation upon LBP deficiency.Notably,LBP^(-/-)reduced the inflammatory response but markedly aggravated high-fat diet(HFD)-induced NAFLD in rats,with pronounced alterations in the histone acetylome and regulatory transcriptome.In total,1128 differential enhancer-target genes significantly enriched in cholesterol and fatty acid metabolism were identified between wild-type(WT)and LBP^(-/-)NAFLD rats.Based on integrative analysis,CCAAT/enhancer-binding proteinβ(C/EBPβ)was identified as a pivotal transcription factor(TF)and contributor to dysregulated histone acetylome H3K27ac,and the lipid metabolism gene SCD was identified as a downstream effector exacerbating NAFLD.This study not only broadens our understanding of the essential role of LBP in the pathogenesis of NAFLD from an epigenetics perspective but also identifies key TF C/EBPβand functional gene SCD as potential regulators and therapeutic targets.

Comprehensive analysis of endoplasmic reticulum stress-related mechanisms in type 2 diabetes mellitus

BACKGROUND The endoplasmic reticulum(ER)is closely related to a wide range of cellular functions and is a key component to maintain and restore metabolic health.Type 2 diabetes mellitus(T2DM)is a serious threat to human health,but the ER stress(ERS)-related mechanisms in T2DM have not been fully elucidated.AIM To identify potential ERS-related mechanisms and crucial biomarkers in T2DM.METHODS We conducted gene set enrichment analysis(GSEA)and gene set variation analysis(GSVA)in myoblast and myotube form GSE166502,and obtained the differentially expressed genes(DEGs).After intersecting with ERS-related genes,we obtained ERS-related DEGs.Finally,functional analyses,immune infiltration,and several networks were established.RESULTS Through GSEA and GSVA,we identified several metabolic and immune-related pathways.We obtained 227 ERS-related DEGs and constructed several important networks that help to understand the mechanisms and treatment of T2DM.Finally,memory CD4^(+)T cells accounted for the largest proportion of immune cells.CONCLUSION This study revealed ERS-related mechanisms in T2DM,which might contribute to new ideas and insights into the mechanisms and treatment of T2DM.

Effect of heat treatment on corrosion resistance and antibacterial ability of 0Cr15Ni5Cu3Mo carbon steel

Before and after heat treatment,0Cr15Ni5Cu3MoN stainless steel(SS)samples were immersed in sulfate-reducing bacteria(SRB)environment to study the effect of heat treatment on the antimicrobial properties of Cu-containing stainless steel.The effect of heat treatment on the corrosion resistance and antibacterial ability of 0Cr15Ni5Cu3MoN SS was studied by scanning electron microscopy,transmission electron microscopy,laser scanning confocal microscopy,and electrochemistry.The results show that 0Cr15Cu3 SS has better antibacterial properties with fewer bacteria adsorbed on the surface after heat treatment,and its corrosion resistance is significantly improved.Interestingly,on the 7th day of immersion,the corrosion resistance of the heat-treated stainless steel was lower than that of the rolled stainless steel.This was due to the fact that the rolled samples were protected by biofilm at this time,while the surface bacteria on the heat-treated samples died,resulting in the exposure of the substrate surface and thus reducing the corrosion resistance of the heat-treated samples.Thus,because of the strong bactericidal properties of the heat-treated Cu-containing stainless steel,the corrosion resistance of the samples suddenly decreases in the middle stage of corrosion,and this discovery provides a new view to study the corrosion process of Cu-containing stainless steel in the SRB environment.

Sintering behavior of aluminum nitride powder prepared by self-propagating high-temperature synthesis method

Fully dense aluminum nitride(AIN) ceramics were synthesized by self-propagating high-temperature synthesis(SHS) method using AIN powder as raw material with Y2O3additive. The sintering behavior was studied at different sintering temperatures and additive contents. The change of phase compositions, secondary phase distributions and grain morphologies during sintering process were investigated. It is shown that fully dense ceramics using AIN powder prepared by SHS method can be obtained when the sintering temperature is above 1830 ℃. Both Y2O3content and sintering temperature have an important influence on the formation of Y-Al-O phase and grain shape. When Y2O3content is identified, the grain morphology converts from polyhedron into sphere-like shape with the rise of sintering temperature. At a certain sintering temperature,the grain size decreases with the increase in Y2O3content. The influencing mechanisms of different YAl-O secondary phases and sintering temperatures on the grain size and morphology were also discussed based on the experimental results.

Interleukin-37b(hereafter called IL-37)was identified as fundamental inhibitor of natural and acquired immunity.The molecular mechanism and function of IL-37 in colorectal cancer(CRC)has been elusive.Here,we found that IL-37 transgenic(IL-37tg)mice were highly susceptible to colitis-associated colorectal cancer(CAC)and suffered from dramatically increased tumor burdens in colon.Nevertheless,IL-37 is dispensable for intestinal mutagenesis,and CRC cell proliferation,apoptosis,and migration.Notably,IL-37 dampened protective cytotoxic T cell-mediated immunity in CAC and B16-OVA models.CD8^(+)T cell dysfunction is defined by reduced retention and activation as well as failure to proliferate and produce cytotoxic cytokines in IL-37tg mice,enabling tumor evasion of immune surveillance.The dysfunction led by IL-37 antagonizes IL-18–induced proliferation and effector function of CD8+T cells,which was dependent on SIGIRR(single immunoglobulin interleukin-1 receptor-related protein).Finally,we observed that IL-37 levels were significantly increased in CRC patients,and positively correlated with serum CRC biomarker CEA levels,but negatively correlated with the CD8+T cell infiltration in CRC patients.Our findings highlight the role of IL-37 in harnessing antitumor immunity by inactivation of cytotoxic T cells and establish a new defined inhibitory factor IL-37/SIGIRR in cancerimmunity cycle as therapeutic targets in CRC.

Emergent symmetry in quantum phase transition:From deconfined quantum critical point to gapless quantum spin liquid

The emergence of exotic quantum phenomena in frustrated magnets is rapidly driving the development of quantum many-body physics,raising fundamental questions on the nature of quantum phase transitions.Here we unveil the behaviour of emergent symmetry involving two extraordinarily representative phenomena,i.e.,the deconfined quantum critical point(DQCP)and the quantum spin liquid(QSL)state.Via large-scale tensor network simulations,we study a spatially anisotropic spin-1/2 square-lattice frustrated antiferromagnetic(AFM)model,namely the J1x-J1y-J2 model,which contains anisotropic nearestneighbor couplings J1x,J1y and the next nearest neighbor coupling J2.For small J1y/J1x,by tuning J2,a direct continuous transition between the AFM and valence bond solid phase is observed.With growing J1y/J1x,a gapless QSL phase gradually emerges between the AFM and VBS phases.We observe an emergent O(4)symmetry along the AFM–VBS transition line,which is consistent with the prediction of DQCP theory.Most surprisingly,we find that such an emergent O(4)symmetry holds for the whole QSL–VBS transition line as well.These findings reveal the intrinsic relationship between the QSL and DQCP from categorical symmetry point of view,and strongly constrain the quantum field theory description of the QSL phase.The phase diagram and critical exponents presented in this paper are of direct relevance to future experiments on frustrated magnets and cold atom systems.

Correlation of BI-RADS grading with cancer cell proliferation and invasion

Objective:To investigate the correlation of BI-RADS grading with cancer cell proliferation and invasion.Methods: A total of 138 patients who were diagnosed with breast cancer in our hospital between August 2015 and October 2017 were divided into 2 cases at stage Ⅲ, 51 cases at stage Ⅳ and 85 cases at stage Ⅴ after the BI-RADS grading was reviewed. Differences in the expression of proliferation-related genes and invasion-related genes were compared among the three groups of patients.Results: Expressions of pro-proliferation genes MTA2, mTOR, PKM2, Notch1 and FSCN1 mRNA in lesion tissue of patients at stage Ⅴ and Ⅳ were higher than those of patients at stage Ⅲ whereas expressions of anti-proliferation genes BRCA1 and PTEN mRNA were lower than those of patients at stage Ⅲ;expressions of pro-invasion genes Gab2 and NUAK1 mRNA were higher than those of patients at stage Ⅲ whereas expressions of anti-invasion genes EBP50, EDN3, FBXW7 and NDRG1 mRNA were lower than those of patients at stage Ⅲ. With the increase of BI-RADS grades, the expression levels of pro-proliferation genes and pro-invasion genes further increased while the expression levels of anti-proliferation and anti-invasion genes further decreased in the lesion tissue.Conclusions: BI-RADS grading of patients with breast cancer is positively correlated with the proliferation and invasion activity of cancer cells.

ThyroidNet:A Deep Learning Network for Localization and Classification of Thyroid Nodules

Aim:This study aims to establish an artificial intelligence model,ThyroidNet,to diagnose thyroid nodules using deep learning techniques accurately.Methods:A novel method,ThyroidNet,is introduced and evaluated based on deep learning for the localization and classification of thyroid nodules.First,we propose the multitask TransUnet,which combines the TransUnet encoder and decoder with multitask learning.Second,we propose the DualLoss function,tailored to the thyroid nodule localization and classification tasks.It balances the learning of the localization and classification tasks to help improve the model’s generalization ability.Third,we introduce strategies for augmenting the data.Finally,we submit a novel deep learning model,ThyroidNet,to accurately detect thyroid nodules.Results:ThyroidNet was evaluated on private datasets and was comparable to other existing methods,including U-Net and TransUnet.Experimental results show that ThyroidNet outperformed these methods in localizing and classifying thyroid nodules.It achieved improved accuracy of 3.9%and 1.5%,respectively.Conclusion:ThyroidNet significantly improves the clinical diagnosis of thyroid nodules and supports medical image analysis tasks.Future research directions include optimization of the model structure,expansion of the dataset size,reduction of computational complexity and memory requirements,and exploration of additional applications of ThyroidNet in medical image analysis.

Patient satisfaction and follow-up adherence to glaucoma case management clinic in China

AIM:To assess glaucoma patient satisfaction and follow-up adherence in case management and identify associated predictors to improve healthcare quality and patient outcomes.METHODS:In this cross-sectional study,a total of 119 patients completed a Patient Satisfaction Questionnaire-18 and a sociodemographic questionnaire.Clinical data was obtained from the case management system.Follow-up adherence was defined as completing each follow-up within±30d of the scheduled time set by ophthalmologists during the study period.RESULTS:Average satisfaction scored 78.65±7,with an average of 4.39±0.58 across the seven dimensions.Age negatively correlated with satisfaction(P=0.008),whilst patients with follow-up duration of 2 or more years reported higher satisfaction(P=0.045).Multivariate logistics regression analysis revealed that longer follow-up durations were associated with lower follow-up adherence(OR=0.97,95%CI,0.95-1.00,P=0.044).Additionally,patients with suspected glaucoma(OR=2.72,95%CI,1.03-7.20,P=0.044)and those with an annual income over 100000 Chinese yuan demonstrated higher adherence(OR=5.57,95%CI,1.00-30.89,P=0.049).CONCLUSION:The case management model proves effective for glaucoma patients,with positive adherence rates.The implementation of this model can be optimized in the future based on the identified factors and extended to glaucoma patients in more hospitals.

Mitogenome-based phylogeny of mosquitoes(Diptera:Culicidae)

Mosquitoes are of great medical significance as vectors of many deadly diseases.Mitogenomes have been widely used in phylogenetic studies,but mitogenome knowledge within the family Culicidae is limited,and Culicidae phylogeny is far from resolved.In this study,we surveyed the mitogenomes of 149 Culicidae species,including 7 newly sequenced species.Comparative analysis of 149 mosquito mitogenomes shows gene composition and order to be identical to that of an ancestral insect,and the AT bias,length variation,and codon usage are all consistent with that of other reported Dipteran mitogenomes.Phylogenetic analyses based on the DNA sequences of the 13 protein-coding genes from the 149 species robustly support the monophyly of the subfamily Anophelinae and the tribes Aedini,Culicini,Mansonini,Sabethini,and Toxorhynchitini.To resolve ambiguous relationships between clades within the subfamily Culicinae,we performed topological tests and show that Aedini is a sister to Culicini and that Uranotaenini is a sister to(Mansonini)+(Toxorhynchitini+Sabethini).In addition,we estimated divergence times using a Bayesian relaxation clock based on the sequence data and 3 fossil calibration points.The results show mosquitoes diverged during the Early Jurassic with massive Culicinae radiations during the Cretaceous,coincident with the emergence of angiosperms and the burst of mammals and birds.Overall,this study,which uses the largest number of Culicidae mitogenomes sequenced to date,comprehensively reveals the mitogenome characteristics and mitogenome-based phylogeny and divergence times of Culicidae,providing information for further studies on the mitogenome,phylogeny,evolution,and taxonomic revision of Culicidae.
