Breast cancer is the most common female cancer around the world.Approximately 5%to 10%of breast cancer is associated with a hereditary predisposition from inherited germline mutations,[1]with about 15% of mutations occurring in the breast cancer susceptibility genes(BRCA1/2).[2-4]Individuals carrying BRCA1/2 mutations may reduce their risk of breast cancer through screening and preventing managements.

Studies have shown that microRNAs(miRNAs) mediate posttranscriptional regulation of target genes and participate in various physiological and pathological processes, including peripheral nerve injury. However, it is hard to select key miRNAs with essential biological functions among a large number of differentially expressed miRNAs. Previously, we collected injured sciatic nerve stumps at multiple time points after nerve crush injury, examined gene changes at different stages(acute, sub-acute, and post-acute), and obtained mRNA expression profiles. Here, we jointly analyzed mRNAs and miRNAs, and investigated upstream miRNAs of differentially expressed mRNAs using Ingenuity Pathway Analysis bioinformatic software. A total of 31, 42, 30, and 23 upstream miRNAs were identified at 1, 4, 7, and 14 days after rat sciatic nerve injury, respectively. Temporal expression patterns and biological involvement of commonly involved upstream miRNAs(miR-21, let-7, miR-223, miR-10 b, miR-132, miR-15 b, miR-127, miR-29 a, miR-29 b, and miR-9) were then determined at multiple time points. Expression levels of miR-21, miR-132, miR-29 a, and miR-29 b were robustly increased after sciatic nerve injury. Biological processes involving these miRNAs include multicellular organismal response to stress, positive regulation of the epidermal growth factor receptor signaling pathway, negative regulation of epithelial cell differentiation, and regulation of myocardial tissue growth. Moreover, we constructed mechanistic networks of let-7, miR-21, and miR-223, the most significantly involved upstream miRNAs. Our findings reveal that multiple upstream miRNAs(i.e., let-7, miR-21, and miR-223) were associated with gene expression changes in rat sciatic nerve stumps after nerve injury, and these miRNAs play an important role in peripheral nerve regeneration. This study was approved by the Experimental Animal Ethics Committee of Jiangsu Province of China(approval No. 20190303-18) on March 3, 2019.

Pathogenic bacteria can proliferate rapidly on porous fabrics to form bacterial plaques/bioflms,resulting in potential sources of cross-transmissions of diseases and increasing cross-infection in public environments.Many works on antibacterial modifcation of cotton fabrics have been reported,while very few works were reported to endow poly(ethylene terephthalate)(PET)fabrics with non-leaching antibacterial function without compromising their innate physicochemical properties though PET is the most widely used fabric.Therefore,it is urgent to impart the PET fabrics with non-leaching antibacterial activity.Herein,a novel N-halamine compound,1-chloro-3-benzophenone-5,5-dimethylhydantoin(Cl-BPDMH),was developed to be covalently bonded onto PET fabrics,rendering non-leaching antibacterial activity while negligible cytotoxicity based on contact-killing principle.Bacterial was easily adhered to Cl-BPDMH fnished PET fabrics,and then it was inactivated quickly within 10 s.Furthermore,the breaking strength,breaking elongation,tearing strength,water vapor permeability,air permeability and whiteness of Cl-BPDMH fnished PET fabrics were improved obviously compared to raw PET fabrics.Hence,this work developed a facile approach to fabricate multifunctional synthetic textiles to render outstanding and rapid bactericidal activity without compromising their physicochemical properties and biocompatibility.

Artificial intelligence generated content(AIGC)has emerged as an indispensable tool for producing large-scale content in various forms,such as images,thanks to the significant role that AI plays in imitation and production.However,interpretability and controllability remain challenges.Existing AI methods often face challenges in producing images that are both flexible and controllable while considering causal relationships within the images.To address this issue,we have developed a novel method for causal controllable image generation(CCIG)that combines causal representation learning with bi-directional generative adversarial networks(GANs).This approach enables humans to control image attributes while considering the rationality and interpretability of the generated images and also allows for the generation of counterfactual images.The key of our approach,CCIG,lies in the use of a causal structure learning module to learn the causal relationships between image attributes and joint optimization with the encoder,generator,and joint discriminator in the image generation module.By doing so,we can learn causal representations in image’s latent space and use causal intervention operations to control image generation.We conduct extensive experiments on a real-world dataset,CelebA.The experimental results illustrate the effectiveness of CCIG.

A signed graph G˙=(G,σ)is a graph G=(V(G),E(G))with vertex set V(G)and edge set E(G),together with a functionσ:E→{+1,−1}assigning a positive or negative sign to each edge.In this paper,we present a more elementary proof for the matrix-tree theorem of signed graphs,which is based on the relations between the incidence matrices and the Laplcians of signed graphs.As an application,we also obtain the results of Monfared and Mallik about the matrix-tree theorem of graphs for signless Laplacians.

The lighter magnesium(Mg) isotopic signatures observed in intraplate basalts are commonly thought to result from deep carbonate recycling, provided that the sharp difference in Mg isotopic composition between surface carbonates and the normal mantle is preserved during plate subduction. However, deep subduction of carbonates and silicates could potentially fractionate Mg isotopes and change their chemical compositions. Subducting silicate rocks that experience metamorphic dehydration lose a small amount of Mg, and preserve the original Mg isotopic signature of their protoliths. When the dehydrated fluids dissolve carbonate minerals, they may evolve into lighter Mg isotopic compositions. The solubility of carbonate minerals in fluids decreases in the order of calcite, aragonite, dolomite, magnesite and siderite, leading to selective and partial dissolution of carbonate minerals along the subduction path. At the island arc depth(70–120 km), the metamorphic fluid dissolves mainly Mg-poor calcites, and thus the fluid has difficulty modifying the Mg isotopic system of the mantle wedge and associated arc basalts. At the greater depth of the back arc system or continental margin(>150 km), the supercritical fluid can dissolve Mg-rich carbonate minerals, and its interaction with the mantle wedge could significantly imprint the light Mg isotopic signature onto the mantle rocks and derivatives. Meanwhile, the carbonate and silicate remaining within the subducting slab could experience elemental and isotopic exchange, during which the silicate can obtain a light Mg isotopic signature and high CaO/Al2O3, whereas the carbonates, particularly the Ca-rich limestone, shift Mg isotopes and Mg O contents towards higher values. If this isotopic and elemental exchange event occurs widely during crustal subduction, subducted Ca-rich carbonates can partially transform into being Mg-rich, and a portion of recycled silicates(e.g. carbonated eclogites) can have light Mg isotopic composition alongside carbonates. Lipid droplet(LD)in vegetative tissues has recently been implicated in environmental responses in plants,but its regulation and its function in stress tolerance are not well understood.Here,we identified a Membrane Occupation and Recognition Nexus 1(MORN1)gene as a contributor to natural variations of stress tolerance through genome-wide association study in Arabidopsis thaliana.Characterization of its loss-of-function mutant and natural variants revealed that the MORN1 gene is a positive regulator of plant growth,disease resistance,cold tolerance,and heat tolerance.The MORN1 protein is associated with the Golgi and is also partly associated with LD.Protein truncations that disrupt these associations abolished the biological function of the MORN1 protein.Furthermore,the MORN1 gene is a positive regulator of LD abundance,and its role in LD number regulation and stress tolerance is highly linked.Therefore,this study identifies MORN1 as a positive regulator of LD abundance and a contributor to natural variations of stress tolerance.It implicates a potential involvement of Golgi in LD biogenesis and strongly suggests a contribution of LD to diverse processes of plant growth and stress responses.

Impact of transcranial electrical stimulation on serum neurotrophic factors and language function in patients with speech disorders

BACKGROUND Speech disorders have a substantial impact on communication abilities and quality of life.Traditional treatments such as speech and psychological therapies frequently demonstrate limited effectiveness and patient compliance.Transcranial electrical stimulation(TES)has emerged as a promising non-invasive treatment to improve neurological functions.However,its effectiveness in enhancing language functions and serum neurofactor levels in individuals with speech disorders requires further investigation.AIM To investigate the impact of TES in conjunction with standard therapies on serum neurotrophic factor levels and language function in patients with speech disorders.METHODS In a controlled study spanning from March 2019 to November 2021,81 patients with speech disorders were divided into a control group(n=40)receiving standard speech stimulation and psychological intervention,and an observation group(n=41)receiving additional TES.The study assessed serum levels of ciliary neurotrophic factor(CNTF),glial cell-derived neurotrophic factor(GDNF),brainderived neurotrophic factor(BDNF),and nerve growth factor(NGF),as well as evaluations of motor function,language function,and development quotient scores.RESULTS After 3 wk of intervention,the observation group exhibited significantly higher serum levels of CNTF,GDNF,BDNF,and NGF compared to the control group.Moreover,improvements were noted in motor function,cognitive function,language skills,physical abilities,and overall development quotient scores.It is worth mentioning that the observation group also displayed superior perfor CONCLUSION This retrospective study concluded that TES combined with traditional speech and psychotherapy can effectively increase the levels of neurokines in the blood and enhance language function in patients with speech disorders.These results provide a promising avenue for integrating TES into standard treatment methods for speech disorders.

Optimal allocation method of energy storage for integrated renewable generation plants based on power market simulation

This study designs and proposes a method for evaluating the configuration of energy storage for integrated re-newable generation plants in the power spot market,which adopts a two-level optimization model of“system simulation+plant optimization”.The first step is“system simulation”which is using the power market simu-lation model to obtain the initial nodal marginal price and curtailment of the integrated renewable generation plant.The second step is“plant optimization”which is using the operation optimization model of the integrated renewable generation plant to optimize the charge-discharge operation of energy storage.In the third step,“sys-tem simulation”is conducted again,and the combined power of renewable and energy storage inside the plant is brought into the system model and simulated again for 8,760 h of power market year-round to quantify and compare the power generation and revenue of the integrated renewable generation plant after applying energy storage.In the case analysis of the provincial power spot market,an empirical analysis of a 1 GW wind-solar-storage integrated generation plant was conducted.The results show that the economic benefit of energy storage is approximately proportional to its capacity and that there is a slowdown in the growth of economic benefits when the capacity is too large.In the case that the investment benefit of energy storage only considers the in-come of electric energy-related incomes and does not consider the income of capacity mechanism and auxiliary services,the income of energy storage cannot fulfill the economic requirements of energy storage investment.
