Long-Term Outcomes of Systemic-to-Pulmonary Artery Shunt in Patients with Functional Single Ventricle and Heterotaxy Syndrome

Objectives: We sought to determine the long-term outcomes and mortality-associated factors after systemic-topulmonaryartery shunt (SPS) in patients with heterotaxy syndrome. Methods: We retrospectively analyzed allpatients with a functional single ventricle and heterotaxy syndrome who underwent SPS at our center from January2001 to April 2022. Results: This study involved 84 patients. Restrictive pulmonary blood flow requiringearly modulation was presented in 34 (40%) patients. Compared with patients without restrictive pulmonaryblood flow (N = 50 [60%]), the postnatal survival of these 34 patients was significantly lower at 10 years (log rank:p = 0.04), but the statistical significance disappeared at 20 years (log rank: p = 0.18). Among 31 patients whounderwent SPS, 11 (35%) had extracardiac total anomalous pulmonary venous connection (TAPVC). The survivalrate after SPS was 80% at 10 years. Cox regression analysis showed that extracardiac TAPVC (hazard ratio6.44, 95% confidence interval 1.23–33.7, p = 0.03) and pulmonary venous obstruction (PVO) at TAPVC repair(hazard ratio 11.2, 95% confidence interval 2.13–58.5, p = 0.004) were significantly associated with death. In25 patients who underwent bidirectional cavopulmonary shunt (BCPS), surgical interventions on the pulmonaryartery (PA) were performed after SPS in 7 of 9 patients with PA coarctation, 3 of 4 with non-confluent PAs, and4 of 12 with normal PAs. At SPS, primary central PA plasty was performed in three patients with PA coarctationand 2 with non-confluent PAs. There was no significant difference in the PA index before BCPS between patientswith and without primary central PA plasty (p = 0.49). Among 20 patients who underwent total cavopulmonaryconnection (TCPC), adverse events occurred in 7 (35%) patients, including death in 1 (5%), intervention for pulmonaryarteriovenous malformation (PAVM) in 3 (15%), and surgical intervention for PVO in 3 (15%). Unsteady Behavior of Cavitating Waterjet in an Axisymmetric Convergent-Divergent Nozzle: High Speed Observation and Image Analysis Based on Frame Difference Method

Although it is well known that cloud cavitation shows unsteady behavior with the growing motion of an attached cavity, the shedding motion of a cloud, the collapsing motion of the cloud shed downstream and a reentrant motion in flow fields such as on a 2-D hydrofoil and in a convergent- divergent channel with a rectangular cross-section, observations for the periodic behavior of cloud cavitation in a cylindrical nozzle with a convergent-divergent part, which is mainly used in an industrial field, have hardly been conducted. From engineering viewpoints, it is important to elucidate the mechanism of periodic cavitation behavior in a cylindrical nozzle. In this study, a high-speed observation technique with an image analysis technique was applied to the cloud cavitation behavior in the nozzle to make clear the mechanism of unsteady behavior. As a result, it was observed in the nozzle that the periodic behavior occurs in the cloud cavitation and pressure waves form at the collapse of clouds shed downstream. Also, it was found through the image analysis based on the present technique that the pressure wave plays a role as a trigger mechanism to cause a reentrant motion at the downstream end of an attached cavity.

Loss-of-Function of a Ubiquitin-Related Modifier Promotes the Mobilization of the Active MITE mPing

Miniature inverted-repeat transposable elements （MITEs） are widespread in both prokaryotic and eukaryotic genomes, where their copy numbers can attain several thousands. Little is known, however, about the genetic factor（s） affecting their transpositions. Here, we show that disruption of a gene encoding ubiquitin-like protein markedly enhances the transposition activity of a MITE roPing in intact rice plants without any exogenous stresses. We found that the transposition activity of roPing is far higher in the lines harboring a non-functional allele at the Rurml （Rice ubiquitin-related modifier-I） locus than in the wild-type line. Although the alteration of cytosine methylation pattern triggers the activation of transposable elements under exogenous stress conditions, the methylation degrees in the whole genome, the roPing-body region, and the roPing-flanking regions of the non-functional Rurml line were unchanged. This study provides experimental evidence for one of the models of genome shock theory that genetic accidents within cells enhance the transposition activities of transposable elements.

Study of Rectangular Underexpanded Microjets

The underexpanded microjet emerging from a rectangular convergent nozzle with a high aspect ratio at the nozzle exit is investigated numerically using the Reynolds-averaged Navier-Stokes (RANS) simulation with the Menter’s shear stress transport (SST) k-ω turbulence model. The simulation is performed at the nozzle pressure ratio of 5.0 to produce a strong shock and it is validated by a comparison with a rainbow schlieren picture of the microjet. The three-dimensional structure of the shock-containing rectangular microjet is demonstrated using the isopycnic surface and bright-field schlieren representations.

Influence of Insertion of the Last Sense Codon on Expression Efficiency of Green Fluorescent Protein Gene in Escherichia coli

We studied the relationship between insertion of the last sense codon (i.e., the codon preceding the stop codon) and the efficiency of gene expression. We inserted 64 kinds of last sense codon at the 5’ end of the stop codon of the green fluorescent protein (GFP) gene and introduced the modified GFP genes into Escherichia coli (E. coli). Measuring the fluorescence intensity of the GFP produced in E. coli showed that the last sense codon influenced GFP gene expression and when CCG was inserted as the last sense codon, fluorescence intensity of E. coli was increased to 2.09 fold. On the other hand, insertion of CUA caused decrease of fluorescence intensity to 0.33 fold. We hope that our findings, which may be applicable to gene engineering, will be useful for further studies of protein expression.

Investigation of the Behavior of a Jet Issued into Two-Layer Density-Stratified Fluid

This study experimentally investigates a jet flow issued into a two-layer density-stratified fluid in a tank and the resultant mixing phenomena. The upper and lower fluids are water and a NaCl- water solution, respectively, with the lower fluid issued vertically upward from a circular nozzle mounted on the tank bottom. Experimental highlights of the jet behavior and mixing phenomena are classified into three patterns according to the jet Reynolds number and mass concentration of the NaCl-water solution. The internal density current clearly occurs along the density interface, and the maximum jet height is predicted by the Froude number defined by the density difference between the upper and lower fluids. The effect of fluid thickness on the maximum jet height is also clarified.

The paper gives description of regular elements of the semigroup B X (D) which are defined by semilattices of the class Σ2 (X, 8), for which intersection the minimal elements is not empty. When X is a finite set, the formulas are derived, by means of which the number of regular elements of the semigroup is calculated. In this case the set of all regular elements is a subsemigroup of the semigroup B X (D) which is defined by semilattices of the class Σ2 (X, 8).

High Speed Observation of Periodic Cavity Behavior in a Convergent-Divergent Nozzle for Cavitating Water Jet

Cloud cavitation shows an unsteady periodic tendency under a certain flow condition. In a cavitating water jet flow with cavitation clouds, the cavities or the clouds produce high impact at their collapse. In order to make clear a mechanism of the periodic cavity behavior, we experimentally examine the behavior in a transparent cylindrical convergent-divergent nozzle using a high-speed video camera. An effect of upstream pressure fluctuation due to a plunger pump is investigated from a viewpoint of unsteady behavior in a cavitating water jet. As a result, it is found that the cavitating flow has two kinds of oscillation patterns in the cavity length (cavitation cloud region). One is due to the upstream pressure fluctuation caused by the plunger pump. The other is much shorter periodic motion related to the characteristic oscillation of cavitation clouds accompanied with the shrinking (reentrant), growing and shedding motion of the clouds.
