A rapid method for extraction,purification and structure analysis of chondroitin sulfate from six marine tissues

This study was designed to establish a strategy for the extraction,purification,and structure analysis of chondroitin sulfate(cs)in milligram amounts.Crude acidic polysaccharides were extracted from six kinds of marine animals by enzymatic hydrolysis and hexadecylpyridinium chloride precipitation and purified by Q Sepharose Fast Flow strong anion exchange column.The purification of each crude polysaccharide was completed within 1 h.The structure of the polysaccharides,i.***.their chemical characterization,functional group,molecular weight and monosaccharide composition,were analyzed by colorimetry,nuclear magnetic resonace and high-performance liquid chromatogrpahy,respectively.All polysaccharides were identified as Cs.The oligosaccharide profile produced by enzyme hydrolysis of polysaccharides was determined by strong anion-exchange high-performance liquid chromatorgraphy.This method can be widely applied to the extraction and characterization of chondroitin sulfate from unknown raw materials,screening high-quality sources of functional polysaccharides,and laying a good foundation for thefollowing studyof the structure-function relationshipof polysaccharides.

Background and objectives:High-pressure processing(HPP)is a promising assistive method to extract pectic polysaccharides with high rhamnogalacturonan I(RG-I)domain and berries are sources of such pectic polysaccharides.This study extracts pectic polysaccharides from goji berry,raspberry,and cranberry,examines how HPP influences the pectic polysaccharide structure of three berries,and provides a basis for the extraction and modification of pectic polysaccharides with specific structure and bioactivity.Materials and methods:An orthogonal test was performed to optimize the HPP-assisted alkali method to extract the high yield and high RG-I content pectic polysaccharides from three berries.Structural information of pectic polysaccharides extracted by the HPP method and conventional methods were compared from the perspectives of monosaccharide composition,molecular weight,Fourier transform infrared spectroscopy,andnuclearmagneticresonancespectroscopy.Results:For raspberry,the optimal conditions consisted of a pressure of 50o MPa,a pH of 13,and a pressure-holding time of 12 min,while the optimal conditions for goji berry and cranberry were both 40o MPa,pH 13,and 15 min.Under the optimal conditions,the yields for goji berry,raspberry,and cranberry were 10.49%,16.63%,and 17.52%,respectively,and RG-l contents were 81.85%,83.30%,and 63.22%,respectively.HPP showed an effect to degrade homogalacturonan backbones and side chains and increase the RG-I content to some extent.Conclusion:HPP-assisted alkali method was revealed to be an efficient method to extract high RG-I content pectic polysaccharides,especially for cranberry,and was a potential method to modify pectic polysaccharide structure in a certain way.

Segregation behavior and its regulating process in as-cast magnesium alloy containing heavy rare earth

Specific gravity segregation that occurs during the smelting process always leads to the low composition homogeneity and poor performance stability of the magnesium-rare earth(Mg-RE)alloys.In this study,the segregation behavior of Mg-Gd alloy was investigated by sampling from different locations in the ingot fabricated in a resistance furnace without a pouring process.The combined application of induction-heating and mechanical stirring with various speeds(0-130 r/min)was applied to promote the distribution homogeneity of Gd atoms.In the resistance-heating fabricated ingot,Gd content at the bottom section reaches 407%of that at the top.The coarse dendrites surrounded by the network-like eutectic structures are responsible for the brittle fracture with a poor elongation of 3.7%.By the combined employment of the induction heating and mechanical stirring with the speed of 87 r/min at 740℃for 40 s,the variation of the Gd content within the whole ingot can be reduced to be the minimum of 0.23 wt%.Corresponding formation and regulating models of segregation were also proposed.However,the cooling rate of the melt is reduced by the continuous increase of the stirring speed to 130 r/min,which results in the grain coarsening and lower homogeneity of the ingot.

A nanocomposite consisting of silica-coated magnetite and phenyl-functionalized graphene oxide for extraction of polycyclic aromatic hydrocarbon from aqueous matrices

In this study,graphene oxide was covalently immobilized on silica-coated magnetite and then modified with 2-phenylethylamine to give a nanocomposite of type Fe3O4@SiO2@GO-PEA that can be applied to the magnetic solid-phase extraction of polycyclic aromatic hydrocarbons（PAHs） from water samples.The resulting microspheres（Fe3O4@SiO2@GO-PEA） were characterized by Fourier transform-infrared spectroscopy（FT-IR）,scanning electron microscopy（SEM）,CHNS elemental analysis,and vibrating sample magnetometry（VSM） techniques.The adsorbent possesses the magnetic properties of Fe3O4 nanoparticles that allow them easily to be separated by an external magnetic field.They also have the high specific surface area of graphene oxide which improves adsorption capacity.Desorption conditions,extraction time,amount of adsorbent,salt concentration,and pH were investigated and optimized.Following desorption,the PAHs were quantified by gas chromatography with flame ionization detection（GC-FID）.The limits of detection（at an S/N ratio of 3） were achieved from 0.005 to0.1 μg/L with regression coefficients（R2） higher than 0.9954.The relative standard deviations（RSDs） were below 5.8%（intraday） and 6.2%（inter-day）,respectively.The method was successfully applied to the analysis of PAHs in environmental water samples where it showed recoveries in the range between 71.7%and 106.7%（with RSDs of 1.6%to 8.4%,for n = 3）.The results indicated that the Fe3O4@SiO2@GO-PEA microspheres had a great promise to extraction of PAHs from different water samples.

Effect of carbon nanotube on physical and mechanical properties of natural fiber/glass fiber/cement composites

The objective of this investigation was to introduce a cement-based composite of higher quality. For this purpose new hybrid nanocomposite from bagasse fiber,glass fiber and multi-wall carbon nanotubes（MWCNTs）were manufactured. The physical and mechanical properties of the manufactured composites were measured according to standard methods. The properties of the manufactured hybrid nanocomposites were dramatically better than traditional composites. Also all the reinforced composites with carbon nanotube, glass fiber or bagasse fiber exhibited better properties rather than neat cement.The results indicated that bagasse fiber proved suitable for substitution of glass fiber as a reinforcing agent in the cement composites. The hybrid nanocomposite containing10 % glass fiber, 10 % bagasse fiber and 1.5 % MWCNTs was selected as the best compound.

Recently the field of cholestasis has expanded enormously reflecting an improved understanding of the molecular mechanisms underlying bile secretion and its perturbation in chronic cholestatic disease. Novel anti-cholestatic therapeutic options have been developed for patients not favorably responding to ursodeoxycholic acid (UDCA), the current standard treatment for cholestatic liver disease. Important novel treatment targets now also include nuclear receptors involved in bile acid (BA) homoeostasis like farnesoid X receptor and G proteincoupled receptors e.g., the G-protein-coupled BA receptor “transmembrane G coupled receptor 5”. Fibroblast growth factor-19 and enterohepatic BA transporters also deserve attention as additional drug targets as does the potential treatment agent norUDCA. In this review, we discuss recent and future promising therapeutic agents and their potential molecular mechanisms in cholestatic liver disorders.

利用限制模压变形(CGP)法对AA6063铝合金进行塑性变形,改善其表面性能。结果发现,在第一道次后,合金的硬度值显著提高,硬度均匀性显著降低。而经后续的道次后,加工材料的硬度行为得到改善。此外,道次数增加后,在CGP第一道次中形成的细长晶粒逐渐转变为等轴晶粒。通过CGP工艺,样品的耐腐蚀性能提高,这是因为钝化膜的快速形成以及第二相和析出物形貌的变化阻碍它们的电化学反应,减少潜在的局部腐蚀位点。

目的：回顾1997／2007伊朗亚兹德地区的200例眼及附属器肿瘤的流行病学特征，确定肿瘤的人群分布及肿瘤位置。方法：回顾观察性病例报道。我们回顾1997／2007医院组织病理学确诊为眼眶肿瘤的病例，分析200例连续病例的年龄分布、病理学、肿瘤来源及肿瘤位置。结果：患者200例（男111例，女89例），其中110例（55．0％）为良性肿瘤，86例（43．0％）为恶性，4例（2．0％）为转移性肿瘤。119例（59．5％）为眼睑肿瘤，21例（10．5％）为眼眶，60例（30．0％）位于眼球。最常见的恶性肿瘤是基底细胞癌（BCC）（25．5％）、鳞状细胞癌（SCC）（6．0％）及黑素瘤（5．5％）。各种类型的痣（15．0％）、皮样囊肿（5．5％）、血管瘤（5．0％）是最常见的良性肿瘤。200例患者按年龄分为两组，18岁以上及18岁以下。18岁以下患者中，最常见的是良性肿瘤（90．1％），恶性肿瘤及转移性肿瘤分别占8．4％和1．5％。然而，18岁以上患者，常见的是恶性肿瘤（51．1％），良性肿瘤占47．1％，转移性肿瘤占1．8％。结论：眼眶肿瘤的病理学特征与患者的年龄及肿瘤在眼及附属器的位置有关。在进行活检及肿瘤切除术之前，患者的发病年龄和肿瘤位置可成为我们制定治疗方案的前提，也是预后评估的重要信息。
