To investigate the link between higher air temperature and ambulance transports due to heat stroke, especially in elderly subjects, using daily data in Sai-tama prefecture in August, 2010, Japan. We obtained daily data of air temperature in Saitama prefecture in August, 2010, Japan from Japan Meteorological Agency. Number of ambulance transports due to heat stroke was also used by Saitama prefecture official website. The effect of high air temperature on am-bulance transports due to heat stroke was evaluated. A total of 1688 ambulance transports due to heat stroke was observed in Saitama prefecture in August, 2010, Japan. Seven hundred and twenty one subjects (42.7%) were over the age of 65. Mean air temperature, the highest air temperature and day light hours were significantly associated with ambulance transports due to heat stroke in total subjects (mean air temperature: r = 0.749, p < 0.0001) and the over the age of 7. In addition, by separate analysis as classified by age groups, the correlation coefficient between ambulance transports due to heat stroke and mean air temperature was the highest in subjects with over 65. However, the lowest air temperature was not cor-related with ambulance transports due to heat stroke. Higher air temperature was closely associated with higher ambulance transports due to heat stroke using daily data in Saitama prefecture in August, 2010, Ja-pan, especially in elderly subjects.

Aims: In the Add-on Lantus®?to Oral Hypoglycaemic Agents 2 (ALOHA 2) Study in Japanese adults with type 2 diabetes mellitus (T2DM), data on the safety and efficacy of combination therapy with insulin glargine (Lantus®) and oral anti-hyperglycaemic drugs (OADs) including dipeptidyl peptidase-4 (DPP-4) inhibitors in a real-life setting were collected and analyzed. Methods: This postmarketing surveillance was a prospective, observational, 24-week study that complied with the pharmaceutical affairs law and the ministerial ordinance of “Good Post-Marketing Study Practice (GPSP)” in Japan. Safety, efficacy and patient-reported outcomes (PROs);patients’ satisfaction with treatment (DTSQs and DTSQc) and patients’ self-reported health (EQ-5D and EQ-VAS) of combination therapy of insulin glargine and OADs were evaluated. Results: A total of 2,630 patients were enrolled. Of the 2,602 patients in the safety analysis population, 161 patients experienced 175 cases of adverse drug reactions, and the major adverse drug reaction was hypoglycaemia (140 patients, 5.38%). Out of those with hypoglycaemia, 11 patients (0.42%) had severe hypoglycaemia and the incidence rate (episodes per patient-year) was 0.019. Basal supported oral therapy (BOT) with insulin glargine substantially reduced the HbA1c, FPG and 2 hour-PPG levels for 24 weeks by -1.61%, -54.4 mg/dL and -74.5 mg/dL respectively. The mean weight was increased, however the change was +0.50 kg. In addition, the treatment satisfaction scores of DTSQs (mean treatment satisfaction score increased 3.6 from baseline to last observation) and DTSQc, EQ-5D index scores and EQ-VAS scores were significantly improved. Conclusion: Insulin glargine and OADs combination therapy was suggested to be effective and well tolerated. Patients’ satisfaction with treatment and their self-reported health improved in spite of the addition of injections to oral agents. Sugarcane advanced variety trials are planted across several locations and harvested for several crop-years to determine genotype by environment interaction and yield stability. Previous studies describe methods for simultaneous screening for yield and stability but did not use parametric statistical tests for comparing genotypes. The objective of this study was to describe a parametric statistical method for simultaneous screening of sugarcane genotypes for yield and stability. Data from 26 crops were collected from trials established at five locations and harvested in the plant, first, second, third and fourth ratoon crops. The mixed procedure of SAS was used for data analysis. The intercept and slope were used to represent yield and stability, respectively. There were significant (P < 0.05) differences in yield and stability among the genotypes. Test genotypes were classified into groups of genotypes that produced high yield, or high stability or both. The method provides fast statistical tests for simultaneous screening for yield and stability. The method was also used to compare two genotypes, an application for variety choice at time of release.

Prevalence and characterization of supplement use among healthy children and adolescents in an urban Japanese city

Although there are numerous studies on the use of supplements in children and adolescents from western countries, only a small number of studies have been conducted in Asian countries. This study was planned to determine the use of supplements and their association with lifestyles in healthy Japanese children and adolescents. This is a community-based cross-sectional study conducted in a Japanese urban city, Nara, in 2008. A total of 681 children and adolescents aged between 3 and 17 years were enrolled. The parents were asked to complete a questionnaire on the use of supplements among their children. Demographic information, health and nutrition status of children as perceived by parents, sleep duration, and a family history of supplement use were also collected. A total of 20.4% of children and adolescents are presently using (in the past one month) or have used (in the past one year excluding the past one month) supplements. The types of supplements most commonly used were minerals, followed by vitamins and blueberry extracts. Univariate analysis indicated that supplement use was associated with older age, poor perception of health status, shorter sleep duration, and a positive family history of supplement use. Multiple logistic regression analysis showed that age and a positive family history affected supplement use. Given the ap-proximately 20% of prevalence of supplement use in Japanese children and adolescents, there is a need for further research to examine the supplements’ effectiveness and safety.

Objective The custom-homebuilding company, Cosmic Garden Co. Ltd., located in Okayama City, Japan was established in 1997 and uses specific natural ore powder （SNOP） in wall materials and surveys customers in order to improve allergic symptoms. Methods To investigate the biological effects of SNOP, patients with a pollen allergy were recruited to stay in a room surrounded by cloth containing SNOP （CCSNOP）, and their symptoms and various biological parameters were compared with those of individuals staying in a room surrounded by control non-woven cloth （NWC）. Each stay lasted 60 min. Before and immediately after the stay, a questionnaire regarding allergic symptoms, as well as POMS （Profile of Mood Status） and blood sampling, was performed. Post-stay minus pre-stay values were calculated and compared between CCSNOP and NWC groups. Results Results indicated that some symptoms, such as nasal obstruction and lacrimation, improved, and POMS evaluation showed that patients were calmer following a stay in CCSNOP. Relative eosinophils, non-specific Ig E, epidermal growth factor, monocyte chemotactic protein-I, and tumor necrosis factor-a increased following a stay in CCSNOP. Conclusion This ore powder improved allergic symptoms, and long-term monitoring involving 1 to 2 months may be necessary to fully explore the biological and physical effects of SNOP on allergic patients.

Internal Rotation of Cyclopentadienyl Rings in Ferrocene Derivatives

The relaxation time of several ferrocene derivatives was measured, and the internal rotation was discussed. For almost all the derivatives, the degree of the internal rotation was constant in spite of the different molecular weights. However, for (triphenylmethyl)ferrocene, the rotation of the unsubstituted ring would be slower due to the bulkiness of the substituent. Furthermore, the derivatives that have a hydroxyl- or acetyl group on the substituent were also discussed. Their rotation would be influenced by the location of these substituents.

We investigated the link between heat stroke and high air temperature from July to September in 2010 in Fukushima prefecture, Japan. Daily data of ambulance transports stratified by heat stroke and the highest air temperature were obtained. Heat stroke was significantly correlated with the highest temperature. The 9.0 magnitude earthquake and following tsunami on March 11, 2011, destroyed many cities in the northwestern part of Japan. Taken together, heat stroke may dramatically increase in Fukushima prefecture, Japan in 2011.

Characterization of macrophage mutants established by their resistance to LPS and cycloheximide-induced apopotic cell death

Macrophages are activated by bacterial lipopolysaccharide (LPS) to produce inflammatory cytokines such as TNF-α or reactive oxygen species such as nitric oxide or superoxide anion. However, in the presence of an inhibitor of protein synthesis, cyclohex-imide (CHX), at 10 μg/mL, LPS at 100 ng/mL induced macrophage apoptosis rapidly without producing phenotypes of activated macrophages. In order to understand the mechanism underlying LPS-induced cytotoxicity toward macrophages, we isolated mutant cells from a macrophage-like cell line, J774.1, as clones resistant against the cytotoxic effects of LPS + CHX by using a somatic cell genetics protocol. All of the mutant clones, designated as LCR mutants, showed resistance to the cell death induced by LPS + CHX as well as to that induced by higher doses of LPS alone, as did the LPS1916 mutant cell line, which had been previously established by its resistance to 100 μg/mL LPS. Characterization of the activated macrophage phenotypes revealed that these mutants showed reduced production of TNF-α and nitric oxide in response to LPS. Further analysis showed a much reduced amount of [125I]LPS-binding and lower CD14 expression on the cell surface, in spite of an adequate intracellular expression of CD14 molecules. Besides, the molecular weight of CD14 on these mutants was around 40-48 kDa, smaller than that of the wild-type JA-4 cells (around 50-55 kDa), suggesting impaired CD14 maturation in these mutants. However, expression of Toll-like receptor 4 (TLR4) and Myd 88 on the cell surface was not different between the wild type and the mutant cells. These results suggest that LCR mutants have common phenotypes of mal-expression of CD14 molecules on the macrophage cell surface, leading to not only reduced responses to LPS-mediated macrophage activation but acquisition of resistance to LPS-induced apoptotic cell death in the presence of CHX.

Insulin resistance, closely linked to inflammation, is recognized as a key factor in the onset and aggravation of diabetes, cardio-renal syndrome, hypertension, and obesity. In the renal proximal tubule, insulin resistance may increase renal sodium reabsorption, leading to hypertension, edema and sometimes heart failure. Recently some anti-diabetic agents have been shown to have effects on the transporters in renal proximal tubule. Because renal proximal tubule mediates about 70% of sodium reabsorption, it is quite important to clarify the function of renal proximal tubule under insulin resistance and inflammation.
