OsIDD2, a zinc finger and INDETERMINATE DOMAIN protein, regulates secondary cell wall formation

Previously, we found 123 transcription factors（TFs） as candidate regulators of secondary cell wall（SCW）formation in rice by using phylogenetic and co-expression network analyses. Among them, we examined in this work the role of OsIDD_2, a zinc finger and indeterminate domain（IDD） family TF. Its overexpressors showed dwarfism, fragile leaves, and decreased lignin content, which are typical phenotypes of plants defective in SCW formation, whereas its knockout plants showed slightly increased lignin *** RNA-seq and quantitative reverse transcription polymerase chain reaction analyses confirmed that some lignin biosynthetic genes were downregulated in the OsIDD_2-overexpressing plants, and revealed the same case for other genes involved in cellulose synthesis and sucrose metabolism. The transient expression assay using rice protoplasts revealed that OsIDD_2 negatively regulates the transcription of genes involved in lignin biosynthesis, cinnamyl alcohol dehydrogenase 2 and 3（CAD_2 and 3）, and sucrose metabolism, sucrose synthase 5（SUS_5）, whereas an Alpha Screen assay, which can detect the interaction between TFs and their target DNA sequences, directly confirmed the interaction between OsIDD_2 and the target sequences located in the promoter regions of CAD_2 and CAD_3. Based on these observations, we conclude that OsIDD_2 is negatively involved in SCW formation and other biological events by downregulating its target genes.

Analysis of aristolochic acids, aristololactams and their analogues using liquid chromatography tandem mass spectrometry

More than 80 aristolochic acids(AAs) and aristololactams(ALs) have been found in plants of the Aristolochiaceae family, but relatively few have been fully studied. The present study aimed at developing and validating a liquid chromatography tandem mass spectrometry(LC/MS^n) for the analysis of these compounds. We characterized the fragmentation behaviors of 31 AAs, ALs, and their analogues via high performance liquid chromatography coupled with electrospray ionization mass spectrometry. We summarized their fragmentation rules and used these rules to identify the constituents contained in Aristolochia contorta, Ar. debilis, Ar. manshurensis, Ar. fangchi, Ar. cinnabarina, and Ar. mollissima. The AAs and ALs showed very different MS behaviors. In MS1 of AAs, the characteristic pseudomolecular ions were [M + NH_4]^+, [M + H]^+, and [M + H - H_2O]^+. However, only [M + H]^+ was found in the MS1 of ALs, which was simpler than that of AAs. Distinct MSn fragmentation patterns were found for AAs and ALs, showing the same skeleton among the different substituent groups. The distribution of the 31 constituents in the 6 species of Aristolochia genus was reported for the first time. 25 Analogues of AAs and ALs were detected in this genus. A hierarchical schemes and a calculating formula of the molecular formula of these nitrophenanthrene carboxylic acids and their lactams were proposed. In conclusion, this method could be applied to identification of similar unknown constituents in other plants.

Worldwide,welfare is trending from a welfare state model to social participation to ensure the sustainability of health care ***,where the aging rate will reach 40%by 2060,reformed public Long-Term Care Insurance(LTCI)on a large scale in 2015 and started the Comprehensive Service project(CS project),which emphasizes the prevention of long-term care through social participation in the *** this project,communities work to develop prevention efforts revolving around the community salon/caféand mutual support,including all daily tasks that previously were the purview of family *** municipality has an obligation to promote a CS project through new community development initiatives among community *** purpose of this study is to investigate the CS projects performed by rural municipalities and to explore the important factors for their *** authors used a case study methodology and constant comprehensive methodology for analyzing interview data to pull out the elements of successful *** study shows that the following factors influence success:recognition of demographic risk,recognition of the ineffectiveness of LTCI services,developing a passionate philosophy and strategy as a municipality,developing community members’initiative,and having many assets and full participation in the *** factors imply that community development is a critical part of any successful CS project.

Associations among pain catastrophizing, muscle strength, and physical performance after total knee and hip arthroplasty

AIM To investigate whether reductions in pain catastrophizing associated with physical performance in the early period after total knee arthroplasty(TKA) or total hip arthroplasty(THA).METHODS The study group of 46 participants underwent TKA or THA. The participants were evaluated within 7d before the operation and at 14d afterwards. Physical performance was measured by the Timed Up and Go(TUG) test, and 10-m gait time was measured at comfortable and maximum speeds. They rated their knee or hip pain using a visual analog scale(VAS) for daily life activities. Psychological characteristics were measured by the Pain Catastrophizing Scale(PCS). Physical characteristics were measured by isometric muscle strength of knee extensors and hip abductors on the operated side. The variables of percent changes between pre-and postoperation were calculated by dividing post-operation score by pre-operation *** Postoperative VAS and PCS were better than preoperative for both TKA and THA. Postoperative physical performance and muscle strength were poorer than preoperative for both TKA and THA. The percent change in physical performance showed no correlation with preoperative variables. In TKA patients, the percent change of PCS showed correlation with percent change of TUG(P = 0.016), 10-m gait time at comfortable speeds(P = 0.003), and 10-m gait time at maximum speeds(P = 0.042). The percent change of muscle strength showed partial correlation with physical *** percent change of VAS showed no correlation with physical performances. On the other hand, in THA patients, the percent change of hip abductor strength showed correlation with percent change of TUG(P =0.047), 10-m gait time at comfortable speeds(P = 0.001),and 10-m gait time at maximum speeds(P = 0.021). Effect of Grains of Paradise (GP) Extract Intake on Obesity and Sympathetic Nerve Activity

The methanol extract of grains of paradise (GP), the seed of Aframomum melegueta, which is distributed throughout West Africa, was administered during an animal breeding test. The extract suppressed body weight gain and decreased the weight of adipose tissues in breeding mice, with a greater effect on mice fed a high-fat diet (HFD) than on those fed a normal diet (ND). Other significant effects of GP intake included increased serum triglyceride (TG) concentration and reduced hepatic total cholesterol (TC) and TG concentrations. GP intake markedly prevented fat accumulation and improved hepatic lipid metabolism in HFD-fed mice. In addition, GP extract at a dosage of 5 mg/kg body weight decreased sympathetic nerve activity (SNA) in brown adipose tissue (BAT), while capsaicin, a major component of chili pepper, activated BAT SNA. This suggested that GP exerts a potential anti-obesity effect by a different mechanism from that of capsaicin.

Automatic evaluation of speech impairment caused by wearing a dental appliance

In dentistry, speech evaluation is important for appropriate orofacial dysfunction rehabilitation. The speech intelligibility test is often used to assess patients’ speech, and it involves an evaluation by human listeners. However, the test has certain shortcomings, and an alternative method, without a listening procedure, is needed. The purpose of this study was to test the applicability of an automatic speech intelligibility test system using a computerized speech recognition technique. Speech of 10 normal subjects, when wearing a dental appliance, was evaluated using an automatic speech intelligibility test system that was developed using computerized speech recognition software. The results of the automatic test were referred to as the speech recognition scores. The Wilcoxon signed rank test was used to analyze differences in the results of the test between the following 2 conditions: with the palatal plate in place and with the palatal plate removed. Spearman correlation coefficients were used to evaluate whether the speech recognition score correlated with the result of conventional intelligibility test. The speech recognition score was significantly decreased when wearing the plate (z = -2.807, P = 0.0050). The automatic evaluation results positively correlated with that of conventional evaluation when wearing the appliance (r = 0.729, P = 0.017). The automatic speech testing system may be useful for evaluating speech intelligibility in denture wearers.

Preparation of Y(Mn1−xFex)O3and Electrical Properties of the Sintered Bodies

YMnO3 and the Fe doped samples were prepared and the lattice constants, electrical conductivities and Seebeck coefficients were measured. The solubility limit of Fe for Mn of YMnO3 was 25 atomic %, the electrical conductivity increased in the range of 350°C - 700°C as Fe increased up to 10%, and the doped samples showed larger Seebeck coefficient than undoped samples.
