A deeper understanding of the biological events occurring when bioprocess parameters changed will be of great value in improving the monoclonal antibodies (mAbs) production. Design of experiment (DoE) was applied to investigate the effect of process parameters (pH, temperature shift and dissolve oxygen (DO)) on protein titer. The key metabolites connecting the critical process parameters (CPPs) with monoclonal antibody production were identified by different chemometrics tools. Finally, the biological events of marker metabolites relating with titer improvement were concluded. pH and temperature shift were identified as CPPs that affect the target protein titer. A series of metabolites influenced by the altered CPPs and correlated with protein titer were screened by principal component analysis (PCA) and Pearson' correlation test. The marker metabolites and their pathways linking CPPs to target protein titer in different culture phases were summarized. Metabolomics and chemometrics are promising data-driven tools to shine light into the biological black box between the bioprocess parameters and process performance.

目的利用锥形束CT(cone-beam computed tomography,CBCT)探讨上颌尖牙阻生与蝶鞍形态变异的相关性,从三维方向上评价蝶鞍变异对上颌尖牙阻生的临床预判。方法收集125例上颌阻生尖牙患者(研究组)和125例与之相匹配的尖牙正常萌出者(对照组)。将所有样本CBCT数据导入Dolphin软件,进行重建头颅侧位片和三维图像,记录观察各组蝶鞍桥接(Ⅰ、Ⅱ、Ⅲ型)的类型分布和发生情况。使用SPSS 25.0软件对各组结果进行统计学分析。结果研究组蝶鞍桥接的发生率显著高于对照组(P=0.004),PIC(腭侧尖牙阻生)组蝶鞍桥接的发生率显著高于对照组(P=0.007),Ⅱ型和Ⅲ型蝶鞍桥接分布和对照组存在统计学差异(P=0.012)。而BIC(唇侧尖牙阻生)组和对照组蝶鞍桥接的发生情况无统计学差异。三维重建图像上各组间蝶鞍桥接发生率无统计学差异(P>0.05)。结论上颌尖牙阻生与蝶鞍桥接的发生情况有相关性,但仅上颌尖牙腭侧阻生患者更易发生蝶鞍桥接,而上颌尖牙唇侧阻生患者蝶鞍桥接的发生情况无变化。

FedDAA:a robust federated learning framework to protect privacy and defend against adversarial attack

Federated learning(FL)has emerged to break data-silo and protect clients’privacy in the field of artificial intelligence.However,deep leakage from gradient(DLG)attack can fully reconstruct clients’data from the submitted gradient,which threatens the fundamental privacy of FL.Although cryptology and differential privacy prevent privacy leakage from gradient,they bring negative effect on communication overhead or model performance.Moreover,the original distribution of local gradient has been changed in these schemes,which makes it difficult to defend against adversarial attack.In this paper,we propose a novel federated learning framework with model decomposition,aggregation and assembling(FedDAA),along with a training algorithm,to train federated model,where local gradient is decomposed into multiple blocks and sent to different proxy servers to complete aggregation.To bring better privacy protection performance to FedDAA,an indicator is designed based on image structural similarity to measure privacy leakage under DLG attack and an optimization method is given to protect privacy with the least proxy servers.In addition,we give defense schemes against adversarial attack in FedDAA and design an algorithm to verify the correctness of aggregated results.Experimental results demonstrate that FedDAA can reduce the structural similarity between the reconstructed image and the original image to 0.014 and remain model convergence accuracy as 0.952,thus having the best privacy protection performance and model training effect.More importantly,defense schemes against adversarial attack are compatible with privacy protection in FedDAA and the defense effects are not weaker than those in the traditional FL.Moreover,verification algorithm of aggregation results brings about negligible overhead to FedDAA.

Sweet osmanthus(Osmanthus fragrans) is a having general approval aromatic tree in China that is widely applied to landscaping and gardening. However, the evergreen tree adaptability is limited by many environmental stresses. Currently, limited information is available regarding the genetic analysis and functional identification of expansin genes in response to abiotic stress in sweet osmanthus. In this study, a total of 29 expansin genes were identified and divided into four groups by genome-wide analysis from the sweet osmanthus genome. Transcriptome and quantitative Real-time PCR analysis showed that the cell wall-localized protein expansin-like A(OfEXLA1) gene was significantly induced by salt and drought treatment. Histochemical GUS staining of transgenic Arabidopsis lines in which GUS activity was driven with the OfEXLA1 promoter, GUS activity was significantly induced by salt, drought, and exogenous abscisic acid(ABA). In yeast, we found OfEXLA1overexpression significantly improved the population of cells compared with wild-type strains after NaCl and polyethylene glycol(PEG)treatment. Additionally, OfEXLA1 overexpression not only promoted plant growth, but also improved the salt and drought tolerance in Arabidopsis. To gain insight into the role of ABA signaling in the regulation of OfEXLA1 improving abiotic tolerance in sweet osmanthus, four differentially expressed ABA Insensitive 5(ABI5)-like genes(OfABL4, OfABL5, OfABL7, and OfABL8) were identified from transcriptome, and dualluciferase(dual-LUC) and yeast one hybrid(Y1H) assay showed that OfABL4 and OfABL5 might bind to OfEXLA1 promoter to accumulate the OfEXLA1 expression by responding to ABA signaling to improve abiotic tolerance in sweet osmanthus. These results provide the information for understanding the molecular functions of expansin-like A gene and molecular breeding of sweet osmanthus in future.

Covalent organic frameworks(COFs)are an emerging type of porous crystalline polymers formed by combining strong covalent bonds with organic building blocks.Due to their large surface area,high intrinsic pore space,good crystallization properties,high stability,and designability of the resultant units,COFs are widely studied and used in the fields of gas adsorption,drug transport,energy storage,photoelectric catalysis,electrochemistry,and sensors.In recent years,the rapid development of the Internet of Things and people’s yearning for a better life have put forward higher and more requirements for sensors,which are the core components of the Internet of Things.Therefore,this paper reviews the recent progress of COFs in synthesis methods and sensing applications,especially in the last five years.This paper first introduces structure,properties,and synthesis methods of COFs and discusses advantages and disadvantages of different synthesis methods.Then,the research progress of COFs in different sensing fields,such as metal ion sensors,gas sensors,biomedical sensors,humidity sensors,and pH sensors,is introduced systematically.Conclusions and prospects are also presented in order to provide a reference for researchers concerned with COFs and sensors.

Straw addition increases enzyme activities and microbial carbon metabolism activities in bauxite residue

Recovery of microbial functions is one of the critical processes in the nutrient cycling of bauxite residue for improving revegetation.Straw is considered to be effective to increase microbial diversity and drive the development of the microbial community,but its effect on microbial carbon metabolism has not been illustrated.The present study evaluated the effects of phosphogypsum(PG),straw(SF)and phosphogypsum plus straw(PGSF)on physicochemical properties,enzyme activities,and microbial carbon metabolism activities in bauxite residue.After 180 days incubation,PG,SF and PGSF treatment significantly reduced the residue pH from 10.85 to 8.64,9.39 and 8.06,respectively.Compared to CK treatment,SF treatment significantly increased the content of total organic carbon(TOC)and organic carbon fractions(DOC,MBC,EOC,and POC).In addition,straw addition significantly increased glucosidase,cellulose,urease,and alkaline phosphatase by 7.2-9.1 times,5.8-7.1 times,11.1-12.5 times,and 1.1-2.2 times,respectively.The Biolog results showed that straw addition significantly increased microbial metabolic activity(AWCD)and diversity in bauxite residue.Redundancy analysis indicated total nitrogen(TN)and carbon fractions(POC,MBC and DOC)were the most important environmental factors affecting microbial metabolic activity and diversity in bauxite residue.These findings provided us with a biogeochemical perspective to reveal soil formation in bauxite residue and suggested that nutrient supplement and regulation of salinity-alkalinity benefit the establishment of microbial communities and functions in bauxite residue.

This paper investigates the design of an attitude autopilot for a dual-channel controlled spinning glideguided projectile(SGGP),addressing model uncertainties and external disturbances.Based on fixed-time stable theory,a disturbance observer with integral sliding mode and adaptive techniques is proposed to mitigate total disturbance effects,irrespective of initial conditions.By introducing an error integral signal,the dynamics of the SGGP are transformed into two separate second-order fully actuated systems.Subsequently,employing the high-order fully actuated approach and a parametric approach,the nonlinear dynamics of the SGGP are recast into a constant linear closed-loop system,ensuring that the projectile's attitude asymptotically tracks the given goal with the desired eigenstructure.Under the proposed composite control framework,the ultimately uniformly bounded stability of the closed-loop system is rigorously demonstrated via the Lyapunov method.Validation of the effectiveness of the proposed attitude autopilot design is provided through extensive numerical simulations.

Analysis of light propagation characteristic in the aero-optic flow field of cone-headed vehicle with side window

Aero-optic imaging deviation research is carried out for infrared-guided vehicle with cone-head side window,with a focus on the propagation characteristics of light in an aero-optic flow field.When the light entering the aero-optic flow field from the free-stream should be close to the normal,numerous data indicate that the light is refracted away from the normal.This paper divides the aero-optic flow field into two parts and uses the gas density distribution in the aero-optic flow field to propose the hypothesis that there are two modes of refraction when light propagates through the flow field.The results show that light propagates from the optically denser medium to the optically thinner medium after passing through the shock wave and eventually leads to refraction away from the normal when the light enters the aero-optic flow field.

This paper proposes a new method for service restoration of distribution network with the support of transportable power sources(TPSs)and repair crews(RCs).Firstly,a coupling model of distribution networks and vehicle routing of TPSs and RCs is proposed,where the TPSs serve as emergency power supply sources,and the RCs are used to repair the faulted lines.Considering the uncertainty of traffic congestion,the probability distribution of the travel time spent on each road is derived based on the Nesterov user equilibrium model,and a two-stage stochastic program is formulated to determine the optimal routings of TPSs and RCs.To efficiently solve the proposed stochastic mixed-integer linear program(MILP),a two-phase scenario reduction method is then developed to scale down the problem size,and an adaptive progressive hedging algorithm is used for an efficient solution.The effectiveness of the proposed methods and algorithms has been illustrated in a modified IEEE 33-bus system.

In the preparation of large-sized ceramics,the use of a green body with relatively high plasticity is crucial to minimize the risk of cracking during processing.To achieve this goal,glycerol and polyethylene glycol(PEG)were utilized as plasticizers in the shaping of green bodies of oxide ceramics through spontaneous coagulation casting(SCC).This study investigated the effects of plasticizers and particle sizes ranging from the submicron to nanoscale on the slurry viscosity,drying shrinkage of wet gels,and mechanical properties of green bodies.The plasticity of the green bodies was assessed by measuring the impact toughness and flexural stress‒strain curves.By incorporating an appropriate plasticizer,the peak width of the flexural stress‒strain curve for dried green bodies from particles of different sizes was nearly twice that without plasticizers,and the impact toughness was enhanced by approximately 71%,34%,and 41%when the particle size decreased from the submicron scale to the nanoscale(0.45μm,0.18μm,and 50 nm,respectively).The drilling test revealed that there was nearly no cracking around the holes in the green bodies with plasticizers.The plasticity mechanism of the green bodies was examined based on ultraviolet–visible(UV–Vis)spectroscopy and scanning electron microscopy(SEM).It was discovered that plasticizers might mitigate the brittleness of green bodies by adjusting the interactions between molecules and modifying the gel network properly.
