miRNAs are endogenous small RNAs that are important regulators of gene expression.miR-1294 was found to be significantly down-regulated in 15 cancers and regulated by 21 upstream regulators.miR-1294 affects the proliferation,migration,invasion,and apoptosis of cancer cells.The target genes of miR-1294 are involved in the PI3K/AKT/mTOR,RAS,and JAK/STAT signaling pathways.Six target genes of miR-1294 are the targets of a variety of drugs.Low expression of miR-1294 is associated with resistance to cisplatin and TMZ and a poorer prognosis in patients with ESCC,GC,EOC,PDAC,or NSCLC.Therefore,this work outlines the molecular mechanisms and provides a basis for the clinical significance of the tumor suppressor miR-1294 in cancer.

PRKCDBP Methylation is a Potential and Promising Candidate Biomarker for Non-small Cell Lung Cancer

Background and objectives:The occurrence and development of lung cancer are closely linked to epigenetic modification.Abnormal DNA methylation in the CpG island region of genes has been found in many cancers.Protein kinase C delta binding protein(PRKCDBP) is a potential tumor suppressor and its epigenetic changes are found in many human malignancies.This study investigated the possibility of PRKCDBP methylation as a potential biomarker for non-small cell lung cancer(NSCLC).Methods:We measured the methylation levels of PRKCDBP in the three groups of NSCLC tissues.Promoter activity was measured by the dual luciferase assay,with S’-aza-deoxycytidine to examine the effect of demethylation on the expression level of PRKCDBP.Results:The methylation levels of PRKCDBP in tumor tissues and 3 cm para-tumor were higher than those of distant(>10 cm)non-tumor tissues.Receiver operating characteristic(ROC) curve analysis between tumor tissues and distant non-tumor tissues showed that the area under the line(AUC) was 0.717.Dual luciferase experiment confirmed that the promoter region was able to promote gene expression.Meanwhile,in vitro methylation of the fragment(PRKCDBP;e) could significantly reduce the promoter activity of the fragment.Demethylation of 5’-aza-deoxycytidine in lung cancer cell lines A549 and H1299 showed a significant up-regulation of PRKCDBP mRNA levels.Conclusion:PRKCDBP methylation is a potential and promising candidate biomarker for non-small cell lung cancer.

It is generally believed that trondhjemitic rock, an important component of TTG rocks, is the anatectic product of mafic rocks. However, in many TTG gneiss terranes, for instance, the granulite facies terrane in Eastern Hebei, trondhjemites occur as small dikes, intrusions or leucosomes in tonalitic gneisses, suggesting their origin of in-situ partial melting. Based on the petrological analysis of a tonalitic gneiss sample from Eastern Hebei, in combination with zircon U-Pb dating, we investigated the petrogenesis of trondhjemite through simulating anatectic reactions and the major and trace element characteristics of the product melt at different pressures(0.7, 1.0 and 2.0 GPa). The results indicate that hornblende dehydration melting in a tonalitic gneiss at 0.9–1.1 GPa and 800–850°C, corresponding to the high-T granulite facies, with melting degrees of 5–10wt.% and a residual assemblage containing 5–10wt.% garnet, can produce felsic melts with a great similarity, for instance of high La/Yb ratios and low Yb contents to the trondhjemitic rocks from Eastern Hebei. However, the modelled melts exhibit relatively higher K2 O, and lower CaO and Mg~# than those in the trondhjemitic dikes and leucosomes from Eastern Hebei, suggesting that the leucosomes may not only contain some residual minerals but also be subjected to the effect of crystal fractionation. The zircon U-Pb dating for the tonalitic and trondhjemitic rocks in the Eastern Hebei yields a protolith age of 2518±12 Ma and a metamorphic age of 2505±19 Ma for the tonalitic gneiss. The latter age is consistent with a crystallization age of 2506±6 Ma for the trondhjemitic rock, confirming a close petrogenetic relation between them.

Microstructural Evolution during Tensile Deformation in TRC-ZA21 Magnesium Alloy with Different Loading Directions and Strain Rates

The variation of texture characteristics and activation of deformation mechanism in magnesium alloys can be achieved by addition of RE and Ca elements and subsequently affect the microstructure evolution during deformation process.In this manuscript,the effects of loading direction and strain rate on mechanical properties,microstructural characteristics,texture evolution and deformation mechanism in TRC-ZA21 magnesium alloy sheet were investigated.Moreover,orientation dependence and strain rate sensitivity of deformation mechanism were also discussed.The results showed that evident difference in mechanical properties in TRC-ZA21 alloy exhibited by the changes in loading direction and strain rate.The variations in grain orientation and basal texture characteristic were attributed to the grain rotation behavior during the plastic deformation,dominated by deformation mechanism.Basal slip,extension twinning and prismatic slip played different contributions to plastic deformation behavior and presented orientation dependence and strain rate sensitivity.The activities of prismatic slip and extension twinning both enhance with increasing strain rate.The phenomenon of weakened effect of twinning and promoted role of prismatic slip was presented to coordinate strain compatibility during plastic deformation.

A highly sensitive fluorescent probe for tracking intracellular zinc ions and direct imaging of prostatic tissue in mice

A highly sensitive fluorescent sensor ZnDN was designed,synthesized and used for tracking intracellular zinc ions in various living cells and direct imaging of prostatic tissue in mice.ZnDN was prepared from the heterocyclic-fused naphthalimide fluorophore,and the zinc receptor,N,N-bis(2-pyridylmethyl)ethylenediamine(BPEN).Upon addition of Zn2+to the solutions of ZnDN,a remarkable fluorescence enhancement was observed,which could be attributed to the photo-induced electron transfer(PET)mechanism.Since ZnDN exhibited high sensitivity toward Zn2+in phosphate buffer solution,with a limit of detection of 4.0 x 10-9 mol/L,it was further applied for the imaging of exogenous and endogenous Zn21 in different living cells.Living cells imaging experiments suggested that ZnDN could image the changes of intracellular free zinc ions,and could be used for two-photon imaging.Moreover,flow cytometry suggested that ZnDN could distinguish cancerous prostate cells from normal cells.Animal experiments indicated that ZnDN had the potential in imaging prostate tissue in vivo.

Increased Expression of Colonic Mucosal Melatonin in Patients with Irritable Bowel Syndrome Correlated with Gut Dysbiosis

Dysregulation of the gut microbiota/gut hormone axis contributes to the pathogenesis of irritable bowel syndrome(IBS).Melatonin plays a beneficial role in gut motility and immunity.However,altered expression of local mucosal melatonin in IBS and its relationship with the gut microbiota remain unclear.Therefore,we aimed to detect the colonic melatonin levels and microbiota profiles in patients with diarrhea-predominant IBS(IBS-D)and explore their relationship in germ-free(GF)rats and BON-1 cells.Thirty-two IBS-D patients and twenty-eight healthy controls(HCs)were recruited.Fecal specimens from IBS-D patients and HCs were separately transplanted into GF rats by gavage.The levels of colon mucosal melatonin were assessed by immunohistochemical methods,and fecal microbiota communities were analyzed using 16S rDNA sequencing.The effect of butyrate on melatonin synthesis in BON-1 cells was evaluated by ELISA.Melatonin levels were significantly increased and negatively correlated with visceral hypersensitivity in IBS-D patients.GF rats inoculated with fecal microbiota from IBS-D patients had high colonic melatonin levels.Butyrate-producing Clostridium cluster XIVa species,such as Roseburia species and Lachnospira species,were positively related to colonic mucosal melatonin expression.Butyrate significantly increased melatonin secretion in BON-1 cells.Increased melatonin expression may be an adaptive protective mechanism in the development of IBS-D.Moreover,some Clostridium cluster XIVa species could increase melatonin expression via butyrate production.Modulation of the gut hormone/gut microbiota axis offers a promising target of interest for IBS in the future.

Long noncoding RNAs(lncRNAs)are a class of nonprotein-coding transcripts that are longer than 200 nucleotides.LINC00355 is a lncRNA located on chromosome 13q21.31 and is consistently upregulated in various cancers.It regulates the expression of downstream genes at both transcriptional and posttranscriptional levels,including eight microRNAs(miR-15a-5p,miR-34b-5p,miR-424-5p,miR-1225,miR-217-5p,miR-6777-3p,miR-195,and miR-466)and three protein-coding genes(ITGA2,RAD18,and UBE3C).LINC00355 plays a role in regulating various biological processes such as cell cycle progression,proliferation,apoptosis,epithelial-mesenchymal transition,invasion,and metastasis of cancer cells.It is involved in the regulation of the Wnt/β-catenin signaling pathway and p53 signaling pathway.Upregulation of LINC00355 has been identified as a high-risk factor in cancer patients and its increased expression is associated with poorer overall survival,recurrence-free survival,and disease-free survival.LINC00355 upregulation has been linked to several unfavorable clinical characteristics,including advanced tumor node metastasis and World Health Organization stages,reduced Karnofsky Performance Scale scores,increased tumor size,greater depth of invasion,and more extensive lymph node metastasis.LINC00355 induces chemotherapy resistance in cancer cells by regulating five downstream genes,namely HMGA2,ABCB1,ITGA2,WNT10B,and CCNE1 genes.In summary,LINC00355 is a potential oncogene with great potential as a diagnostic marker and therapeutic target for cancer.

Electrical properties of yttrium calcium oxyborate crystal annealed at high temperature and low oxygen partial pressure

The yttrium calcium oxyborate crystal(YCa_(4)O(BO_(3))_(3),YCOB)has been actively studied for hightemperature piezoelectric sensing applications.In this work,the stability of electric properties of YCOB crystal annealed in critical conditions(high-temperatures of 900-1100℃ with a low oxygen partial pressure of 4×10^(-6) atm for 24 h)was investigated and the recovery mechanism for the electrical resisitivity,dielectric permittivity and dielectric loss were studied,taking advantage of the X-ray photoelectron spectra and the first principle calculations.The electrical resistivity of the annealed YCOB crystal was slightly decreased when compared to the pristine counterpart,being(2-5)×10^(7) Ω·cm at 850C.The dielectric permittivity and dielectric loss were found to increase after annealing,showing recoverable behaviours after thermal treatment above 650℃ in air.The calculated vacancy formation energy indicates that the oxygen vacancy is the dominant defects in YCOB.The formation of oxygen vacancy weakens the chemical bonding strength between B(Ca or Y)and O atoms,introduces extra donor levels in the band gap,which excites the electrons to conduction band more easily thus enhances the electrical conductivity and dielectric loss.The recovered electrical properties are believed to be associated with the reduced vacancy defects at elevated temperatures in air.
