Enzymatically prepared alginate oligosaccharides improve broiler chicken growth performance by modulating the gut microbiota and growth hormone signals

Background Alginate oligosaccharide(AOS)holds great potential as a novel feed supplement in farm ***,the effects of AOS on chicken health and the underlying mechanisms are not fully *** study aimed to optimize the enzymatic preparation of AOS by using bacterial alginate lyases expressed in yeast,investigate the effects of the prepared AOS on the growth performance and gut health of broiler chickens,and reveal the underlying *** Five alginate lyases from bacteria were cloned into Pichia pastoris GS115 and the alginate lyase PDE9 was expressed at relatively high yield,activity and stability in *** trials were carried out using 3201-day-old male Arbor Acres broilers(four groups;8 replicates/group×10 chicks/replicate)receiving either a basal diet or the same diet supplemented with 100,200 and 400 mg/kg PDE9-prepared AOS for 42 *** results showed that dietary supplementation of 200 mg/kg AOS displayed the highest activity in promoting the birds’ADG and ADFI(P<0.05).AOS ameliorated the intestinal morphology,absorption function and barrier function,as indicated by the enhanced(P<0.05)intestinal villus height,maltase activity,and the expression of PEPT,SGLT1,ZNT1,and *** also increased serum insulin-like growth factor-1,ghrelin(P<0.05),and growth hormone(P<0.1).Moreover,the concentrations of acetate,isobutyrate,isovalerate,valerate,and total SCFAs in cecum of birds fed AOS were significantly higher than the control birds(P<0.05).Metagenomic analysis indicated that AOS modulated the chicken gut microbiota structure,function,and microbial interactions and promoted the growth of SCFAs-producing bacteria,for example,Dorea sp.002160985;SCFAs,especially acetate,were found positively correlated with the chicken growth performance and growth-related hormone signals(P<0.05).We further verified that AOS can be utilized by Dorea *** grow and to produce acetate in *** We demonstrated that the enzymatically

Sequential pyrolysis for understanding specific influence of cellulose- and lignin-derived volatiles on properties of counterpart char

Interactions of cellulose-and lignin-derived intermediates have been well documented during pyrolysis of lignocellulosic *** reaction network for the interactions is rather complex,as cellulose-derived volatiles could interact/react with not only lignin-derived volatiles but also lignin-derived char and vice *** probe specifically the impacts of cellulose-derived volatiles on the lignin-derived char or the opposite,herein the sequential pyrolysis was performed by arranging cellulose in the upper bed with lignin in the lower bed or lignin above with cellulose below at 350 and 650℃,*** results indicated that the sequential pyrolysis of cellulose→lignin or lignin→cellulose at 350℃induced increased char yield from formation of carbonaceous deposits via volatiles-char *** with the lignin-derived volatiles,the cellulose-derived volatiles,especially aldehyde fractions,were more reactive towards the lignin-derived char at 350℃,forming oxygen-rich lignin-derived char with a higher yield,an abundance of aliphatic structures and consequently lower thermal *** sequential pyrolysis of lignin→cellulose,more aromatics-rich species were deposited on cellulose-derived char,but the lignin-derived volatiles were less reactive for enhancing the char *** 650℃,instead of polymerisation of the volatiles on the char,either the cellulose-or lignin-derived char catalyzed the cracking of the counterpart volatiles to remove the aliphatic functionalities,which made the char more aromatic and thermally more stable.

Micromorphological Analysis of Soil Structure under No Tillage Management in the Black Soil Zone of Northeast China

The structure of the ＂black soil＂ in Northeast China has been greatly deteriorated by long-term intensive conventional mouldboard plow tillage （CT） practices. In this study, micro- morphological observation and image analysis of soil thin sections were conducted to evaluate the impacts of 21 years （1986-2007） of no tillage （NT） on soil structure as compared to CT in an experiment near Gongzhuling City, Jilin Province. Soil organic matter （SOM）, wet aggregate stability and saturated hydraulic conductivity （Ks） were also analyzed. Total SOM was not significantly affected by tillage systems, but fresher SOM was observed in the surface layer under NT. The aggregates under NT showed different hierarchies in the form of crumbs, and the mean weight diameter （MWD） of NT was significant higher than that of CT in the surface layer. Platy and blocky aggregates were frequently observed in the lower layers under CT practice. The compound pore structure with intertwined intra- and inter- aggregates pores under NT was well developed in a layer from 0-5 cm to 20-25 era. While under CT system, more inter-aggregate pores and fewer intra- aggregate pores were observed, and planes and channels were frequently found in the 20-25 cm layer, where maeroporosity decreased significantly and a plow pan was evident. The Ks values of NT weresignificantly lower at o-5 cm but significantly higher at 20-95 cm compared with CT, which showed the same trend with macroporosity. These results confirmed that long-term CT practice fragmented the tillage layer soil and compacted the lower layer soil and formed a plow pan. While long-term NT practice in the black soil region favored soil aggregation and a stable porous soil structure was formed, which are important to the water infiltration and prevent soil erosion.
