Hypomyelination leukodystrophies constitute a group of heritable white matter disorders exhibiting defective myelin *** identified as a lysosomal protein,the TMEM106B D252N mutant has recently been associated with ***,how lysosomal TMEM106B facilitates myelination and how the D252N mutation disrupts that process are poorly *** used superresolution Hessian structured illumination microscopy(Hessian-SIM)and spinning discconfocal structured illumination microscopy(SD-SIM)to find that the wild-type TMEM106B protein is targeted to the plasma membrane,filopodia,and lysosomes in human *** D252N mutation reduces the size of lysosomes in oligodendrocytes and compromises lysosome changes upon starvation *** importantly,we detected reductions in the length and number of filopodia in cells expressing the D252N ***1 is the most abundant myelin protein that almost entirely colocalizes with TMEM106B,and coexpressing PLP1 with the D252N mutant readily rescues the lysosome and filopodia phenotypes of ***,interactions between TMEM106B and PLP1 on the plasma membrane are essential for filopodia formation and myelination in oligodendrocytes,which may be sustained by the delivery of these proteins from lysosomes via exocytosis.

Despite the wide application of super-resolution(SR)microscopy in biological studies of cells,the technology is rarely used to monitor functional changes in live *** combining fast spinning disc-confocal structured illumination microscopy(SD-SIM)with loading of cytosolic fluorescent Ca2+indicators,we have developed an SR method for visualization of regional Ca2+dynamics and related cellular organelle morphology and dynamics,termed SR calcium lantern *** COS-7 cells stimulated with ATP,we have identified various calcium macrodomains characterized by different types of Ca2+release from endoplasmic reticulum(ER)***,we demonstrated various roles of mitochondria in mediating calcium signals from different sources;while mitochondria can globally potentiate the Ca2+entry associated with store release,mitochondria also locally control Ca2+release from the neighboring ER stores and assist in their refilling processes.

In fluorescence microscopy,computational algorithms have been developed to suppress noise,enhance contrast,and even enable super-resolution(SR).However,the local quality of the images may vary on multiple scales,and these differences can lead to *** mapping methods fail to finely estimate the local quality,challenging to associate the SR scale ***,we develop a rolling Fourier ring correlation(rFRC)method to evaluate the reconstruction uncertainties down to SR *** visually pinpoint regions with low reliability,a filtered rFRC is combined with a modified resolution-scaled error map(RSM),offering a comprehensive and concise map for further *** demonstrate their performances on various SR imaging modalities,and the resulting quantitative maps enable better SR images integrated from different ***,we expect that our framework can become a routinely used tool for biologists in assessing their image datasets in general and inspire further advances in the rapidly developing field of computational imaging.

The application of high-performance lithium-sulfur(Li-S)batteries is severely influenced by the“shuttle effect”of polysulfides and the volume change of sulfur ***,two different polymeric binders SOT-A and SOT-C with three-dimensional network structure containing polar groups(sulfhydryl groups,amide groups and amino groups)are synthesized by the nucleophilic ring-opening polymerization(ROP)of thiolactone with amino *** network structure formed by hydrogen bonds and functional groups can resist the volume change of the *** sulfhydryl groups and the S-S bond formed by oxidative dehydrogenation of sulfhydryl group participate in the charge and discharge process of the battery as active materials,which improves the discharge specific capacity of the *** functional groups have strong chemisorption on polysulfides and effectively inhibit the“shuttle effect”.The electrochemical performances of Li-S batteries containing SOT-A and SOT-C binders are significantly *** 1 C rate,the batteries achieve initial discharge specific capacity of 871 and 837 mAh·g^(-1),respectively,and have 83.9%and 62.5%capacity retention after 500 cycles.

背景口服中药在慢性阻塞性肺疾病急性加重(AECOPD)的治疗中应用广泛,但相关临床试验的结局指标尚未得到统一和规范。目的通过筛选已发表的以口服中药为AECOPD治疗措施的随机对照试验(RCT),总结其文献特征和选用的结局指标情况,为中医药治疗AECOPD临床试验设计和结局指标选择提供参考。方法计算机检索中国知网、万方数据知识服务平台、维普网、中国生物医学文献数据库、PubMed、Embase、Web of Science、Cochrane Library、***、中国临床试验注册中心,获取口服中药治疗AECOPD的RCT和临床试验注册方案,检索时限为2018年1月—2022年10月。由2位评价员独立筛选文献、提取资料后,采用定性分析的方法,对纳入研究的结局指标选择情况进行描述。结果纳入578篇文献,包含574篇已报告试验结果的RCT及4个临床试验注册方案。574篇RCT共纳入51508例患者。88篇文献在纳入标准中限定了患者的疾病分级,361篇文献在纳入标准中限定了中医证型,6篇文献报告了盲法,6篇文献提及了随访。纳入文献共涉及4030个结局指标,单篇文献结局指标数量范围为1~24个。按照结局指标的功能属性,将其归为8个指标域:中医症状/证候、症状/体征、理化检测、生活质量、远期预后、经济学评估、安全性评价、其他,报告率最高的指标域是理化检测,报告频次排名前5位的结局指标项目是:有效率(11.5%)、第1秒用力呼气容积(7.5%)、中医症状/证候评分(7.0%)、第1秒用力呼气容积/用力肺活量(6.8%)、用力肺活量(4.6%)。445篇文献报告了有效率的组成,报告率排名前5位的研究指标依次为症状(423篇)、体征(281篇)、中医证候评分(203篇)、实验室检查(89篇)、肺功能(71篇)。结论口服中药治疗AECOPD的RCT涉及的结局指标数量多、范围广,纳入的文献在不同程度上关注了口服中药对AECOPD患者的症状体征、理化检测指标、生活质量、远期预后、经济学评价和安全性结局等方面的影响。但结局指标的选择仍存在多方面的问题:结局指标主次不清;主要关注替代终点,对临床终点的关注不足;对卫生经济学指标关注不足;有效性参考来源不一、判断标准不一。研究者可参考已发表的核心指标集,合理设计结局指标,以提高中医药临床研究的质量。

Microbial flocculant （MBF） is a kind of novel flocculant, which is widely used for the food industry, highly concentrated organic waste water treatment and the urban water supply technology. Due to its characteristics of superior flocculation effect, without secondary pollution to the environment and being easily degraded, MBF has been emerging as a hotspot of water treatment research in recent years. In this paper, the latest progress of the preparation of MBFs and the flocculating mechanism were presented. The practical application research of MBFs for water treatment was reviewed. Finally, suggestions and development prospect of research on MBFs were discussed.

裸燕麦膳食纤维组成含量研究及优异资源筛选

为了研究裸燕麦不同品种间膳食纤维组成、含量与营养品质的多样性,完善裸燕麦营养品质评价体系,筛选出优异资源,测定了24个裸燕麦品种籽粒的膳食纤维组成(β-葡聚糖、半纤维素、果胶、纤维素、低聚糖和抗性淀粉)和营养品质指标(蛋白质、淀粉、灰分和脂肪),并运用主成分分析和聚类分析方法对24个裸燕麦品种的营养品质进行综合评价。结果表明,β-葡聚糖和半纤维素占总膳食纤维的比例范围分别为31.2%~49.0%和25.8%~48.7%,是裸燕麦膳食纤维的主要组成成分。主成分分析结果表明,蛋白质、半纤维素、β-葡聚糖和果胶可作为裸燕麦营养品质评价的核心指标;品质综合评价前10名的燕麦品种分别为张莜8号、迪燕1号、白燕16号、坝莜13号、魏都莜5号、白燕2号、晋燕17号、坝莜16号、坝莜18号和白燕18号。聚类分析结果表明,24个裸燕麦品种可划分为5类,类群Ⅲ中品种的蛋白质、β-葡聚糖和脂肪含量最高,类群Ⅳ中品种的半纤维素含量较高。综合品质评价前十名的燕麦品种有1个来自类群Ⅲ,2个来自类群Ⅱ,有3个来自类群Ⅳ,有4个来自类群Ⅰ,这10个品种可用于改良燕麦籽粒品质性状的优异材料。

建立了同时测定连翘叶茶中连翘酯苷A、连翘苷、芦丁和连翘脂素的液相色谱方法。采用70%甲醇在70℃恒温水浴回流提取目标物,以0.3%醋酸水溶液-甲醇为流动相,选用Waters sunfire C18色谱柱在277 nm下进行液相色谱分析。该方法线性、精密度和回收率好,检出限和定量限低,适用于连翘叶茶中上述四种活性成分的测定。采用标准方法和自建方法对连翘绿茶(FGT)和连翘红茶(FBT)的营养成分进行了比较分析,其中可溶性糖含量较高(14.15%和15.02%),利于茶汤甘甜滋味及香气形成;FGT的水浸出物(50.62%)、维生素C(48.63 mg/100 g)、总多酚(11.06%)、黄酮(2.39%)、连翘酯苷A(8.77%)、连翘苷(4.98%)和芦丁含量(2.19%)显著高于FBT(p<0.05),其中特征成分连翘酯苷A和连翘苷含量远高于传统中药连翘(果);FBT虽特征成分含量低,但在发酵过程中生成了更易吸收和更强药理活性的连翘脂素(2.88%),含量远高于FGT(0.10%)。此外,FBT茶汤呈橙黄色,含有11.83 mg/100 g的茶黄素。综上,两种连翘叶茶均具有极高的药用价值,极具市场潜力。
