Apple polyphenol phloretin inhibits type II glucose transporter and enhances anti-HER2 antibody drug binding as an adjuvant treatment for HER2-positive breast cancer

This study presents novel findings on the potential of phloretin， an apple polyphenol， to enhance the effectiveness of anti-HER2 antibody therapy in HER2-positive breast cancer patients. Our research reveals that phloretin inhibits type II glucose transporter (GLUT2) activity， significantly reducing cancer cell glucose uptake. We confirmed the overexpression of GLUT1 and GLUT2 mRNA in paired human breast tumor tissues， with GLUT2 overexpression associated explicitly with poorer survival rates in breast cancer patients. Treatment with phloretin was observed to increase the interaction between GLUT2 and HER2 proteins， attenuate glycolysis， and enhance the binding affinity of anti-HER2 antibody drugs to target human breast cancer cells. Furthermore， the efficacy of the combination therapy involving phloretin and antibody drugs was reaffirmed in a cell-derived xenograft (CDX) tumor animal model， particularly in suppressing the growth of Trastuzumab-resistant HER2+ breast cancer. These significant findings suggest that targeting GLUT2 activity with phloretin in combination with anti-HER2 antibody drugs may help mitigate the development of drug-resistant breast cancer， offering valuable insights for enhancing tumor treatment strategies and contributing to developing more effective therapies.

Trastuzumab resistance is one of the causes of poor prognosis in patients with HER2-positive (HER2+) breast cancer (BC). The truncated isoform of dopamine- and cAMP-regulated phosphoprotein (t-DARPP) has been reported to be involved in Trastuzumab therapy resistance and promoting tumor progression. To evaluate the t-DARPP expression in BC， paired tumors and surrounding normal tissues were analyzed by real-time PCR and confirmed higher DARPP-32 family mRNA expression in HER2+ BC tumor tissues. We established two patient-derived xenografts (PDX) mice models to test the efficacy of Trastuzumab， named model 1 (non-responder) and model 2 (responder). t-DARPP and p95-HER2 protein-protein interactions were detected in PDX tumor tissue from non-responders using F?rster resonance energy transfer assays. Instead， there is no response from the responder. Furthermore， mechanistic studies using transwell and western blot assays demonstrated that t-DARPP could upregulate epithelial-mesenchymal transition signaling proteins， enhance p95-HER2 expression and promote cell migration. We found that Quercetin effectively reduced t-DARPP expression in HER2+ BC cells. In t-DARPP ShRNA-suppressed cells， Quercetin synergistically enhanced Trastuzumab-induced apoptotic cell death and G2/M phase arrest. In conclusion， the combination of Quercetin and Trastuzumab treatment by targeting t-DARPP in HER2+ BC patients has the potential as a biomarker for mitigating drug resistance.

A treatment recommendation for trichoepithelioma: Thoughts on selection of treatment timing and method

In this report, we present the case of a 55-year-old woman with a 30-year history of multiple familial trichoepithelioma(MFT), with facial surface tumors that progressively worsened, causing ventilation disorders. In order to improve the patient's appearance and functional impairment, we performed a two-step surgical excision and skin grafting procedure post diagnosis through genetic testing. The 18-month follow-up confirmed that the effect was satisfactory without obvious recurrence. Based on this outcome, we have developed a diagnosis and treatment process for patients with MFT, aiming to provide a reference for standardized treatment of this rare disease.

Random vibration analysis of FGM plates subjected to moving load using a refined stochastic finite element method

The article introduces a finite element procedure using the bilinear quadrilateral element or four-node rectangular element(namely Q4 element) based on a refined first-order shear deformation theory(rFSDT) and Monte Carlo simulation(MCS), so-called refined stochastic finite element method to investigate the random vibration of functionally graded material(FGM) plates subjected to the moving *** advantage of the proposed method is to use r-FSDT to improve the accuracy of classical FSDT, satisfy the stress-free condition at the plate boundaries, and combine with MCS to analyze the vibration of the FGM plate when the parameter inputs are random quantities following a normal distribution. The obtained results show that the distribution characteristics of the vibration response of the FGM plate depend on the standard deviation of the input parameters and the velocity of the moving ***, the numerical results in this study are expected to contribute to improving the understanding of FGM plates subjected to moving loads with uncertain input parameters.

Cooperation among enterprises can bring overall and individual performance improvement,and a smooth coordination method is ***,due to the lack of customized coordination methods,cooperation in the downstream oil supply chain cannot be carried out *** paper intends to propose a multi-party coordination method to promote cooperation between oil shippers and pipeline operator by optimizing oil transportation,oil substitution and pipeline pricing *** integrated game-theoretic modeling and analysis approach is developed to characterize the operation behaviors of all stakeholders in the downstream oil supply *** proposed mixed integer nonlinear programming model constrains supply and demand capacity,transportation routes,oil substitution rules and pipeline freight *** transformation and price discretization are introduced for model linear *** experiments are carried out in the oil distribution system in South *** results show that compared to the business-as-usual scheme,the new scheme saves transportation cost by 3.48%,increases pipeline turnover by 5.7%,and reduces energy consumption and emissions by 7.66%and 6.77%.It is proved that the proposed method improves the revenue of the whole system,achieves fair revenue distribution,and also improves the energy and environmental benefits of the oil supply chain.

With the development of satellite miniaturization and remote sensing,the establishment of microsatellite constellations is an inevitable *** to their limited size,weight,and power,spaceborne storage systems with excellent scalability,performance,and reliability are still one of the technical bottlenecks of remote sensing *** on the commercial off-the-shelf field-programmable gate array and memory devices,a spaceborne advanced storage system(SASS)is proposed in this *** work provides a dynamic programming,queue scheduling multiple-input multiple-output cache technique and a high-speed,high-reliability NAND flash controller for multiple microsatellite payload *** results show that SASS has outstanding scalability with a maximum write rate of 2429 Mb/s and preserves at least 78.53%of the performance when a single NAND flash *** scheduling technique effectively shortens the data scheduling time,and the data remapping method of the NAND flash controller can reduce the retention error by at least 50.73%and the program disturbance error by at least 37.80%.
