Berberine induces cell cycle arrest and apoptosis in human gastric carcinoma SNU-5 cell line

AIM： To investigate the relationship between the inhibited growth （cytotoxic activity） of berberine and apoptotic pathway with its molecular mechanism of action. METHODS： The in vitro cytotoxic techniques were complemented by cell cycle analysis and determination of sub-G1 for apoptosis in human gastric carcinoma SNU-5 cells. Percentage of viable cells, cell cycle, and sub431 group （apoptosis） were examined and determined by the flow cytometric methods. The associated proteins for cell cycle arrest and apoptosis were examined by Western blotting. RESULTS： For SNU-5 cell line, the IC （50） was found to be 48 μmol/L of berberine. In SNU-5 cells treated with 25-200 μmol/L berberine, G2/M cell cycle arrest was observed which was associated with a marked increment of the expression of p53, Wee1 and CDk1 proteins and decreased cyclin B. A concentration-dependent decrease of cells in G0/G1 phase and an increase in G2/M phase were detected. In addition, apoptosis detected as sub-Go cell population in cell cycle measurement was proved in 25-200 μmol/L berberine-treated cells by monitoring the apoptotic pathway. Apoptosis was identified by sub-Go cell population, and upregulation of Bax, downregulation of Bcl-2, release of Ca^2＋, decreased the mitochondrial membrane potential and then led to the release of mitochondrial cytochrome C into the cytoplasm and caused the activation of caspase-3, and finally led to the occurrence of apoptosis. CONCLUSION： Berberine induces p53 expression and leads to the decrease of the mitochondrial membrane potential, Cytochrome C release and activation of caspase-3 for the induction of apoptosis.

Vegetable/fruit, smoking, glutathione S-transferase polymorphisms and risk for colorectal cancer in Taiwan

AIM: To investigate the colorectal cancer risk associated with polymorphic GSTM1, GSTT1 and GSTP1 and the effect of diet and ***: With consents, genotypes of the genes were determined using PCR methods for 727 cases and 736sex and age-matched healthy controls recruited at a medical center in the Northern Taiwan. Nurses who were blind to the study hypothesis conducted interviews with study participants for the information of socio-demographic variables, diet and ***: There was no significant association between GSTM1 genotypes and the disease. Men, not women, with GSTT1 null genotype were at significant risk of colorectal cancer, but limited to rectal tumor, and in men aged 60 years and less. The corresponding association with the GSTP1 with G allele compared to GSTP1 A/A genotype was at borderline significance. Compared to men with GSTT1 present and GSTP1 A/A combined, men with both GSTT1 null and GSTP1 with G allele genotypes were at significant risk (odds ratio (OR) = 1.91, 95% confidence interval (CI) = 1.21-3.02), also limited to the rectal tumor and younger men. The beneficial effects of vegetable/fruit intake on colorectal cancer were much higher for men with GSTT1 present (OR = 0.32, 95%CI = 0.20-0.50) or GSTP1 A/A genotypes (OR = 0.40, 95%CI = 0.25-0.64).These effects remained significant for women. But, the greatest protective effect from vegetable/fruit intake for women was observed in those with GSTT1 null or GSTP1 with G allele genotypes. In addition, non-smoking men benefitted significantly from combined effect of higher vegetable/fruit intake and GSTT1 present or GSTP1 A/A genotypes with OR = 0.17 and 0.21 ***: This study suggests that the GSTT1 gene can modulate the colorectal cancer risk and vegetable/fruit-related colorectal cancer risk, particularly in men of no smoking history.

Decomposition of perfluorooctanoic acid by microwaveactivated persulfate： Effects of temperature, pH, and chloride ions

Microwave-hydrothermal treatment of persis- tent and bioaccumulative perfluorooctanoic acid （PFOA） in water with persulfate （S2O2-） has been found effective. However, applications of this process to effectively remediate PFOA pollution require a better understanding on free-radical scavenging reactions that also take place. The objectives of this study were to investigate the effects ofpH （pH= 2.5, 6.6, 8.8, and 10.5）, chloride concentra- tions （0.01-0.15 mol.L-1）, and temperature （60℃, 90℃, and 130℃） on persulfate oxidation of PFOA under microwave irradiation. Maximum PFOA degradation occurred at pH 2.5, while little or no degradation at pH 10.5. Lowering system pH resulted in an increase in PFOA degradation rate. Both high pH and chloride concentrations would result in more scavenging of sulfate free radicals and slow down PFOA degradation. When chloride concentrations were less than 0.04 mol.L^-1 at 90℃ and 0.06mol.L^-1 at 60℃, presence of chloride ions had insignificant impacts on PFOA degradation. However, beyond these concentration levels, PFOA degradation rates reduced significantly with an increase in chloride concentrations, especially under the higher temperature.

Enterprises have always sacrificed the benefits of the human and environmental causes in order to pursue the maximization of companies’ *** more awareness of the concept of environmental protection and Corporate Social Responsibility（CSR）,enterprises have to commence to take the potential duty for the human and the environment beyond the going-concern assumption and the achievement of profits maximization for ***,with reference to vigorous environmental issues（unstable variation of climate,huge fluctuation of the Earth’s crust,abnormal raises of the sea and so on）,the traditional accounting principles and assumptions do conform to the contemporarily social ***,based on the full disclosure principle,enterprises are further supposed to disclose the full financial cost of complying with environment regulations（regarding the dystrophication resulted from operation,amended honorable of contamination,environmental protection policies and so on） which are going to lead to enterprises are still able to achieve "economy-efficiency" for the environments under the go-concern ***,the innovative accounting theory（the Green Accounting Theory） is created to establish new accounting principles and regulations for confronting these *** paper utilizes three essential relations in consolidation that are evaluated by nineteen assessable sub-criteria of four evaluated criteria through the comparison of Multiple Criteria Decision Making（MCDM）-Analytical Network Process（ANP）.Specifically,the specific feature of the three-approach models is to calculate the priority vector weights of each assessable characteristic,criteria and sub-criteria by pairwise comparative ***,in the content,the analytical hierarchical relations are definitely expressed in four levels among each criterion which enable enterprises to choose the potential role of green accounting in CSR in a thriving hypercompetitive commerce environment.

Eradication of Helicobacter pylori significantly reduced gastric damage in nonsteroidal anti-inflammatory drug-treated Mongolian gerbils

AIM: To examine the effect of eradication of Hellcobacter pylori prior to usage of NSAIDs, by investigating gastric inflammatory activity, myeloperoxidase (MPO) activity, prostaglandin (PG) E_2 synthesis in H Pylori-infected, and H pylori-eradicated gerbils followed by administration of indomethacin and rofecoxib. METHODS: Six-week-old male gerbils were orally inoculated with H pylori. Seven weeks later, anti-H pylori triple therapy and vehicle were given to gerbils respectively and followed by oral indomethacin (2 mg/kg·d) or rofecoxib (10 mg/kg·d) for 2 wk. We examined the area of lesions, gastric inflammatory activity, PGE_2 synthesis and MPO activity in the stomach. RESULTS: In indomethacin and rofecoxib-treated gerbils, the following results were obtained in H pylori-infected group vs H pylori-eradicated group respectively: hyperplasia area of the stomach (mm^2): 82.4±9.2 vs 13.9±3.5 (P＜0.05), 30.5±5.1 vs 1.3±0.6 (P＜0.05); erosion and ulcer area (mm^2): 14.4±4.9 vs 0.86±0.5 (P＜0.05), 1.3±0.6 vs0.4±0.3 (P＜0.05); score of gastritis: 7.0±0.0 vs3.6±0.5 (P＜0.05), 7.0±0.0 vs 2.7±0.5 (P＜0.05); MPO activity (μmol H_2O_2/min/g tissue): 104.7±9.2 vs9.0±2.3(P＜0.05), 133.5±15.0 vs2.9±0.7 (P＜0.05); PGE_2 synthesis (pg/mg wet weight/min): 299.2±81.5 vs102.8±26.2 (P＜0.05), 321.4±30.3 vs 11.9±4.8(P＜0.05). CONCLUSION: Eradication of H pylori reduced gastric damage of NSAID-treated Mongolian gerbils. Rofecoxib caused less severe gastric damage than indomethacin in H pylori-eradicated gerbils.

Cytosine methylation is a covale nt modificatio n of DNA that regulates important processes in eukaryotic genomes,including gene transcription,transposon silencing,and genomic imprinting(Law and Jacobsen,2010).DNA methylation patterns are faithfully duplicated upon cell division to ensure genome integrity and to maintain lineage-specific cell ***,DNA methylation also needs to be dynamically reprogramed or reconfigured during development to allow establishment of new cellular identity and transcriptional state,which plays a prominent role in animal development and reproduction,and is increasingly being appreciated for reproductive success in flowering plants(Walker et al.,2018;Ono and Kinoshita,2021).
