Objective: To identify the core targets of Rheum palmatum L. and Salvia miltiorrhiza Bge.,(Dahuang-Danshen, DH-DS) and the mechanism underlying its therapeutic efficacy in acute pancreatitis(AP)using a network pharmacology approach and validate the findings in animal experiments. Methods: Network pharmacology analysis was used to elucidate the mechanisms underlying the therapeutic effects of DH-DS in AP. The reliability of the results was verified by molecular docking simulation and molecular dynamics ***, the results of network pharmacology enrichment analysis were verified by immunohistochemistry,Western blot analysis and real-time quantitative PCR, respectively. Results: Sixty-seven common targets of DH-DS in AP were identified and mitogen-activated protein kinase 3(MAPK3), Janus kinase 2(JAK2), signal transducer and activator of transcription 3(STAT3), protein c-Fos(FOS) were identified as core targets in the protein interaction(PPI) network analysis. Gene ontology analysis showed that cellular response to organic substance was the main functions of DH-DS in AP, and Kyoto Encyclopedia of Genes and Genomes analysis showed that the main pathway included Th17 cell differentiation. Molecular docking simulation confirmed that DH-DS binds with strong affinity to MAPK3, STAT3 and FOS. Molecular dynamics simulation revealed that FOS-isotanshinone Ⅱ and STAT3-dan-shexinkum d had good binding capacity. Animal experiments indicated that compared with the AP model group, DH-DS treatment effectively alleviated AP by inhibiting the expression of interleukin-1β, interleukin-6 and tumor necrosis factor-α, and blocking the activation of Th17 cell differentiation(P<0.01). Conclusion: DH-DS could inhibit the expression of inflammatory factors and protect pancreatic tissues,which would be functioned by regulating Th17 cell differentiation-related m RNA and protein expressions.

Dietary fishmeal replacement by Clostridium autoethanogenum protein meal influences the nutritional and sensory quality of turbot(Scophthalmus maximus)via the TOR/AAR/AMPK pathways

Clostridium autoethanogenum protein(CAP)is a promising protein source for aquaculture;however,how CAP influences fish quality is worth extensive *** randomly allocated 630 turbot with initial body weights of about 180 g into 6 groups,with fishmeal-based control diet or diet with CAP replacing 15%(CAP15),30%(CAP30),45%(CAP45),60%(CAP60),or 75%(CAP75)of fishmeal *** a 70-d feeding trial,the fillet yield(P=0.015)and content of protein(P=0.017),collagen(P 0.05).By contrast,the muscle hardness increased linearly with increasing CAP(P=0.004),accompanied by linear reduction of muscle fiber area(P=0.003)and expression of myogenesis-related genes,including cathepsin D(ctsdPlinearly;however,phosphorylation of AMP-activated protein kinase(AMPK,Pling pathway was inhibited,and the amino acid response(AAR)and AMPK pathways were ***,expression of genes related to protein degradation,including myogenic factor 5(myf 5,Plinearly with increasing *** conclusion,CAP could be used to replace up to 15%of fishmeal without negatively impacting turbot ***,higher levels of CAP decreased fillet yield,muscle protein content,and muscle fiber diameter while increasing muscle hardness,which could be attributed to the inhibition of the TOR pathway and activation of the AAR and AMPK pathways.

The disparity in the transfer of carriers(electrons/mass)during the reaction in zinc-air batteries(ZABs)results in sluggish kinetics of the oxygen reduction reaction(ORR)and oxygen evolution reaction(OER),along with elevated overpotentials,thereby imposing additional constraints on its ***,the pre-design and target-development of inexpensive,high-performance,and long-term stable bifunctional catalysts are urgently *** this work,an apically guiding dual-functional electrocatalyst(Ag-FeN_(x)-N-C)was prepared,in which a hierarchical porous nitrogen-doped carbon with three-dimensional(3D)hollow star-shaped structure is used as a substrate and high-conductivity Ag nanoparticles are coupled with iron nitride(FeN_(x))*** calculations indicate that the Mott-Schottky heterojunction as an inherent electric field comes from the two-phase bound of Ag and FeN_(x),of which electron accumulation in the FeN_(x)phase region and electron depletion in the Ag phase region promote orientated-guiding charge *** effective modulation of local electronic structures felicitously reforms the d-band electron-group distribution,and intellectually tunes the masstransfer reaction energy barriers for both ORR/***,the hollow star-s haped hierarchical porous structure provides an apical region for fast mass *** results show that the halfwave potential for ORR is 0.914 V,and the overpotential for OER is only 327 mV at 10 mA cm^(-2).A rechargeable ZAB with Ag-FeN_(x)-N-C as the air cathode demonstrates long-term cycling performance exceeding 1500 cycles(500 h),with a power density of 180 mW cm^(-2).Moreover,when employing AgFeN_(x)-N-C as the air cathode,flexible ZABs demonstrate a notable open-circuit voltage of 1.42 V and achieve a maximum power density of 65.6 mW cm^(-2).Ag-FeN_(x)-N-C shows guiding electron/mass transfer route and apical reaction microenvironment for the electrocatalyst architecture in the explo

Predictability of the 7·20 extreme rainstorm in Zhengzhou in stochastic kinetic-energy backscatter ensembles

The scale-dependent predictability of the devastating 7·20 extreme rainstorm in Zhengzhou,China in 2021 was investigated via ensemble experiments,which were perturbed on different scales using the stochastic kinetic-energy backscatter(SKEB)scheme in the WRF model,with the innermost domain having a 3-km grid *** daily rainfall(RAIN24h)and the cloudburst during 1600-1700 LST(RAIN1h)were *** demonstrated that with larger perturbation scales,the ensemble spread for the rainfall maximum widens and rainfall forecasts become closer to the *** ensembles with mesoscale or convective-scale perturbations,RAIN1h loses predictability at scales smaller than 20 km and RAIN24h is predictable for all *** in ensembles with synoptic-scale perturbations,the largest scale of predictability loss extends to 60 km for both RAIN1h and ***,the average positional error in forecasting the heaviest rainfall for RAIN24h(RAIN1h)was 400 km(50-60)*** southerly low-level jet near Zhengzhou was assumed to be directly responsible for the forecast uncertainty of *** rapid intensification in low-level cyclonic vorticity,mid-level divergence,and upward motion concomitant with the jet dynamically facilitated the *** analysis of the divergent,rotational and vertical kinetic spectra and the corresponding error spectra showed that the error kinetic energy at smaller scales grows faster than that at larger scales and saturates more quickly in all ***-scale perturbations not only boost larger-scale error growth but are also conducive to error growth at all scales through a downscale cascade,which indicates that improving the accuracy of larger-scale flow forecast may discernibly contributes to the forecast of cloudburst intensity and position.

TULAM:trajectory-user linking via attention mechanism

Recently,the application of location-based services(LBS) has become a prevalent means to provide convenience in customers’ everyday ***,because massive volumes of location information are collected by LBS applications,users may suffer from serious privacy *** studies have shown that the identities of users can be inferred from historical anonymous trajectories,which is formulated as the trajectory-user linking(TUL) *** some recurrent neural network(RNN)-based models have been proposed to capture implicit movement patterns among trajectories to improve TUL performance,they cannot learn the sequential and contextual semantics within any individual trajectory completely,leaving the advantages of RNNs *** therefore propose an RNN model with an attention mechanism called TULAM to improve the accuracy of the TUL *** learns sequential relationships within individual trajectories via RNN and captures contextual semantics from trajectories via a multi-head attention ***,we propose a novel location encoding method called approximate one-hot to solve the corpus shortage problem of trajectory *** were conducted on real datasets from the Gowalla and Foursquare LBS *** experimental results indicate that TULAM is a practical solution that achieves significant improvements over existing methods with satisfactory model complexity and convergence.

Data-driven surrogate model for aerodynamic design using separable shape tensor method

In the context of increasing dimensionality of design variables and the complexity of con-straints,the effcacy of Surrogate-Based Optimization(SBO)is *** traditional linear and nonlinear dimensionality reduction algorithms are mainly to decompose the mathematical matrix composed of design variables or objective functions in various forms,the smoothness of the design space cannot be guaranteed in the process,and additional constraint functions need to be added in the optimization,which increases the calculation *** study presents a new parameterization method to improve both problems of *** new parameterization is addressed by decoupling affine transformations(dilation,rotation,shearing,and translation)within the Grassmannian sub-manifold,which enables a separate representation of the physical information of the airfoil in a high-dimensional *** upon this,Principal Geodesic Analysis(PGA)is employed to achieve geometric control,compress the design space,reduce the number of design variables,reduce the dimensions of design variables and enhance predictive performance during the surrogate optimiza-tion *** comparison,a dimensionality reduction space is defined using 95%of the energy,and RAE 2822 for transonic conditions are used as *** method significantly enhances the optimization efficiency of the surrogate model while effectively enabling geometric *** three-dimensional problems,it enables simultaneous design of planar shapes for various components of the aircraft and high-order perturbation *** was applied to the ONERA M6 wing,achieving a lift-drag ratio of 18.09,representing a 27.25%improvement com-pared to the baseline *** comparison to conventional surrogate model optimization methods,which only achieved a 17.97%improvement,this approach demonstrates its superiority.

Polyethylene interfacial dielectric layer for organic semiconductor single crystal based field-effect transistors

Organic semiconductor single crystals(OSSCs) have shown their promising potential in high-performance organic field-effect transistors(OFETs). The interfacial dielectric layers are critical in these OFETs as they not only govern the key semiconductor/dielectric interface quality but also determine the growth of OSSCs by their wetting properties. However, reported interfacial dielectric layers either need rigorous preparation processes, rely on certain surface chemistry reactions, or exhibit poor solvent resistance, which limits their applications in low-cost, large-area, monolithic fabrication of OSSC-based OFETs. In this work, polyethylene(PE) thin films and lamellar single crystals are utilized as the interfacial dielectric layers, providing solvent resistive but wettable surfaces that facilitate the crystallization of 6,13-bis(tri-isopropylsilylethynyl)pentacene(TIPS-PEN) and 6,13-bis(triisopropylsilylethynyl)-5,7,12,14-tetraazapentacene(TIPS-TAP). As evidenced by the presence of ambipolar behavior in TIPS-PEN single crystals and the high electron mobility(2.3 ± 0.34 cm^(2)V^(-1)s^(-1)) in TIPS-TAP single crystals, a general improvement on electron transport with PE interfacial dielectric layers is revealed, which likely associates with the chemically inertness of the saturated C-H bonds. With the advantages in both processing and device operation, the PE interfacial dielectric layer potentially offers a monolithic way for the enhancement of electron transport in solution-processed OSSC-based OFETs.

Highly elastic and degradable vitrimeric elastomers using polycondensation

Introducing covalently crosslinked network to polymer matrix can merge the advantages in reprocessing and durability of *** this contribution,a series of high-performance vitrimeric elastomers were achieved via *** topological structures of polymers were tuned by varying the feeding ratios of bisacetoacetate,hex–substituted bisacetoacetate,bisamine and tris(2-aminoethyl)*** these structural manipulations,the vitrimeric elastomers presented great elastic recovery properties(strain recovery value up to 80%)benefiting from the introduction of long chain ***,the elastomers exhibited excellent reprocessing property,water vapor/oxygen barrier and adhesive ***,the elastomers could be degraded into monomer under acid conditions which enabled the elastomer synthesis again in closed loop recycling *** ease of the polycondensation in this work to prepare highly elastic and recyclable vitrimeric elastomers demonstrated exciting opportunities for the synthesis of sustainable polymers.
