Effect of serum concentration on adhesion of monocytic THP-1 cells onto cultured EC monolayer and EC-SMC co-culture

Background:The adhesion of monocytes to the endothelium following accumulation of low-density lipoprotein (LDL) in subendothelial spaces is an important step in the development of intimal hyperplasia in arterially implanted vein grafts and atherosclerosis in both animals and humans. However, it is not well known how serum factors affect the adhesion of monocytes. Methods: We have studied the effect of fetal calf serum (FCS), which we considered a source of LDL, on the adhesion of monocytes to endothelial cells (ECs) by using human monocytic THP-1 cells and both a monolayer of cultured bovine aortic endothelial cells (EC monoculture) and a co-culture with bovine aortic smooth muscle cells (EC-SMC co-culture). Results: It was found that the addition of FCS to the medium greatly affected the adhesion of THP-1 cells, and the higher the concentration of FCS in the medium, the greater the adhesion of THP-1 cells to endothelial cells. Adhesion of THP-1 cells to an EC-SMC co-culture was approximately twofold greater than that to an EC monoculture, and after adhering to endothelial cells, many THP-1 cells trans-migrated into the layer of smooth muscle cells. Conclusion: The results suggest that the elevation of the LDL (cholesterol) level in blood provides a favorable condition for the development of intimal hyperplasia and atherosclerosis by promoting the adhesion of monocytes to the endothelium and their subsequent migration into subendothelial spaces.

The paper presents a review of dynamic stabilization mechanisms for plasma instabilities. One of the dynamic stabilization mechanisms for plasma instability was proposed in the paper [Kawata, Phys. Plasmas 19, 024503(2012)],based on a perturbation phase control. In general, instabilities emerge from the perturbations. Normally the perturbation phase is unknown, and so the instability growth rate is discussed. However, if the perturbation phase is known, the instability growth can be controlled by a superimposition of perturbations imposed actively. Based on this mechanism we present the application results of the dynamic stabilization mechanism to the Rayleigh–Taylor instability(RTI) and to the filamentation instability as typical examples in this paper. On the other hand, in the paper [Boris, Comments Plasma Phys. Control. Fusion 3, 1(1977)] another mechanism was proposed to stabilize RTI, and was realized by the pulse train or the laser intensity modulation in laser inertial fusion [Betti et al., Phys. Rev. Lett. 71, 3131(1993)]. In this latter mechanism, an oscillating strong force is applied to modify the basic equation, and consequently the new stabilization window is created. Originally the latter was proposed by Kapitza. We review the two stabilization mechanisms, and present the application results of the former dynamic stabilization mechanism.

High blood pressure (hypertension) is implicated in the development of atherosclerosis. Blood vessels are constantly subjected to stretch due to blood pressure and changes in stretch usually instigate adaptive vascular remodeling, including abnormal growth and proliferation of vascular smooth muscle cells (VSMCs) as well as extracellular matrix (ECM). In this experiment, we used bovine aortic endothelial cells and smooth muscle cells (EC-SMC) co-cultured ePTFE vascular grafts subjected to normal atmospheric pressure (as a control), and 100 mmHg hydrostatic pressure for 7 d. The increase of cell layer thickness was observed. When measured, the cell layer thickness increased by 116.2%. The increase of collagen (Type Ⅳ)synthesis was also observed in the immunohistochemistry assay. When stained with toluidine blue, the cells showed metachromatic phenomenon.

In this review paper on heavy ion inertial fusion(HIF),the state-of-the-art scientific results are presented and discussed on the HIF physics,including physics of the heavy ion beam(HIB)transport in a fusion reactor,the HIBs-ion illumination on a direct-drive fuel target,the fuel target physics,the uniformity of the HIF target implosion,the smoothing mechanisms of the target implosion non-uniformity and the robust target *** HIB has remarkable preferable features to release the fusion energy in inertial fusion:in particle accelerators HIBs are generated with a high driver efficiency of~30%-40%,and the HIB ions deposit their energy inside of ***,a requirement for the fusion target energy gain is relatively low,that would be~50-70 to operate a HIF fusion reactor with the standard energy output of 1 GWof *** HIF reactor operation frequency would be~10-15 Hz or ***-MJ HIBs illuminate a fusion fuel target,and the fuel target is imploded to about a thousand times of the solid *** the DT fuel is ignited and *** HIB ion deposition range is defined by the HIB ions stopping length,which would be~1 mm or so depending on the ***,a relatively large density-scale length appears in the fuel target *** of the critical issues in inertial fusion would be a spherically uniform target compression,which would be degraded by a non-uniform *** implosion non-uniformity would be introduced by the Rayleigh-Taylor(R-T)instability,and the large densitygradient-scale length helps to reduce the R-T growth *** the other hand,the large scale length of the HIB ions stopping range suggests that the temperature at the energy deposition layer in a HIF target does not reach a very-high temperature:normally about 300 eV or so is realized in the energy absorption region,and that a direct-drive target would be appropriate in *** addition,the HIB accelerators are operated repetitively and *** precis

Cardiopulmonary resuscitation,commonly known as CPR,is an emergency procedure that normally combines chest compression with artificial ventilation in an effort to preserve intact brain function manually until further measures can be taken to restore spontaneous blood circulation and breathing in a person who is in cardiac *** this study,we evaluated the skills of CPR practitioners on the basis of kinematic data obtained from their body movements while performing *** particular,we used a Microsoft Kinect sensor to evaluate CPR performance by new and more experienced practitioners and to analyze CPR skill-building *** measurement using the Kinect sensor enabled detailed information about motion at body joints to be displayed quickly and objectively,thus facilitating identification of any ***,we could not confirm gesture recognition and detailed motion analysis based on using a high-speed camera to capture three-dimensional(3-D)motion of the entire body,we determined the Kinect sensor to be an easily applied evaluation tool that can provide body-motion information quickly and thus serve as an objective index for evaluating CPR performance.

我们研究了在2.7Ni-0.45Cr-0.5Mo-0.1V钢中,降低硅含量对机械性能的影响,以便为了解决钢锭偏析问题,采用低硅钢来制造高压-低压整体转子轴。降低硅合量时,可以改善韧性和回火脆化,但在500℃以下的温度,抗张强度也会降低。这一点可以用增加少量的铬来得到充分补偿,而不致损害其他性能。不管降低硅或是增加铬,蠕变断裂强度都不会发生变化。川崎钢铁公司曾经发展一种特殊的炼钢方法,可以制出磷硫和其他杂质都很低的高级钢。用此法实地制成了一根整体的转子轴,试验证明它具有令人满意的机械性能,正象我们进行初步的研究后所预期的那样。
